| 1 | Photoexcited phytochrome B interacts with brassinazole resistant 1 to repress brassinosteroid signaling in Arabidopsis显示文摘Photoreceptor phytochrome B(phy B)mediates a variety of light responses in plants.To further elucidate the molecular mechanisms of phy B-regulated hypocotyl elongation,we performed firefly luciferase complementation imaging(LCI)screening for phy B-interacting transcription factors(TFs).LCI assays showed that phy B possibly interacts with brassinazoleresistant 1(BZR1),BZR2,AUXIN RESPONSE FACTOR 6(ARF6),and several WRKY DNA-binding TFs in a red light-dependent manner.Furthermore,biochemical assays demonstrated that photoexcited phy B specifically interacts with nonphosphorylated BZR1,the physiologically active form of a master TF in brassinosteroid(BR)signaling,and this interaction can be competitively interfered by phytochrome-interacting factor 4.Furthermore,we showed that phy B can directly interact with the DNA-binding domain of BZR1 and affect the enrichment of BZR1 on the chromatin of target genes.Moreover,our genetic evidence and RNA-seq analysis demonstrated that phy B negatively regulates BR signaling.Together,we revealed that photoexcited phy B directly interacts with the TF BZR1 to repress BR signaling in Arabidopsis. | Huixue Dong Jie Liu Guanhua He Pan Liu Jiaqiang Sun | 2020 | Journal of Integrative Plant Biology2020,62,5: | 5 |
| 2 | An enhanced method for nucleic acid detection with CRISPR-Cas12a using phosphorothioate modified primers and optimized gold-nanopaticle strip显示文摘CRISPR-Cas12a system has been shown promising for nucleic acid diagnostics due to its rapid,portable and accurate features.However,cleavage of the amplicons and primers by the cis-and trans-activity of Cas12a hinders the attempts to integrate the amplification and detection into a single reaction.Through phosphorothioate modification of primers,we realized onepot detection with high sensitivity using plasmids of SARS-CoV-2,HPV16 and HPV18.We also identified the activated Cas12a has a much higher affinity to C nucleotide-rich reporter than others.By applying such reporters,the reaction time required for a lateral-flow readout was significantly reduced.Furthermore,to improve the specificity of the strip-based assay,we created a novel reporter and,when combined with a customized gold-nanopaticle strip,the readout was greatly enhanced owing to the elimination of the nonspecific signal.This established system,termed Targeting DNA by Cas12a-based Eye Sight Testing in an Onepot Reaction(TESTOR),was validated using clinical cervical scrape samples for human papillomaviruses(HPVs)detection.Our system represents a general approach to integrating the nucleic acid amplification and detection into a single reaction in CRISPR-Cas systems,highlighting its potential as a rapid,portable and accurate detection platform of nucleic acids. | Jiaojiao Gong Lijuan Kan Xiuming Zhang Ying He Jiaqiang Pan Liping Zhao Qianyun Li Menghao Liu Jie Tian Sili Lin Zhouyu Lu Liang Xue Chaojun Wang Guanghui Tang | 2021 | Bioactive Materials2021,6,12: | 1 |

| 9 | Wheat male-sterile 2 reduces ROS levels to inhibit anther development by deactivating ROS modulator 1显示文摘Ms2 is an important dominant male-sterile gene in wheat,but the biochemical function of Ms2 and the mechanism by which it causes male sterility remain elusive.Here,we report the molecular basis underlying Ms2-induced male sterility in wheat.We found that activated Ms2 specifically reduces the reactive oxygen species(ROS)signals in anthers and thereby induces termination of wheat anther development at an early stage.Furthermore,our results indicate that Ms2 is localized in mitochondria,where it physically interacts with a wheat homolog of ROS modulator 1(TaRomo1).Romo1 positively regulates the ROS levels in humans but has never been studied in plants.We found that single amino acid substitutions in the Ms2 protein that rescue the ms2 male-sterile phenotype abolish the interaction between Ms2 and TaRomo1.Significantly,Ms2 promotes the transition of TaRomo1 proteins from active monomers to inactive oligomers.Taken together,our findings unravel the molecular basis of Ms2-induced male sterility and reveal a regulatory mechanism in which ROS act as essential signals guiding the anther development program in wheat. | Jie Liu Chuan Xia Huixue Dong Pan Liu Ruizhen Yang Lichao Zhang Xu Liu Jizeng Jia Xiuying Kong Jiaqiang Sun | 2022 | Molecular Plant2022,15,9: | 1 |

| 14 | Surface superconductivity in the type Ⅱ Weyl semimetal TaIrTe4显示文摘The search for unconventional superconductivity in Weyl semimetal materials is currently an exciting pursuit,since such superconducting phases could potentially be topologically non-trivial and host exotic Majorana modes.The layered material TaIrTe4 is a newly predicted time-reversal invariant type II Weyl semimetal with the minimum number of Weyl points.Here,we report the discovery of surface superconductivity in Weyl semimetal TaIrTe4.Our scanning tunneling microscopy/spectroscopy(STM/STS)visualizes Fermi arc surface states of TaIrTe4 that are consistent with the previous angle-resolved photoemission spectroscopy results.By a systematic study based on STS at ultralow temperature,we observe uniform superconducting gaps on the sample surface.The superconductivity is further confirmed by electrical transport measurements at ultralow temperature,with an onset transition temperature(Tc)up to 1.54 K being observed.The normalized upper critical field h?(T/Tc)behavior and the stability of the superconductivity against the ferromagnet indicate that the discovered superconductivity is unconventional with the p-wave pairing.The systematic STS,and thickness-and angular-dependent transport measurements reveal that the detected superconductivity is quasi-1D and occurs in the surface states.The discovery of the surface superconductivity in TaIrTe4 provides a new novel platform to explore topological superconductivity and Majorana modes. | Ying Xing Zhibin Shao Jun Ge Jiawei Luo Jinhua Wang Zengwei Zhu Jun Liu Yong Wang Zhiying Zhao Jiaqiang Yan David Mandrus Binghai Yan Xiong-Jun Liu Minghu Pan Jian Wang | 2020 | National Science Review2020,7,3: | 0 |
