1Development of synthetic biology tools to engineer Pichia pastoris as a chassis for the production of natural products显示文摘The methylotrophic yeast Pichia pastoris(a.k.a.Komagataella phaffii)is one of the most commonly used hosts for industrial production of recombinant proteins.As a non-conventional yeast,P.pastoris has unique biological characteristics and its expression system has been well developed.With the advances in synthetic biology,more efforts have been devoted to developing P.pastoris into a chassis for the production of various high-value compounds,such as natural products.This review begins with the introduction of synthetic biology tools for the engineering of P.pastoris,including vectors,promoters,and terminators for heterologous gene expression as well as Clustered Regularly Interspaced Short Palindromic Repeats/CRISPR-associated System(CRISPR/Cas)for genome editing.This review is then followed by examples of the production of value-added natural products in metabolically engineered P.pastoris strains.Finally,challenges and outlooks in developing P.pastoris as a synthetic biology chassis are prospected.Jucan Gao Lihong Jiang Jiazhang Lian 2021Synthetic and Systems Biotechnology2021,6,2:9

4Identification of Peptides Inhibiting Adhesion of Monocytes to the Injured Vascular Endothelial Cells through Phage-displaying Screening显示文摘Using oxidized low-density lipoprotein (LDL)-injured vascular endothelial cells (ECs) as target cells, peptides specifically binding to the injured ECs were screened from a phage-displaying peptide library by using the whole-cell screening technique after three cycles of the ''''''''''''''''''''''''''''''''''''''''''''''''''''''''''''''''adsorption-elution-amplification'''''''''''''''''''''''''''''''''''''''''''''''''''''''''''''''' procedure. Positive phage clones were identified by ELISA, and the inserted amino acid sequences in the displaying peptides were deduced from confirmation with DNA sequencing. The adhesion rate of ECs to monocytes was evaluated by cell counting. The activity of endothelial nitric oxide synthase (eNOS), and the expression levels of caveolin-1 and intercellular adhesion molecule-1 (ICAM-1) were determined by Western blotting. Six positive clones specifically binding to injured ECV304 endothelial cells were selected from fourteen clones. Interestingly, four phages had peptides with tandem leucine, and two of these even shared an identical sequence. Functional analysis demonstrated that the YCPRYVRRKLENELLVL peptide shared by two clones inhibited the expression of ICAM-1, increased nitric oxide concentration in the culture media, and upregulated the expression of caveolin- 1 and eNOS. As a result, the adhesion rate of monocytes to ECV304 cells was significantly reduced by 12.1%. 6Exo^+ proofreading polymerases mediate genetic analysis and its application in biomedical studies显示文摘Polymerases with a proofreading function in their internal 3' to 5' exonuclease possess high fidelity for DNA replication both in vivo and in vitro. The obstacle facing Exo^+ polymerases for single nucleotide polymorphism (SNP) detection could be bypassed by using primer-3'-termini modification. This hypothesis has been well tested using three types of modified allele specific primers with: 3' labeling, 3'to 5'exonuclease resistance, and 3'dehydroxylation. Accordingly, three new SNP assaying methods have been developed to carry out genome-wide genotyping, taking advantage of the enzymatic properties of Exo^+ polymerases. These new mutation detection assays are widely adaptable to a variety of platforms, including multi-well plate and microarray technologies. Application of Exo^+ polymerases to genetic analysis, including genotyping that is mostly relevant to pharmacogenetics, high-fidelity gene expression profiling, rare mutation detection and mutation load assay, will help to accelerate the pace of personalized medicine. 8Research Progress of Rice-Fish Integrated Farming显示文摘Rice-fish integrated farming( RFIF) can change the agricultural production pattern from mono-farming model to the co-farming model,and the stereoscopic and integrated farming system established by RFIF can change the flat production to the stereoscopic production. Therefore,RFIF is the important development direction for agriculture to change pattern and adjust structure. In order to further promote development of the research and application of RFIF,the general development situation of RFIF in China was reviewed; the effects of RFIF on increasing farmer' income and enhancing agricultural efficiency were clarified. The technologies matched with RFIF,such as rice paddy engineering technology,safe plant protection technology,security fertilization technology,fish disease prevention and control technology and water quality management technology,were summarized,and the supporting role of RFIF for agriculture was pointed out. 14Biomanufacturing ofγ-linolenic acid-enriched galactosyldiacylglycerols:Challenges in microalgae and potential in oleaginous yeasts显示文摘γ-Linolenic acid-enriched galactosyldiacylglycerols(GDGs-GLA),as the natural form ofγ-linolenic acid in microalgae,have a range of functional activities,including anti-inflammatory,antioxidant,and anti-allergic properties.The low abundance of microalgae and the structural stereoselectivity complexity impede microalgae extraction or chemical synthesis,resulting in a lack of supply of GDGs-GLA with a growing demand.At present,there is a growing interest in engineering oleaginous yeasts for mass production of GDGs-GLA based on their ability to utilize a variety of hydrophobic substrates and a high metabolic flux toward fatty acid and lipid(triacylglycerol,TAG)production.Here,we first introduce the GDGs-GLA biosynthetic pathway in microalgae and challenges in the engineering of the native host.Subsequently,we describe in detail the applications of oleaginous yeasts with Yarrowia lipolytica as the representative for GDGs-GLA biosynthesis,including the development of synthetic biology parts,gene editing tools,and metabolic engineering of lipid biosynthesis.Finally,we discuss the development trend of GDGs-GLA biosynthesis in Y.lipolytica.Xiaosong Gu Lei Huang Jiazhang Lian 2023Synthetic and Systems Biotechnology2023,8,3:0

19Testis of Male Tilapia Under Methomyl Stress: Transcriptome Changes and Signal Pathway Analysis显示文摘This study was conducted to find the differentially expressed genes and understand the transcriptome change characteristics in the testis of tilapia(Oreochromis niloticus) stressed by methomyl.The tilapia were exposed to 200 μg/L methomyl for 30 d,and then the testis samples were collected and used for the highthroughput sequencing.The results showed that 369 significant differentially expressed genes were obtained,including 160 significantly down-regulated genes,and209 significantly up-regulated genes.The enrichment of the differentially expressed genes in ECM-receptor interaction signaling pathway and MAPK signaling pathway was obvious.The adhesion of testis tissue cells was influenced by methomyl stress,which induced the expression of related gene in ECM-receptor interaction up-regulating,such as laminins,integrin α6 and integrin β1,in order to stabilize testis tissue structure.JNK,MAP3 K and MKK4 in the JNK pathway were significantly up-regulated,which enhanced transcription factor AP-1 to promote the expression of P53,Bax and other pro-apoptotic proteins.The expression of growth-related factors in the ERK pathway of MAPK signal pathway was influenced by methomyl stress.The apoptosis-related genes and pregnancy-related genes have significant differential expression under methomyl stress,and thereby,the proliferation of reproduction cells in the testis tissue may be affected.Shunlong MENG Tao LIU Chao SONG Cong ZHANG Jiazhang CHEN 2020Agricultural Biotechnology2020,9,2:0
