| 1 | Effects of lactic acid bacteria isolated from fermented mustard on lowering cholesterol显示文摘Objective:To evaluate the ability of lactic acid bacteria(LAB)strains isolated from fermented mustard to lower the cholesterol in vitro.Methods:The ability of 50 LAB strains isolated from fermented mustard on lowering cholesterol in vitro was determined by modified o-phtshalaldehyde method.The LAB isolates were analyzed for their resistance to acid and bile salt.Strains with lowering cholesterol activity,were determined adherence to Caco-2 cells.Results:Strain B0007,B0006 and B0022 assimilated more cholesterol than BCRC10474 and BCRC17010.The isolated strains showed tolerance to pH 3.0 for 3h despite variations in the degree of viability and bile-tolerant strains,with more than 10~s CFU/mL after incubation for 24 h at 1%oxigall in MRS.In addition,strain B0007 and B0022 identified as Lactobacillus plantarum with 16S rDNA sequences were able to adhere to the Caco-2 cell lines.Conclusions:These strains B0007 and B0022 may be potential functional sources for cholesterollowering activities as well as adhering to Caco-2 cell lines. | Shu Chen Wang Chen Kai Chang Shu Chang Chan Jiunn Shiuh Shieh Chih Kwang Chiu Pin-Der Duh | 2014 | Asian Pacific Journal of Tropical Biomedicine2014，4，7： | 10 |
| 2 | Comparison of peripapillary retinal nerve fiber layer thickness between myopia severity groups and controls显示文摘AIM: To compare the peripapillary retinal nerve fiber layer(RNFL) thickness measured via optical coherence tomography(OCT) between different groups of myopia severity and controls. METHODS: This was a prospective cross-sectional study. All subjects underwent a full ophthalmic examination, refraction, visual field analysis and A-scan biometry. Myopic patients were classified as low myopia(LM) [spherical equivalent(SE) from greater than-0.5 D, up to-3.0 D], moderate myopia(MM; SE greater than-3.0 D, up to-6.0 D) and high myopia(HM; SE greater than-6.0 D). The control group consisted of emmetropic(EM) patients(SE from +0.5 D to-0.5 D). A Zeiss Cirrus HD-OCT machine was used to measure the peripapillary RNFL thickness of both eyes of each subject. The mean peripapillary RNFL thickness between groups was compared using both analysis of variance and analysis of covariance.RESULTS: A total of 403 eyes of 403 subjects were included in this study. The mean age was 31.48±10.23 y. There were 180(44.7%) eyes with EM, 124(30.8%) with LM, 73(18.1%) with MM and 26(6.5%) with HM. All groups of myopia severity had a thinner average RNFL than the EM group, but after controlling for gender, age, and axial eye length, only the HM group differed significantly from the EM group(P=0.017). Likewise, the superior, inferior and nasal RNFL was thinner in all myopia groups compared to controls, but after controlling for confounders, only the inferior quadrant RNFL was significantly thinner in the HM group, when compared to the EM group(P=0.017). CONCLUSION: The average and inferior quadrant RNFL is thinner in highly myopic eyes compared to emmetropic eyes. | 3 | An in-vitro cocktail assay for assessing compound-mediated inhibition of six major cytochrome P450 enzymes显示文摘An efficient screening assay was developed and validated for simultaneous assessment of compound-mediated inhibition of six major human cytochrome P450(CYP) enzymes.This method employed a cocktail of six probe substrates(i.e.,phenacetin.amodiaquine.diclofenac,S-mephenytoin,dextromethorphan and midazolam for CYP1A2,2C8.2C9,2C19,2D6 and 3A4.respectively) as well as individual prototypical inhibitors of the six CYP enzymes in human liver microsomes under optimized incubation conditions.The corresponding marker metabolites(i.e.,acetaminophen,N-deselhylamodiaquine,4-OH-diclofenac.4-OH-Smephenytoin,dextrorphan and 1-OH-midazolam) in the incubates were quantified using LC-MS/MS methods either by an internal standaid(IS) calibration curve or a simplified analyte-to-IS peak area ratio approach.The results showed that the IC_(50) values determined by the cocktail approach were in good agreement with those obtained by the individual substrate approach as well as those reported in the literature.Besides,no remarkable difference was observed between the two quantification approaches.In conclusion,this new cocktail assay can be used for reliable screening of compound-mediated CYP inhibition. | Jing-Jing Wang Jian-Jun Guo Jenny Zhan Hai-Zhi Bu Jiunn H.Lin | 2014 | Journal of Pharmaceutical Analysis2014，4，4： | 6 |
