6Radiosensitization by microRNA30a-5p in a nude mouse model with subcutaneous lung-cancer xenograft显示文摘Objective We aimed to observe the radiosensitization effect of mir-30a-5p in a nude mouse model with subcutaneous lung-cancer xenograft and to explore the underlying mechanism.Methods A549 cell lines with either stable upregulation or downregulation of mir-30a-5p,and their negative control,were transfected with lentivirus vectors.These cell lines were used to establish a nude mouse model with subcutaneous lung-cancer xenograft.Each group was randomly divided into irradiated and non-irradiated groups.The radiosensitization effect of mir-30a-5p in vivo was studied by observing xenograft growth trends and tumor weight.The mechanisms involved in this radiosensitization were investigated by detecting expressed radiosensitization-related proteins,using immunohistochemistry and Western blotting.Results The expression level of mir-30a-5p in the lenti-mir-30a-5p group was higher than that in the negative control(lenti-GFP)group and lower in the lenti-inhibitor group(P<0.05).Subcutaneous lung-cancer xenografts in the irradiation group and lenti-mir-30a-5p increased in size slowly;tumors were lighter and tumor inhibition rates were higher than those in the non-irradiation and lenti-GFP groups.In contrast,the opposite of these effects was observed in the lenti-inhibitor group.Immunohistochemistry and Western blotting indicated that ATM protein expression level was lower in the lenti-mir-30a-5p group,with or without irradiation,compared to that in the lenti-GFP group.ATM protein levels were higher in the lenti-inhibitor groups.The phosphorylation level of ATM at residue 1981 was low in the groups without irradiation and increased significantly after irradiation(P<0.05).Moreover,the phosphorylation level was lower in the lenti-mir-30a-5p group and higher in the lenti-inhibitor group than that in the lenti-GFP group after irradiation(P<0.05).Conclusion Mir-30a-5p enhanced the radiosensitivity of nude mice with subcutaneous lung-cancer xenografts by inhibiting ATM phosphorylation.Yuyan Guo Yingtao Cui Xing Bao Yue Ke Hongtao Ren Jiyuan Pan Liping Song Hongbing Ma 2022Oncology and Translational Medicine2022,8,4:0
7泰素帝和顺铂联合放疗同步治疗中晩期宫颈癌56例的近期疗效分析(英文)显示文摘Objective: The aim of our study was to investigate the early outcome of the taxotere and cisplatin chemoradiotherapy to the advanced cervical cancer. Methods: Fifty-six cases with cervical cancer (FIGO IIb to IVa) were divided randomly into two groups in the oncology hospital of Jingzhou from September 2009 to October 2010, radiotherapy alone (28 cases) and radiation plus chemotherapy (TP) group. There was no difference of radiotherapy between the two groups, the RT + C cases who accepted TP regimen during the radiation, and DDP once weekly injection of vain, according to 20 mg/m2 and taxotere once weekly i.v. according to 35 mg/m2. These regimen were given for 4-5 weeks, and some medicine for vomiting was given to the RT + C cases. Two groups were received an oral medicine MA 160 mg every day during the treatment. Results: The early outcome: the complete remission rate was 64.3% and partial remission rate was 35.7% in RT + C. The complete remission rate was 32.1% and partial remission rate was 39.3% in RT. The total response rate and complete remission of RT + C group was higher than that of the RT group. There was significant difference between the two groups. In RT + C group, 1-year survive rate was 100.00% (28/28); in RT group, 1-year survive rate was 85.71% (24/28). There was significant difference between the two groups (χ2 = 4.31 > 3.84, P < 0.05). 8奈达铂联合甲地孕酮同步放化疗治疗中晚期宫颈癌的近期疗效分析(英文)显示文摘Objective:The aim of this study was to investigate the early outcome of the nedaplatin and megestrol combine chemoradiotherapy to the advanced cervical cancer.Methods:Forty-two cases with cervical cancer (FIGO IIb to IVa) were divided randomly into two groups,radiotherapy alone (RT group:21 cases) and radiation combines chemotherapy (nedaplatin and megestrol) (RT+ C group:21 cases).There was no difference of radiotherapy between the two groups,the RT+ C group accepted nedaplatin injection during the radiation weekly,according to 30 mg/m 2,these regimen were given for 4-5 weeks.This group was received an oral medicine megestrol 160 mg every day during the treatment.Results:The RT+ C group:the complete remission rate was 80.9% (17/21),the partial remission rate was 19.1% (4/21),the effective rate was 100%.The RT group:the complete remission rate was 38.1% (8/21) and partial remission rate was 32.9% (9/21),the effective rate was 81.0%.The total effective rate and complete remission rate of RT+ C group were higher than RT group.There was significant difference between the two groups.The 1-year survival rates respectively were 100% (21/21) in RT+ C group,80.9% (17/21) in RT group.There was statistically significant difference between the two groups (χ2=4.42>3.84,P<0.05).Conclusion:The nedaplatin and megestrol combine chemoradiotherapy can improve the early outcome of the advanced cervical cancer,and the adverse effects was raised,but that can be endured.Qinghua Ke Shiqiong Zhou Xiaoyan Su Zheng Liu Wentao Zhang Jiyuan Yang 2011The Chinese-German Journal of Clinical Oncology2011,10,8:0
9In vivo study on heparin/poly-L-lysine-copper coating for surface functionalization of ureteral stent显示文摘Polyurethane(PU)ureteral stents are used in clinics to maintain the ureteral patency.Due to biofilm formation and encrustation complications,long-term clinical usage has been limited.It is therefore necessary to develop an effective response to this unmet medical need.A heparin/poly-L-lysine/copper(NPs)coating was developed in our previous work that showed the effect of preventing infection and encrustation in vitro.In this work,a further study was conducted by grafting NPs on clinical ureteral stents that then were implanted into the infectious bladders of Wistar rats to investigate the effects of nanoparticles on bacterial growth and crystal deposition in vivo.It was found that decreased numbers of adherent microbes,urease amount splitting by bacteria,and deposited crystals were observed on the NPs stents with significant differences in comparison with PU stents.Besides,histological analysis showed that the NPs stents decreased the host tissue inflammation in close relation to the decrease biofilm formation and encrustation after 28 days of implantation.Bukola O.Awonusi Jianzhong Li Hongwei Li Zhenyu Wang Jiyuan Hu Ke Yang Jing Zhao Shi-Changxu 2022Regenerative Biomaterials2022,9,1:0
