| 1 | 绞股蓝叶水提物对糖尿病大鼠降血糖作用研究显示文摘目的探讨绞股蓝Gynostemmapentaphyllum叶水提物对链脲佐菌素(STZ)诱导的糖尿病大鼠的降血糖作用及其机制。方法采用ip STZ的方法制备糖尿病大鼠模型,将21只造模成功的SD雄性大鼠随机分为模型组和绞股蓝叶水提物低、高剂量(100、500 mg/kg)组,另取7只正常大鼠作为对照组,分别于给药2周末和3周末禁食取血,检测血浆葡萄糖(空腹血糖)、三酰甘油(TG)的浓度,实时荧光定量PCR(q RT-PCR)检测骨骼肌肿瘤坏死因子α(TNF-α)和葡萄糖转运蛋白4(GLUT4)m RNA的表达,Western blotting检测骨骼肌GLUT-4总蛋白表达,免疫荧光染色观察GLUT-4蛋白在骨骼肌肌膜上的表达分布情况。结果与对照组比较,模型组大鼠的饮食、饮水量明显增加,而体质量以及骨骼肌的质量明显下降;血浆TG和空腹血糖以及骨骼肌中TNF-αmRNA的表达明显升高,而骨骼肌GLUT-4 mRNA、GLUT-4总蛋白及骨骼肌肌膜上GLUT-4蛋白的表达明显降低;与模型组比较,高剂量的绞股蓝叶水提物明显降低了STZ诱导的糖尿病大鼠的空腹血糖,逆转了骨骼肌TNF-αmRNA和肌膜GLUT-4蛋白的表达。结论绞股蓝叶水提物能够降低STZ诱导的糖尿病大鼠的血糖,其作用机制可能与增加骨骼肌肌膜GLUT-4蛋白表达和抑制骨骼肌炎症有关。 | 王同壮 王尚 马朋 姚玲 王猛 LI Yu-hao JOHJI Yamahara 蒋丽蓉 王建伟 | 2020 | 中草药2020,51,10: | 16 |
| 2 | 生姜乙醇提取物改善果糖所致大鼠脂肪肝的作用机制研究显示文摘目的观察姜提取物改善果糖所致大鼠脂肪肝的作用,并探讨其可能的作用机制。方法取SD大鼠24只,随机分为4组,每组6只,即正常对照组(给予正常饮食饮水),模型组(给予10%果糖水),姜提取物低、高剂量组20,50 mg·kg^(-1),给予姜提取物灌胃,连续28 d。用酶法和ELISA检测血浆葡萄糖、甘油三酯(TG)、胆固醇(TC)和胰岛素含量,苏木精-伊红(HE)染色和油红-O染色观察肝脏组织中的脂肪沉积程度,RT-PCR和Western blot法检测肝脏相关基因和蛋白表达。结果姜提取物逆转了果糖过量消耗引起的大鼠肝脏甘油三酯的过度堆积(P<0.05)、肝细胞空泡化的增加和肝脏油红-O染色面积的增加(P<0.05)。然而,姜提取物并不影响大鼠食物摄入量和体质量。姜提取物能够抑制肝脏二脂酰甘油酰基转移酶(DGAT)-2 mRNA和蛋白的过表达(P<0.05),但对肝X受体(LXR)、DGAT-1、单酰甘油酰基转移酶(MGAT)-1、MGAT-2、激素敏感脂肪酶(HSL)和脂肪甘油三酯脂肪酶(ATGL)mRNA的表达无影响。结论姜提取物可能通过抑制介导肝脏甘油三酯生物合成的关键酶DGAT-2的过表达,从而改善果糖所致大鼠脂肪肝。 | 陈婷 姚玲 柯大智 左国伟 曹纬国 王尚 马朋 蒋丽蓉 Johji Yamahara Yuhao Li 王建伟 | 2016 | 中药新药与临床药理2016,27,1: | 10 |
| 3 | Ectopic hepatocellular carcinoma arising from pancreas: A case report and review of the literature显示文摘A 56-year-old man was found to have a pancreatic tail tumor. His blood chemistry showed no infection with hepatitis B or C virus and no elevations of tumor markers or pancreatic hormones. Abdominal ultrasound showed an encapsulated, rather heterogeneous, hypoechoic tumor, 6.5 cm in maximum diameter, with a beak sign. Helical dynamic CT revealed an irregularly enhanced tumor with pooling of contrast medium in the delayed phase. Abdominal angiography showed a hypervascular tumor. With a tentative diagnosis of non-functional islet-cell tumor, the patient underwent resection of the pancreatic body and tail with splenectomy. The contour of the liver and its surface were normal. In microscopic examination, tumor cells arranged in a trabecular pattern with focal bile pigment resembling hepatocellular carcinoma (HCC). Immunohistochemically, these tumor cells were positivefor HEPPAR-1, CAM5.2, cytokeratin 18 and COX-2, but negative for MUC-1, and cytokeratins 7, 20 and 8. These results supported a diagnosis of HCC without any adenocarcinoma component. The patient is currently doing well without any signs of recurrence in either the remaining pancreas or liver three years after surgery.
