| 1 | Single-cell RNA-seq highlights intra-tumoral heterogeneity and malignant progression in pancreatic ductal adenocarcinoma显示文摘Pancreatic ductal adenocarcinoma (PDAC) is the most common type of pancreatic cancer featured with high intra-tumoral heterogeneity and poor prognosis. To comprehensively delineate the PDAC intra-tumoral heterogeneity and the underlying mechanism for PDAC progression, we employed single-cell RNA-seq (scRNA-seq) to acquire the transcriptomic atlas of 57,530 individual pancreatic cells from primary PDAC tumors and control pancreases, and identified diverse malignant and stromal cell types, including two ductal subtypes with abnormal and malignant gene expression profiles respectively, in PDAC. We found that the heterogenous malignant subtype was composed of several subpopulations with differential proliferative and migratory potentials. Cell trajectory analysis revealed that components of multiple tumor-related pathways and transcription factors (TFs) were differentially expressed along PDAC progression. Furthermore, we found a subset of ductal cells with unique proliferative features were associated with an inactivation state in tumor-infiltrating T cells, providing novel markers for the prediction of antitumor immune response. | 2 | Experimental Investigation on Noise of Cavitation Nozzle and Its Chaotic Behaviour显示文摘The researches of cavitation noise mainly focus on the incipiency and developing of cavitation to prevent the cavititation erosion in the hydraulic machinery, while there is few report about the collapse strength of cavitation bubbles produced by water jet through the cavitation nozzle to utilize efficiently the collapse energy of cavitation bubbles. The cavitation noise signals are collected with hydrophones for the cavitation nozzle and general nozzle at the target position and the nozzle exit separately in the conditions of different standoff distance. The features of signal's frequency spectrum and power spectrum are analyzed for various nozzles by way of classical methods. Meanwhile, based on chaotic theory, phase space reconstruction is processed and the maximum Lyapunov index is calculated separately for each cavitation signal's time series. The results of chaotic analysis are compared with the one of conventional analysis. The analyzed data show that there are the marked differences at the spectrum between the cavitation nozzle and general nozzle at the target position while the standoff distance is 35 mm, which mainly displays at the high frequency segment (60-120 kHz). The maximum Lyapunov index calculated appear at standoff distance 35 mm, which is an optimum standoff distance for the most bubbles to collapse at the target. At the nozzle exit, the noise signal of cavitation nozzle is different from the general nozzle, which also displays at the high frequency segment. The results demonstrate that the water jet modulated by the cavitation nozzle can produce effectually cavitation, and at the target position the amplitude and energy of noise spectrum in high frequency segment for cavitation nozzle are higher than conventional nozzle and the Lyapunov index of cavitation nozzle is larger than conventional nozzle as the standoff distance is less than 55 mm. | 5 | Porphyromonas gingivalis exacerbates ulcerative colitis via Porphyromonas gingivalis peptidylarginine deiminase显示文摘Abstract:Ulcerative Colitis(UC)has been reported to be related to Porphyromonas gingivalis(P.gingivalis).Porphyromonas gingivalis peptidylarginine deiminase(PPAD),a virulence factor released by P.gingivalis,is known to induce inflammatory responses.To explore the pathological relationships between PPAD and UC,we used homologous recombination technology to construct a P.gingivalis strain in which the PPAD gene was deleted(Δppad)and aΔppad strain in which the PPAD gene was restored(comΔppad).C57 BL/6 mice were orally gavaged with saline,P.gingivalis,Δppad,or comΔppad twice a week for the entire 40 days(days 0-40),and then,UC was induced by dextran sodium sulfate(DSS)solution for 10 days(days 31-40).P.gingivalis and comΔppad exacerbated DDS-induced colitis,which was determined by assessing the parameters of colon length,disease activity index,and histological activity index,butΔppad failed to exacerbate DDS-induced colitis.Flow cytometry and ELISA revealed that compared withΔppad,P.gingivalis,and comΔppad increased T helper 17(Th17)cell numbers and interleukin(IL)-17 production but decreased regulatory T cells(Tregs)numbers and IL-10 production in the spleens of mice with UC.We also cocultured P.gingivalis,Δppad,or comΔppad with T lymphocytes in vitro and found that P.gingivalis and comΔppad significantly increased Th17 cell numbers and decreased Treg cell numbers.Immunofluorescence staining of colon tissue paraffin sections also confirmed these results.The results suggested that P.gingivalis exacerbated the severity of UC in part via PPAD. | Xida Zhao Jingbo Liu Chong Zhang Ning Yu Ze Lu Shuwei Zhang Yuchao Li Qian Li Junchao Liu Dongjuan Liu Yaping Pan | 2021 | International Journal of Oral Science2021,13,3: | 5 |

| 10 | Neuroprotective effects of cromakalim on cerebral ischemia-reperfusion injury in rats Correlation with hippocampal metabotropic glutamate receptor 1 alpha and glutamate transporter 1显示文摘BACKGROUND:Studies have reported that potassium channel openers exhibit a protective effect on cerebral ischemia-reperfusion injury and inhibit glutamate excitotoxicity in rats.However,the effects of the glutamate receptor 1α and glutamate transporter 1 remain poorly understood.OBJECTIVE:To investigate the prophylactic use of the adenosine triphosphate-sensitive potassium channel opener cromakalim on neurological function and cerebral infarct size,as well as glutamate receptor 1α and glutamate transporter 1 expression,in rats with cerebral ischemia-reperfusion injury,and to explore action mechanisms underlying reduced glutamate excitotoxicity and neuroprotection in rats.DESIGN,TIME AND SETTING:Randomized,controlled,animal experiment was performed at the Brain Institute,Qingdao University Medical College,Between July 2008 and April 2009.MATERIALS:Cromakalim was purchased from Sigma,USA;rabbit anti-glutamate receptor 1α polyclonal antibody was offered by Wuhan Boster,China;rabbit anti-glutamate transporter 1 polyclonal antibody was offered by Santa Cruz Biotechnology,USA.METHODS:Sixty male,Wistar rats,aged 6 months,were randomly assigned to three groups (n= 20):sham-surgery,model,and cromakalim.Intraluminal thread methods were used to establish middle cerebral artery occlusion in rats from the model and cromakalim groups.Rats from the sham-surgery group were subjected to exposed common carotid artery,external carotid artery,and internal carotid artery,without occlusion.Cromakalim (10 mg/kg) was administered 30 minutes prior to middle cerebral artery occlusion,but there was no intervention in the model and sham-surgery groups.MAIN OUTCOME MEASURES:At 24 hours post-surgery,neurological behavioral functions were evaluated using Bederson's test,cerebral infarction volume was determined following tetrazolium chloride staining,and glutamate receptor 1α and glutamate transporter 1 expressions were detected using immunohistochemistry.RESULTS:Following cerebral ischemia-reperfusion injury,neurological behavioral malfunctions were obvious in all mice.Focal cerebral infarction was detected in ischemic hemispheres,glutamate receptor 1α expression increased,and glutamate transporter 1 expression decreased in the ischemic hemisphere (P < 0.05).Compared with the model group,neurological behavioral functions significantly improved,cerebral infarction volume was significantly reduced (P < 0.05),glutamate receptor 1α expression was significantly decreased,and glutamate transporter 1 expression was increased in the cromakalim group (P < 0.05).CONCLUSION:Improved neurological function and reduced cerebral infarction volume in rats through the preventive use of cromakalim could be related to decreased glutamate receptor 1α expression and enhanced glutamate transporter 1 expression. | Shilei Wang Junchao Liu Qingxian Chang Yu Li, Yan Jiang Shiduan Wang | 2010 | Neural Regeneration Research2010,5,9: | 2 |

| 15 | Stego key searching for LSB steganography on JPEG decompressed image显示文摘At present, steganalysis research focuses on detecting the existence of a hidden message. However,extracting the hidden information, i.e., an extracting attack, is crucial in obtaining effective evidence in computer forensics. Due to the difficulty of an extracting attack, research in this field is limited. In steganography with a stego key, an extracting attack is equivalent to recovering the stego key. In this paper we study a method for recovering the stego key in least significant bit(LSB) steganography with a decompressed JPEG image as the cover image. Firstly, the recovery of the stego key is translated into a cryptanalysis problem for a sequential cipher. The method for recovering the stego key is based on estimating the modification positions. The minimum size of the data used to recover the stego key successfully is discussed. Secondly, when a decompressed JPEG image is used as the cover image, the probability of recovering the cover pixels using recompression is discussed.Recompression is used to compute the error of the estimated sequence. Finally, an algorithm to recover the stego key in LSB steganography with a decompressed JPEG image as the cover image is proposed. | 16 | Highly time-resolved aerosol characteristics during springtime in Weizhou Island显示文摘A single particle aerosol mass spectrometer was deployed to measure the changes of single particle species and sizes during March 2015 in Weizhou Island of the Beibu Gulf, Guangxi province, South China. In this campaign, a total of 3,100,597 particles were sized, and 25.8%particles with both positive and negative mass spectrum were collected and 24.8%characterized in combination with the ART-2 a neural network algorithm. The distribution of sized particles was mainly in from 520 to 600 nm, and the diameters ranging from 340 to1000 nm accounted for above 90%. Eight types of particles were classified: Elemental Carbon containing(EC), Organic Carbon containing(OC), EC and OC combined containing particles,Na containing particles, K containing particles(K), Levoglucosan containing particles,mineral containing particles, and Heavy Metal containing particles(HM). EC, OC and K were the major containing particles, which accounted for 84.3% in the eight types particles. The relative ratio and size distribution of the three types were EC(48.1%, 620 nm), OC(12.7%,440 nm), and K(23.5%, 600 nm), respectively.
