1A bioinformatics method for predicting long noncoding RNAs associated with vascular disease显示文摘Long noncoding RNAs(lncRNAs)play important roles in human diseases including vascular disease.Given the large number of lncRNAs,however,whether the majority of them are associated with vascular disease remains unknown.For this purpose,here we present a genomic location based bioinformatics method to predict the lncRNAs associated with vascular disease.We applied the presented method to globally screen the human lncRNAs potentially involved in vascular disease.As a result,we predicted 3043 putative vascular disease associated lncRNAs.To test the accuracy of the method,we selected 10 lncRNAs predicted to be implicated in proliferation and migration of vascular smooth muscle cells(VSMCs)for further experimental validation.The results confirmed that eight of the 10 lncRNAs(80%)are validated.This result suggests that the presented method has a reliable prediction performance.Finally,the presented bioinformatics method and the predicted vascular disease associated lncRNAs together may provide helps for not only better understanding of the roles of lncRNAs in vascular disease but also the identification of novel molecules for the diagnosis and therapy of vascular disease.LI JianWei GAO Cheng WANG YuChen MA Wei TU Jian WANG JunPei CHEN ZhenZhen KONG Wei CUI QingHua 2014Science China(Life Sciences)2014,57,8:2
2Construction of Expression Vector for Porcine Gastrin-releasing Peptide Fusion Protein显示文摘[Objectives]This study was conducted to obtain porcine gastrin-releasing peptide(GRP)fusion protein.[Methods]The constructed pET32a(+)-GRP plasmid was transformed into Escherichia coli BL21(DE3)competent cells to obtain the pET32a(+)-GRP-BL21(DE3)fusion protein expression strain,which was induced with 0.5 mM IPTG at 25℃and 150 r/min for 12 h,and the His-tagged GRP fusion protein was detected by SDS-Page gel electrophoresis and Western Blot.[Results]After optimizing the IPTG-induced expression conditions,it was confirmed that the porcine GRP fusion protein was obtained,and the porcine GRP fusion protein was soluble,stable and highly active.[Conclusions]This study lays a foundation for the subsequent preparation of anti-pig GRP antibodies.Zhiyu MA Jie ZHANG Junpei GUO Zhuo MA Chang YU Ying ZHANG Jinlong ZHANG 2022Agricultural Biotechnology2022,11,3:1
3Optical spectroscopy study of Ca_3(Ru_(0.91)Mn_(0.09))_2O_7 single crystal in high magnetic fields显示文摘The magneto-optical spectrum, with magnetic fields up to 42 T, of double layered ruthenates Ca_3(Ru_(0.91)Mn_(0.09))_2O_7(CRMO) single crystal is studied. Both the temperature and magnetic field induced insulator-to-metal transitions(IMTs) are observed via magneto-optical properties of the crystal. The critical magnetic field(H//c) of IMT for CRMO is found to be as large as 35 T at 5 K. The fine structure of optical spectra identified the antiferromagnetic/ferro-orbital-ordering configurations of Ru 4d orbitals at low temperatures. Meanwhile, the configuration of orbital polarization of such double-layer CRMO single crystal is discussed. 4Effects of LPS on the Gene Expression of NMB and Its Receptor in the Hypothalamic-pituitary-testicular Axis of Rabbits显示文摘[Objectives]This study was conducted to investigate the effect of lipopolysaccharide(LPS)on the gene expression levels of neuromedin B(NMB)and its receptor(NMBR)in the hypothalamic-pituitary-testis(HPT)axis of male rabbits.[Methods]Forty male New Zealand white rabbits were randomly divided into an experimental group injected with LPS and a control group.At 0,1.5,3,6 and 12 h after LPS injection,four rabbits in each group were sacrificed,and the hypothalamus,pituitary and testicular tissues were collected.RT-PCR was carried out to detect the expression of rabbit NMB and NMBR mRNA in the HPT axis.[Results]NMB and NMBR mRNA were expressed in the hypothalamus,pituitary gland and testis of rabbits,and there was a certain time effect,indicating that the NMB/NMBR system is involved in the activities of the hypothalamus-pituitary-testis(HPT)axis.At 1.5 h after LPS stimulation,the expression of NMB mRNA in the hypothalamus increased significantly,and showed an extremely significant difference from the control group(P<0.01),while the expression of NMBR mRNA in the hypothalamus increased,but was not significantly different from the control group(P>0.05).The expression levels of NMB and NMBR mRNA in the pituitary and testis tissues in the HPT axis at different time after the LPS treatment were not significantly different from those in the control group(P>0.05).Therefore,LPS stimulation still has a certain effect on the expression of NMB and its receptor mRNA in the HPT axis.[Conclusions]This study is of great significance for the in-depth study on the mechanism of damage caused by immune stress to livestock and poultry reproduction.Zhiyu MA Ying ZHANG Junpei GUO Jinlong ZHANG 2020Agricultural Biotechnology2020,9,6:0
5Intelligent Industrial Auxiliary System Based on AR Technology显示文摘The traditional industrial assembly method is inefficient,and it is difficult to meet the needs of rapid production in modern society.Although many virtual assembly systems based on augmented reality technology have appeared in recent years,most of these assembly systems are noninteractive,nonintelligent,and inefficient.In response to this problem,we combined AR technology and AI technology to design and implement a strong interactive,high-intelligence virtual assembly guidance system in four levels.Finally,we took a UAV assembly as an example to show our design results.Operators can use this system to quickly master the assembly process.Finally,according to some problems encountered in the process of system implementation,the improvement direction of the virtual assembly system is proposed,and the future development of the AR auxiliary industry is proposed.This article describes the functions and implementation process of the four levels of the AR assembly system in detail,which can help readers quickly understand the connection between AR technology and AI technology and understand the principle of the virtual assembly guidance system.Tao Wang Xinqi Shen Junpei Ma Zhuorui Chang Linyan Guo 2022国际计算机前沿大会会议论文集2022,,2:0
6Industry-Oriented Cloud Edge Intelligent Assembly Guidance System显示文摘In view of the low efficiency of production and assembly in traditional industries,we use AR technology to replace traditional assembly instructions and design an industry-oriented cloud-edge intelligent assembly guidance system.Since the computing power ofARglasses cannot meet the high complexity requirements of scene understanding,we adopt the joint solution of Cloud-Edge.First,the sensor data collected by the AR glasses are streamed to the edge server using high-speed and low-latency wireless interconnection technology.Then,the product artifacts in the data scene are identified and understood through the instance segmentation network BlendMask based on deep learning.Then,the 3D pose of the object is calculated in real time by combining pose estimation and 3D reconstruction.Furthermore,an accurate 3D guidance animation is generated,and the virtual 3D model in the AR glasses is accurately superimposed on the real object to determine whether the assembly is correct in real time.Experiments show that the system effectively combines artificial intelligence and intelligent manufacturing,integrates various elements in the scene in real time to provide operators with multimodal and multidimensional immersive guidance,and corrects in time when assembly errors occur.It can not only quickly guide the operator to complete the learning of the assembly process but also assist the staff in the assembly in real time.Ultimately,it improves assembly speed and accuracy,which in turn improves enterprise productivity.Difei Liu Zhuorui Chang Junpei Ma Tao Wang Mei Li 2022国际计算机前沿大会会议论文集2022,,2:0
7Fidgetin interacting with microtubule end binding protein EB3 affects axonal regrowth in spinal cord injury显示文摘Fidgetin,a microtubule-severing enzyme,regulates neurite outgrowth,axonal regeneration,and cell migration by trimming off the labile domain of microtubule polymers.Because maintenance of the microtubule labile domain is essential for axon initiation,elongation,and navigation,it is of interest to determine whether augmenting the microtubule labile domain via depletion of fidgetin serves as a therapeutic approach to promote axonal regrowth in spinal cord injury.In this study,we constructed rat models of spinal cord injury and sciatic nerve injury.Compared with spinal cord injury,we found that expression level of tyrosinated microtubules in the labile portion of microtubules continuously increased,whereas fidgetin decreased after peripheral nerve injury.Depletion of fidgetin enhanced axon regeneration after spinal cord injury,whereas expression level of end binding protein 3(EB3)markedly increased.Next,we performed RNA interference to knockdown EB3 or fidgetin.We found that deletion of EB3 did not change fidgetin expression.Conversely,deletion of fidgetin markedly increased expression of tyrosinated microtubules and EB3.Deletion of fidgetin increased the amount of EB3 at the end of neurites and thereby increased the level of tyrosinated microtubules.Finally,we deleted EB3 and overexpressed fidgetin.We found that fidgetin trimmed tyrosinated tubulins by interacting with EB3.When fidgetin was deleted,the labile portion of microtubules was elongated,and as a result the length of axons and number of axon branches were increased.These findings suggest that fidgetin can be used as a novel therapeutic target to promote axonal regeneration after spinal cord injury.Furthermore,they reveal an innovative mechanism by which fidgetin preferentially severs labile microtubules.Chao Ma Junpei Wang Qifeng Tu Weijuan Bo Zunlu Hu Run Zhuo Ronghua Wu Zhangji Dong Liang Qiang Yan Liu Mei Liu 2023Neural Regeneration Research2023,18,12:0
