1Influence of Water Stress on Endogenous Hormone Contents and Cell Damage of Maize Seedlings显示文摘Phytohormones play critical roles in regulating plant responses to stress. We investigated the effects of water stress induced by adding 12% (w/v) polyethylene glycol to the root medium on the levels of abscisic acid (ABA), indole-3-acid (IAA), zeatin (ZT), and gibberellin3 (GA3) in maize leaves. The results suggested that water stress had significant effects on the four hormone levels. There was a transient increase in the IAA content during the initial stage of adaptation to water stress in maize leaves, but it dropped sharply thereafter in response to water stress. ABA content increased dramatically in maize leaves after 24 h of exposure to water stress, and then the high levels of ABA were maintained to the end. The contents of ZT and GA3 rapidly declined in maize leaves subjected to water stress. The effects of water stress on chlorophyll content, electrolyte leakage and malondialdehyde levels in maize leaves were also studied. The variation of cell damage was negatively correlated with ZT and GA3 levels in maize leaves under water stress. Thus, we explored the roles of ZT and GA3 on the growth of maize seedlings under water stress by exogenous application. 2Genetic transformation of calli from maize and regeneration of herbicide-resistant plantlets显示文摘The herbicide-resistant gene als of Arabidopsis thaliana has been transferred into embryonic calli of maize by microprojectile bombardment. We have obtained chlor-sulfuron-resistant calli and regenerated plantlets through selection by herbicide chlorsulfuron. The results of PCR analysis and Southern blotting indicate that gene als has been transferred into some plantlets. The test of spraying chlorsulfuron indicated that the transgenic plantlets had favorable herbicide-resistant trait. The purpose of the research was to obtain chiorsulfuron-resistant transgenic maize and hope that this kind of high efficient herbicide could be widely used in rotation soil of wheat and maize. 15Comparative Studies on Preparation of Large Plant DNA Fragments by Pulse Field Gel Electrophoresis显示文摘The preparation of large plant DNA fragments is extremely important to the construction of large insert DNA libraries (YAC, BAC, PAC and TAC). Although several techniques have been developed in each step of large plant DNA fragments preparation, the whole processing remains complicated and difficult. Based on authors research experience and the recent worldwide development in this field, the following aspects are discussed in this paper: techniques of plant high molecular weight (HMW) DNA purification by pre-electrophoresis, the optimal conditions for the partial digestion of the HMW DNA by HindIII, the isolation effects of of large plant DNA fragments (100~400 kb) with different parameters of pulse field gel electrophoresis (PFGE), and the recovery of large DNA fragments. Through comparative studies, the advantages and disadvantages of each technique are discussed and some recommendations are proposed for preparing high quality large plant DNA fragments. The suggested techniques have been used in preparing the large DNA fragments of maize, rice, moss, laver, sea tangle and peach,and similar results are obtained among all the materials. 17Cloning and molecular characterization of a gene encoding MAP kinase from maize and its expression in E.coli显示文摘A new MAPK gene, ZmSIMK1 (Zea mays L. salt-induced mitogen-activated protein kinase 1), is isolated from a maize cDNA library. The full-length ZmSIMK1 gene contains 1636 bp and an open reading frame of 1122 nucleotides capable of encoding 373 amino acid polypeptides with a predicted molecular mass of 42.3 kda and pI of 6.01. The putative ZmSIMK1 protein contains all 11 conserved subdomains that are characteristics of serine/threonine protein kinases and the TEY motif, which is the putative phosphorylation site. Northern blot analysis shows that ZmSIMK1 is ubiquitously expressed in roots, stems, and leaves of maize seedlings and its mRNA accumulation is observed in maize seedlings treated with 30 mmol/L PEG-6000 and 137 mmol/L NaCl, but the expression of ZmSIMK1 is not significantly affected by 4℃ treatment. The expression vector pET-ZmSIMK1 is constructed by inserting the coding region of ZmSIMK1 cDNA into pET-42a(+), and transformed into E. coli strain BL21(DE3).
