6Nigral dopaminergic neuron replenishment in adult mice through VE-cadherin-expressing neural progenitor cells显示文摘The function of dopaminergic neurons in the substantia nigra is of central importance to the coordination of movement by the brain’s basal ganglia circuitry.This is evidenced by the loss of these neurons,resulting in the cardinal motor deficits associated with Parkinson’s disease.In order to fully understand the physiology of these key neurons and develop potential therapies for their loss,it is essential to determine if and how dopaminergic neurons are replenished in the adult brain.Recent work has presented evidence for adult neurogenesis of these neurons by Nestin^+/Sox2~–neural progenitor cells.We sought to further validate this finding and explore a potential atypical origin for these progenitor cells.Since neural progenitor cells have a proximal association with the vasculature of the brain and subsets of endothelial cells are Nestin^+,we hypothesized that dopaminergic neural progenitors might share a common cell lineage.Therefore,we employed a VE-cadherin promoter-driven CRE^(ERT2):TH^(lox)/TH^(lox) transgenic mouse line to ablate the tyrosine hydroxylase gene from endothelial cells in adult animals.After 26 weeks,but not 13 weeks,following the genetic blockade of tyrosine hydroxylase expression in VE-cadherin^+cells,we observed a significant reduction in tyrosine hydroxylase^+neurons in the substantia nigra.The results from this genetic lineage tracing study suggest that dopaminergic neurons are replenished in adult mice by a VE-cadherin^+progenitor cell population potentially arising from an endothelial lineage.Abir A.Rahman Nathan K.Lai Joshua E.Albright Paige E.Urquhart Abby R.Webb Brad E.Morrison 2017Neural Regeneration Research2017,12,11:1
7Effects of Porosity on Piezoelectric Characteristics of Polyvinylidene Fluoride Films for Biomedical Applications显示文摘Objective:The objective of this work is to study the effects of porosity on mechanical and piezoelectric properties of polyvinylidene fluoride(PVDF)films for biomedical applications.Impact Statement:By investigating the piezoelectric properties of PVDF and the porosity effect on its electromechanical performance,there is potential for further development of PVDF as a hemodynamic sensor that can lead to further technological advancements in the biomedical field,benefiting patients and physicians alike.Introduction:PVDF thin films have shown potential in the application of hemodynamic flow sensing and monitoring the effects on blood flow caused by prosthetic valve implantation via the transcatheter aortic valve replacement operation.The piezoelectric performance of PVDF films can be influenced by the porosity of the material.Methods:In this study,strain tracking was performed on thin film PVDF specimens with various levels of porosity and pore sizes to determine the mechanical properties of the specimens.The mechanical properties were used to model the PVDF material in COMSOL multiphysics software,in which compression test simulations were performed to determine the piezoelectric coefficient d33 of the PVDF.Results:A decline in the elastic modulus was found to be highly inversely correlated with porosity of the specimens and the simulation results show that elastic modulus had a much greater effect on the piezoelectric properties than Poisson’s ratio.Conclusion:A combination of experimental and computational techniques was able to characterize and correlate the mechanical properties of PVDF films of varying porosities to their piezoelectric properties.Jack T.Kloster Matthew J.Danley Victor K.Lai Ping Zhao 2023Biomedical Engineering Frontiers2023,4,1:0
