| 3 | Development of nanoscale drug delivery systems of dihydroartemisinin for cancer therapy: A review显示文摘Dihydroartemisinin(DHA),a first-line antimalarial drug,has demonstrated great anticancer effects in many types of tumors,including liver cancer,glioblastoma,and pancreatic cancer.Due to its abilities to induce programmed cell death(PCD;apoptosis,autophagy and ferroptosis),inhibit tumor metastasis and angiogenesis,and modulate the tumor microenvironment,DHA could become an antineoplastic agent in the foreseeable future.However,the therapeutic efficacy of DHA is compromised owing to its inherent disadvantages,including poor stability,low aqueous solubility,and short plasma halflife.To overcome these drawbacks,nanoscale drug delivery systems(NDDSs),such as polymeric nanoparticles(NPs),liposomes,and metal-organic frameworks(MOFs),have been introduced to maximize the therapeutic efficacy of DHA in either single-drug or multidrug therapy.Based on the beneficial properties of NDDSs,including enhanced stability and solubility of the drug,prolonged circulation time and selective accumulation in tumors,the outcomes of DHA-loaded NDDSs for cancer therapy are significantly improved compared to those of free DHA.This reviewfirst summarizes the current understanding of the anticancer mechanisms of DHA and then provides an overview of DHA-including nanomedicines,aiming to provide inspiration for further application of DHA as an anticancer drug. | Ka Hong Wong Donglin Yang Shanshan Chen Chengwei He Meiwan Chen | 2022 | Asian Journal of Pharmaceutical Sciences2022,17,4: | 3 |
| 4 | Aortic heterogeneity across segments and under high fat/salt/glucose conditions at the single-cell level显示文摘The aorta,with ascending,arch,thoracic and abdominal segments,responds to the heartbeat,senses metabolites and distributes blood to all parts of the body.However,the heterogeneity across aortic segments and how metabolic pathologies change it are not known.Here,a total of 216612 individual cells from the ascending aorta,aortic arch,and thoracic and abdominal segments of mouse aortas under normal conditions or with high blood glucose levels,high dietary salt,or high fat intake were profiled using single-cell RNA sequencing.We generated a compendium of 10 distinct cell types,mainly endothelial(EC),smooth muscle(SMC),stromal and immune cells.The distributions of the different cells and their intercommunication were influenced by the hemodynamic microenvironment across anatomical segments,and the spatial heterogeneity of ECs and SMCs may contribute to differential vascular dilation and constriction that were measured by wire myography.Importantly,the composition of aortic cells,their gene expression profiles and their regulatory intercellular networks broadly changed in response to high fat/salt/glucose conditions.Notably,the abdominal aorta showed the most dramatic changes in cellular composition,particularly involving ECs,fibroblasts and myeloid cells with cardiovascular risk factor-related regulons and gene expression networks.Our study elucidates the nature and range of aortic cell diversity,with implications for the treatment of metabolic pathologies. | Dongxu He Aiqin Mao Chang-Bo Zheng Hao Kan Ka Zhang Zhiming Zhang Lei Feng Xin Ma | 2020 | National Science Review2020,7,5: | 2 |
| 5 | Walking pace and the risk of stroke:A meta-analysis of prospective cohort studies显示文摘Purpose:The extent to which walking pace is associated with a reduced risk for stroke remains unclear.This study examined the association between walking pace and stroke risk based on prospective cohort studies.Methods:Databases of PubMed,EMBASE,Web of Science,Scopus,and China National Knowledge Internet were searched from the inception dates to January 31,2019,for prospective cohort studies focusing on walking pace and risk of stroke in adults.Two reviewers independently extracted data and assessed the quality of the studies.The dependent measure was stroke incidence.Using random-effects models,a metaanalysis was performed to estimate the overall relative risks(RR)of stroke incidence and 95%confidence intervals(CIs)for the individuals with the fastest walking paces vs.individuals with the slowest walking paces.A dose-response relationship was also examined.Results:After screening 1294 titles/abstracts and 14 full-text studies identified in the search,7 studies(from 8 cohorts)were included in the metaanalysis.The 7 studies included a total of 135,645 participants(95.2%women;mean age 63.6 years)and 2229 stroke events(median follow-up time=8.0 years).Compared to individuals in the slowest walking-pace category(median=1.6 km/h),individuals in the fastest walking-pace category(median=5.6 km/h)had a 44%lower risk of stroke(pooled RR=0.56,95%CI:0.48-0.65).There was also a linear dose-response relationship(RR=0.87;95%CI:0.83-0.91),with the risk of stroke decreased by 13%for every 1 km/h increment in baseline walking pace.We observed similar results across walking-pace assessment,type of stroke ascertainment,stroke subtypes,sex,sample size,and duration of follow-up.Conclusion:Findings from this meta-analysis indicate that walking pace is inversely associated with the risk of stroke. | Minghui Quan Pengcheng Xun Ru Wang Ka He Peijie Chen | 2020 | Journal of Sport and Health Science2020,9,6: | 2 |
