| 1 | Therapeutic efficacy of anti-CD19 CAR-T cells in a mouse model of systemic lupus erythematosus显示文摘Dysregulated B-cell activation plays pivotal roles in systemic lupus erythematosus(SLE),which makes B-cell depletion a potential strategy for SLE treatment.The clinical success of anti-CD19 CAR-T cells in treating B-cell malignancies has attracted the attention of researchers.In this study,we aimed to investigate the feasibility of applying anti-CD19 CAR-T cell therapy to SLE treatment in a mouse disease model.We constructed murine anti-CD19 CARs with either CD28 or 4-1BB as the intracellular costimulatory motif and evaluated the therapeutic function of the corresponding CAR-T cells by infusing them into MRL-lpr mice.Furthermore,anti-CD19 CAR-T cells were transferred to MRL-lpr mice before the onset of disease to determine their role in SLE prevention.According to our observations,compared with antibody treatment,the adoptive transfer of our anti-CD19 CAR-T cells showed a more sustained B-cell-depletion effect in MRL-lpr mice.The transfer of syngeneic anti-CD19 CAR-T cells not only prevented disease pathogenesis before the onset of disease symptoms but also displayed therapeutic benefits at a later stage after disease progression.We also tried to optimize the treatment strategy and found that compared with CAR-T cells with the CD28 costimulatory motif,CAR-T cells with the 4-1BB costimulatory motif showed better therapeutic efficiency without cell enrichment.Taken together,these results show that anti-CD19 CAR-T cell therapy was effective in the prevention and treatment of a murine model of SLE,indicating its potential for clinical use in patients. | Xuexiao Jin Qin Xu Chengfei Pu Kaixiang Zhu Cheng Lu Yu Jiang Lei Xiao Yongmei Han Linrong Lu | 2021 | Cellular & Molecular Immunology2021,18,8: | 12 |

| 3 | Dark matter direct search sensitivity of the PandaX-4T experiment显示文摘The Panda X-4T experiment, a 4-ton scale dark matter direct detection experiment, is being planned at the China Jinping Underground Laboratory. In this paper we present a simulation study of the expected background in this experiment. In a 2.8-ton fiducial mass and the signal region between 1-10 keV electron equivalent energy, the total electron recoil background is found to be 4.9 × 10^(-5) kg^(-1) d^(-1) keV^(-1). The nuclear recoil background in the same region is 2.8 × 10^(-7) kg^(-1) d^(-1) keV^(-1). | 4 | Remaining Useful Life Prediction for a Roller in a Hot Strip Mill Based on Deep Recurrent Neural Networks显示文摘Accurate estimation of the remaining useful life(RUL)and health state for rollers is of great significance to hot rolling production.It can provide decision support for roller management so as to improve the productivity of the hot rolling process.In addition,the RUL prediction for rollers is helpful in transitioning from the current regular maintenance strategy to conditional-based maintenance.Therefore,a new method that can extract coarse-grained and fine-grained features from batch data to predict the RUL of the rollers is proposed in this paper.Firstly,a new deep learning network architecture based on recurrent neural networks that can make full use of the extracted coarsegrained fine-grained features to estimate the heath indicator(HI)is developed,where the HI is able to indicate the health state of the roller.Following that,a state-space model is constructed to describe the HI,and the probabilistic distribution of RUL can be estimated by extrapolating the HI degradation model to a predefined failure threshold.Finally,application to a hot strip mill is given to verify the effectiveness of the proposed methods using data collected from an industrial site,and the relatively low RMSE and MAE values demonstrate its advantages compared with some other popular deep learning methods. | Ruihua Jiao Kaixiang Peng Jie Dong | 2021 | IEEE/CAA Journal of Automatica Sinica2021,8,7: | 5 |

| 6 | A novel entry point for pedicle screw placement in the thoracic spine显示文摘This study was aimed to introduce a novel entry point for pedicle screw fixation in the thoracic spine and compare it with the traditional entry point. A novel entry point was found with the aim of improving accuracy, safety and stability of pedicle screw technique based on anatomical structures of the spine. A total of 76 pieces of normal thoracic CT images at the transverse plane and the thoracic pedicle anatomy of 6 cadaveric specimens were recruited.Transverse pedicle angle(TPA), screw length, screw placement accuracy rate and axial pullout strength of the two different entry point groups were compared. There were significant differences in the TPA, screw length, and the screw placement accuracy rate between the two groups(P<0.05). The maximum axial pullout strength of the novel entry point group was slightly larger than that of the traditional group. However, the difference was not significant(P > 0.05). | 7 | Priming of NLRP3 inflammasome activation by Msn kinase MINK1 in macrophages显示文摘The nucleotide-binding domain,leucine-rich-repeat containing family,pyrin domain-containing 3(NLRP3)inflammasome is essential in inflammation and inflammatory disorders.Phosphorylation at various sites on NLRP3 differentially regulates inflammasome activation.The Ser725 phosphorylation site on NLRP3 is depicted in multiple inflammasome activation scenarios,but the importance and regulation of this site has not been clarified.The present study revealed that the phosphorylation of Ser725 was an essential step for the priming of the NLRP3 inflammasome in macrophages.We also showed that Ser725 was directly phosphorylated by misshapen(Msn)/NIK-related kinase 1(MINK1),depending on the direct interaction between MINK1 and the NLRP3 LRR domain.MINK1 deficiency reduced NLRP3 activation and suppressed inflammatory responses in mouse models of acute sepsis and peritonitis.Reactive oxygen species(ROS)upregulated the kinase activity of MINK1 and subsequently promoted inflammasome priming via NLRP3 Ser725 phosphorylation.Eliminating ROS suppressed NLRP3 activation and reduced sepsis and peritonitis symptoms in a MINK1-dependent manner.Altogether,our study reveals a direct regulation of the NLRP3 inflammasome by Msn family kinase MINK1 and suggests that modulation of MINK1 activity is a potential intervention strategy for inflammasomerelated diseases. | Kaixiang Zhu Xuexiao Jin Zhexu Chi Sheng Chen Songquan Wu Richard D.Sloan Xuai Lin Dante Neculai Di Wang Hu Hu Linrong Lu | 2021 | Cellular & Molecular Immunology2021,18,10: | 2 |

In order to make the controlled system stable and having good performance, H∞ control theory was put forward to solve this practical problem. Taking the position of a rolling mill as the controlled object, it was rectified by optimal engineering way. | 19 | N,O co-doped porous carbon with rich pseudocapacitive groups exhibiting superior energy density in an acidic 2.4 V Li_(2)SO_(4)electrolyte显示文摘Designing a carbon material with a unique composition and surface functional groups for offering high specific capacity in a wide voltage window is of great significance to improve the energy density for the supercapacitor in a cheap and eco-friendly aqueous electrolyte.Herein,we develop an efficient strategy to synthesize a N,O co-doped hierarchically porous carbon(NODPC-1.0)with moderate specific surface area and pore volume as well as rich heteroatoms using a deep eutectic solvent(DES)as an activator.It is found that NODPC-1.0 with a large proportion of pseudocapacitive functional groups(pyrrole-N,pyridineN and carbonyl-quinone)can work stable in an acidic 2 mol/L Li_(2)SO_(4)(pH 2.5)electrolyte,exhibiting specific capacities of 375 and 186 F/g at the current densities of 1.0 and 100 A/g,respectively.Also,the assembled symmetric capacitor using the NODPC-1.0 as the active material and 2 mol/L acidic Li_(2)SO_(4)(pH 2.5)as the electrolyte shows an outstanding energy density of 74.4 Wh/kg at a high power density of 1.44 k W/kg under a broad voltage window(2.4 V).Relevant comparative experiments indicate that H+of the acidic aqueous electrolyte plays a crucial part in enhancement the specific capacity,and the abundant pseudocapacitive functional groups on the surface of the NODPC-1.0 sample play the key role in the improvement of electrochemical cycle stability under a broad voltage window. | Xiao Wang Kaixiang Zou Weijing Wu Yuanfu Deng Guohua Chen | 2022 | Chinese Chemical Letters2022,33,8: | 0 |
