1Phase 1 human trial of autologous bone marrow-hematopoietic stem cell transplantation in patients with decompensated cirrhosis显示文摘AIM: To evaluate safety and feasibility of autologous bone marrow-enriched CD34+ hematopoietic stem cell Tx through the hepatic artery in patients with decompensated cirrhosis.METHODS: Four patients with decompensated cirrhosis were included. Approximately 200 mL of the bone marrow of the patients was aspirated, and CD34+ stem cells were selected. Between 3 to 10 million CD34+ cells were isolated. The cells were slowly infused through the hepatic artery of the patients.RESULTS: Patient 1 showed marginal improvement in serum albumin and no significant changes in other test results. In patient 2 prothrombin time was decreased; however, her total bilirubin, serum creatinine, and Model of End-Stage Liver Disease (MELD) score worsened at the end of follow up. In patient 3 there was improvement in serum albumin, porthrombin time (PT), and MELD score. Patient 4 developed radiocontrast nephropathy after the procedure, and progressed to type 1 hepatorenal syndrome and died of liver failure a few days later. Because of the major side effects seen in the last patient, the trial was prematurely stopped.CONCLUSION: Infusion of CD34+ stem cells through the hepatic artery is not safe in decompensated cirrhosis. Radiocontrast nephropathy and hepatorenal syndrome could be major side effects. 2Novel hepatocellular carcinoma molecules with prognostic and therapeutic potentials显示文摘Hepatocellular carcinoma(HCC), the predominant form of primary liver cancer, is the sixth most common cancer worldwide and the third leading cause of cancerrelated death. The difficulty to diagnose early cancer stages, the aggressive behaviors of HCC, and the poor effectiveness of therapeutic treatments, represent the reasons for the quite similar deaths per year and incidence number. Considering the fact that the diagnosis of HCC typically occurs in the advanced stages of the disease when the therapeutic options have only modest efficacy, the possibility to identify early diagnostic markers could be of significant benefit. So far, a large number of biomarkers have been associated to HCC progression and aggressiveness, but many of them turned out not to be of practical utility. This is the reason why active investigations are ongoing in this field. Given the huge amount of published works aimed at the identification of HCC biomarkers, in this review we mainly focused on the data published in the last year, with particular attention to the role of(1) molecular and biochemical cellular markers;(2) micro-interfering RNAs;(3) epigenetic variations; and(4) tumor stroma. It is worth mentioning that a significant number of the HCC markers described in the present review may be utilized also as targets for novel therapeutic approaches, indicating the tight relation between diagnosis and therapy. 3Hot deformation behavior of microstructural constituents in a duplex stainless steel during high-temperature straining显示文摘The hot deformation characteristics of 1.4462 duplex stainless steel(DSS)were analyzed by considering strain partitioning between austenite and ferrite constituents.The individual behavior of ferrite and austenite in microstructure was studied in an iso-stress condition.Hot compression tests were performed at temperatures of 800-1100 C and strain rates of 0.001-1 s 1.The flow stress was modeled by a hyperbolic sine constitutive equation,the corresponding constants and apparent activation energies were determined for the studied alloys.The constitutive equation and law of mixture were used to measure the contribution factor of each phase at any given strain.It is found that the contribution factor of ferrite exponentially declines as the Zener-Hollomon parameter(Z)increases.On the contrary,the austenite contribution polynomially increases with the increase of Z.At low Z values below 2.6.×1015(lnZ=35.5),a negative contribution factor is determined for austenite that is attributed to dynamic recrystallization.At high Z values,the contribution factor of austenite is about two orders of magnitude greater than that of ferrite,and therefore,austenite can accommodate more strain.Microstructural characterization via electron back-scattered difraction(EBSD)confirms the mechanical results and shows that austenite recrystallization is possible only at high temperature and low strain rate.Amir Momeni Shahab Kazemi Ali Bahrani 2013International Journal of Minerals,Metallurgy and Materials2013,20,10:11
4油藏数值模拟自动历史拟合方法——以Nelson油田为例显示文摘针对北海中部Nelson油田,研究油藏数值模拟自动历史拟合方法。完整的油藏自动历史拟合工作流程包括选择要调整的油藏变量、选择参数调整方法、自动历史拟合、数据分析、组合最优结果得到最优的油藏模型集5个步骤。以净毛比、水平渗透率、垂直渗透率为调整参数,采用全局单变量法、区域多变量法、局部多变量法等3种油藏参数调整方法,并结合生产数据和时移地震数据,对Nelson油田进行自动历史拟合。结果表明,局部参数调整方法减少了模拟中模型的数目和计算时间,提高了模拟精度,使得油藏中油水产量拟合误差大大降低,在Nelson油田应用效果最好。全局单变量法适合于历史拟合参数相互独立的情况;对于井网密度大且井间关联性强、历史拟合参数相互影响的油藏,最好选用区域多变量法进行参数调整;对于油藏历史拟合参数相互影响但每个选定调整区域相互独立的油藏,局部多变量法更加适用。Kazemi Alireza Stephen Karl D 2012石油勘探与开发2012,39,3:10
5Nonlinear adaptive optimal control for vehicle handling improvement through steer-by-wire system显示文摘A control algorithm for improving vehicle handling was proposed by applying right angle to the steering wheel,based on the nonlinear adaptive optimal control(NAOC).A nonlinear 4-DOF model was initially developed,then it was simplified to a 2-DOF model with reasonable assumptions to design observer and optimal controllers.Then a simplified model was developed for steering system.The numerical simulations were carried out using vehicle parameters for standard maneuvers in dry and wet road conditions.Moreover,the hardware in the loop method was implemented to prove the controller ability in realistic conditions.Simulation results obviously show the effectiveness of NAOC on vehicle handling and reveal that the proposed controller can significantly improve vehicle handling during severe maneuvers.Vahid Tavoosi Reza Kazemi Atta Oveisi 2014Journal of Central South University2014,21,1:8
6Post-transplant lymphoproliferative disorder after liver transplantation: Incidence, long-term survival and impact of serum tacrolimus level显示文摘AIM To investigate incidence and survival of post-transplant lymphoproliferative disorder(PTLD) patients after liver transplantation. METHODS A cross-sectional survey was conducted among patients who underwent liver transplantation at Shiraz Transplant Center(Shiraz, Iran) between August 2004 and March 2015. Clinical and laboratory data of patients were collected using a data gathering form. RESULTS There were 40 cases of PTLD in the pediatric age group and 13 cases in the adult group. The incidence of PTLD was 6.25% in pediatric patients and 1.18% in adult liver transplant recipients. The post-PTLD survival of patients at 6 mo was 75.1% ± 6%, at 1 year was 68.9% ± 6.5% and at 5 years was 39.2% ± 14.2%. Higher serum tacrolimus level was associated with lower post-PTLD survival in pediatric patients(OR = 1.07, 95%CI: 1.006-1.15, P = 0.032). A serum tacrolimus level over 11.1 ng/mL was predictive of post PTLD survival(sensitivity = 90%, specificity = 52%, area under the curve = 0.738, P = 0.035). CONCLUSION Incidence of PTLD in our liver transplant patients is comparable to other centers. 7m TOR signaling in liver regeneration: Rapamycin combined with growth factor treatment显示文摘AIM: To investigate the effects of mammalian target of rapamycin(mT OR) inhibition on liver regeneration and autophagy in a surgical resection model.METHODS: C57BL/6 mice were subjected to a 70% partial hepatectomy(PH) and treated intraperitoneally every 24 h with a combination of the m TOR inhibitor rapamycin(2.5 mg/kg per day) and the steroid dexamethasone(2.0 mg/kg per day) in phosphate bufferedsaline(PBS) or with PBS alone as vehicle control. In the immunosuppressant group, part of the group was treated subcutaneously 4 h prior to and 24 h after PH with a combination of human recombinant interleukin 6(IL-6; 500 μg/kg per day) and hepatocyte growth factor(HGF; 100 μg/kg per day) in PBS. Animals were sacrificed 2, 3 or 5 d after PH and liver tissue and blood were collected for further analysis. Immunohistochemical staining for 5-Bromo-2'-deoxyuridine(Brd U) was used to quantify hepatocyte proliferation. Western blotting was used to detect hepatic microtubule-associated protein 1 light chain 3(LC3)-Ⅱ protein expression as a marker for autophagy. Hepatic gene expression levels of proliferation-, inflammation- and angiogenesisrelated genes were examined by real-time reverse transcription-polymerase chain reaction and serum bilirubin and transaminase levels were analyzed at the clinical chemical core facility of the Erasmus MC-University Medical Center.RESULTS: m TOR inhibition significantly suppressed regeneration, shown by decreased hepatocyte proliferation(2% vs 12% Brd U positive hepatocyte nuclei at day 2, P < 0.01; 0.8% vs 1.4% at day 5, P = 0.02) and liver weight reconstitution(63% vs 76% of initial total liver weight at day 3, P = 0.04), and furthermore increased serum transaminase levels(aspartate aminotransferase 641 U/L vs 185 U/L at day 2, P = 0.02). Expression of the autophagy marker LC3-Ⅱ, which was reduced during normal liver regeneration, increased after mT OR inhibition(46% increase at day 2, P = 0.04). Hepatic gene expression showed an increased inflammation-related response [tumor necrosis factor(TNF)-α 3.2-fold upregulation at day 2, P = 0.03; IL-1Ra 6.0-fold upregulation at day 2 and 42.3-fold upregulation at day 5, P < 0.01] and a reduced expression of cell cycle progression and angiogenesis-related factors(HGF 40% reduction at day 2; vascular endothelial growth factor receptor 2 50% reduction at days 2 and 5; angiopoietin 1 60% reduction at day 2, all P ≤ 0.01). Treatmentwith the regeneration stimulating cytokine IL-6 and growth factor HGF could overcome the inhibitory effect on liver weight(75% of initial total liver weight at day 3, P = 0.02 vs immunosuppression alone and P = 0.90 vs controls) and partially reversed gene expression changes caused by rapamycin(TNF-α and IL-1Ra levels at day 2 were restored to control levels). However, no significant changes in hepatocyte proliferation, serum injury markers or autophagy were found.CONCLUSION: mT OR inhibition severely impairs liver regeneration and increases autophagy after PH. 9Performance of low-salinity water flooding for enhanced oil recovery improved by SiO_2 nanoparticles显示文摘Low-salinity water injection has been utilized as a promising method for oil recovery in recent years. Low-salinity water flooding changes the ion composition or brine salinity for improving oil recovery. Recently, the application of nanoparticles with low-salinity water flooding has shown remarkable results in enhanced oil recovery(EOR). Many studies have been performed on the effect of nanofluids on EOR mechanisms. Their results showed that nanofluids can improve oil recovery when used in low-salinity water flooding. In this work, the effects of injection of low-salinity water and low-salinity nanofluid(prepared by adding SiO_2 nanoparticles to low-salinity water) on oil recovery were investigated. At first, the effects of ions were investigated with equal concentrations in low-salinity water flooding. The experimental results showed that the monovalent ions had better performance than the divalent ions because of them having more negative zeta potential and less ionic strength. Also, low-salinity water flooding recovered 6.1% original oil in place(OOIP) more than the high-salinity flooding. Contact angle measurements demonstrated that low-salinity water could reduce the contact angle between oil and water. Then in the second stage, experiments were continued by adding SiO_2 nanoparticles to the K+ solution which had the highest oil recovery at the first stage. 10Development of fragility curves by incorporating new spectral shape indicators and a weighted damage index:case study of steel braced frames in the city of Mashhad,Iran显示文摘In this study, strong ground motion record(SGMR) selection based on Eta(η) as a spectral shape indicator has been investigated as applied to steel braced frame structures. A probabilistic seismic hazard disaggregation analysis for the definition of the target Epsilon(ε) and the target Eta(η) values at different hazard levels is presented, taking into account appropriately selected SGMR's. Fragility curves are developed for different limit states corresponding to three representative models of typical steel braced frames having significant irregularities in plan, by means of a weighted damage index. 11Transcriptional down regulation of hTERT and senescence induction in HepG2 cells by chelidonine显示文摘AIM: To investigate the potential effects of chelidonine, the main alkaloid of Chelidonium majus, on telomerase activity and its regulation in HepG2 cells. METHODS: Cytotoxicity of chelidonine for HepG2 cells was determined by neutral red assay. A modified polymerase chain reaction (PCR)-based telomerase repeat amplification protocol was used to estimate relative telomerase activity in chelidonine-treated cells in comparison with the untreated control cells. Relative expression level of the catalytic subunit of telomerase (hTERT) gene and P-glycoprotein (pgp) were estimated using semi-quantitative real-time reverse transcription-PCR (RT-PCR). Cell senescence in treated cells was demonstrated using a β-galactosidase test. RESULTS: Cytotoxicity of chelidonine in HepG2 cells was not dose-dependent and tended to reach plateau immediately after the living cells were reduced in number to slightly higher than 50%. However, 12 μmol/L concentration of chelidonine was considered as LD50, where the maximal attainable effects were realized. Real-time RT-PCR data showed that the expression of pgp increased three-fold in chelidonine treated HepG2 cells in comparison with the untreated controls. Morphologically, treated HepG2 cells showed apoptotic features after 24 h and a small fraction of cells appeared with single blister cell death. The relative expressionlevel of Bcl-2 dropped to less than 50% of control cells at a sub-apoptotic concentration of chelidonine and subsequently increased to higher than 120% at LD50. Telomerase activity was reduced considerably after administration of very low doses of chelidonine, whereas higher concentrations of chelidonine did not remarkably enhance the effect. Real-time RT-PCR experiments indicated a drastic decrease in expression level of hTERT subunit of telomerase under treatment with chelidonine. Repeated treatment of cells with very low doses of chelidonine caused a decline in growth rate by 4 wk and many of the cells appeared to be aged with large volume and dark staining in the β-galactosidase assay. CONCLUSION: Chelidonine reduces telomerase activity through down-regulation of hTERT expression. 12Process mineralogy as a key factor affecting the flotation kinetics of copper sulfide minerals显示文摘The aim of this study is to apply process mineralogy as a practical tool for further understanding and predicting the flotation kinetics of the copper sulfide minerals. The minerals' composition and association, grain distribution, and liberation within the ore samples were analyzed in the feed, concentrate, and the tailings of the flotation processes with two pulp densities of 25 wt% and 30 wt%. The major copper-bearing minerals identified by microscopic analysis of the concentrate samples included chalcopyrite(56.2 wt%), chalcocite(29.1 wt%),covellite(6.4 wt%), and bornite(4.7 wt%). Pyrite was the main sulfide gangue mineral(3.6 wt%) in the concentrates. A 95% degree of liberation with d_(80) > 80 μm was obtained for chalcopyrite as the main copper mineral in the ore sample. The recovery rate and the grade in the concentrates were enhanced with increasing chalcopyrite particle size. Chalcopyrite particles with a d_(80) of approximately 100 μm were recovered at the early stages of the flotation process. The kinetic studies showed that the kinetic second-order rectangular distribution model perfectly fit the flotation test data. 13FOLFIRINOX and translational studies: Towards personalized therapy in pancreatic cancer显示文摘Pancreatic cancer is an extremely aggressive disease; although progress has been made in the last few years, the prognosis of these patients remains dismal. FOLFIRINOX is now considered a standard treatment in first-line setting, since it demonstrated an improved overall and progression-free survival vs gemcitabine alone. However, the enthusiasm over the benefit of this three-drug regimen is tempered by the associated increased toxicity profile, and many efforts have been made to improve the feasibility of this schedule. After a more recent phase Ⅲ trial showing an improved outcome over gemcitabine, the combination of gemcitabine/nab-paclitaxel emerged as another standard first-line treatment. However, this treatment is also associated with more side effects. In addition, despite initial promising data on the predictive role of SPARClevels, recent studies showed that these levels are not associated with nab-paclitaxel efficacy. The choice to use this treatment over FOLFIRINOX is therefore a topic of debate, also because no validated biomarkers to guide FOLFIRINOX treatment are available. In the era of actionable mutations and target agents it would be desirable to identify molecular factors or biomarkers to predict response to therapy in order to maximize the efficacy of treatment and avoid useless toxic effects for non-responding patients. However, until today the milestone of treatment for pancreatic cancer remains chemotherapy combinations, without predictive or monitoring tools existing to optimize therapy. 15Structural characterization and strengthening mechanism of forsterite nanostructured scaffolds synthesized by multistep sintering method显示文摘In this study, highly porous forsterite scaffolds with interconnected porosities were synthesized using multi-step sintering(MSS) method. The starting powder was nanosized forsterite, which was synthesized from talc and magnesium carbonate powders. The phase composition, average particle size and morphology of the produced forsterite powder were characterized by X-ray diffraction technique(XRD) and transition electron microscopy(TEM). Forsterite scaffolds were produced by foamy method using polymeric sponges. MSS process including three steps was used to efficiently sinter the forsterite nanopowders without destroying the initial porous structure of polymeric sponges. The results showed that MSS technique is an efficient and appropriate procedure to produce highly porous forsterite scaffolds with pore size in the range of 100-300 μm. The compressive strength, compressive modulus and porosity of C12 specimen(sintered at 1650℃ for 1 h with subsequent annealing at 1000℃ for 1000 min) was 1.88 MPa, 29.2 MPa, and 72.4%, respectively, which is very close to that of cancellous bone. 17Effects of fixed and dynamic mesh methods on simulation of stepped planing craft显示文摘In the present study,we investigate the effects of fixed and dynamic mesh methods on CFD simulation of stepped planing craft.To this accomplishment,three different body forms of without step,one step,and two step Cougar planing crafts are considered.Three-dimensional CFD analysis is conducted using finite volume method(FVM).Volume of fluid(VOF)model is used for free surface modeling.Overset mesh technique(dynamic mesh)and fixed mesh method are conducted by STAR CCM and ANSYS CFX CFD toolboxes,respectively.CFD results of total hydrodynamic resistance and dynamic trim angle are compared against our measured experimental data to assess the performance and accuracy of considered mesh methods.For more details,pressure distributions,wave patterns and streamlines around the hull models are also presented.Based on our CFD results,dynamic mesh method is more accurate and precise to simulate the stepped planing crafts compared to fixed mesh method.However,computational cost for dynamic mesh method is significantly greater than the fixed mesh method.We also found that the porpoising phenomenon is only detectable using dynamic mesh method in simulation of stepped Cougar planing craft.©2018 Shanghai Jiaotong University.Published by Elsevier B.V.M.Mehdi Doustdar Hamid Kazemi 2019Journal of Ocean Engineering and Science2019,4,1:3
18Dynamic recrystallization and precipitation in high manganese austenitic stainless steel during hot compression显示文摘Dynamic recrystallization and precipitation in a high manganese austenitic stainless steel were investigated by hot compression tests over temperatures of 950-1150°C at strain rates of 0.001 s-1-1 s-1.All the flow curves within the studied deformation regimes were typical of dynamic recrystallization.A window was constructed to determine the value of apparent activation energy as a function of strain rate and deformation temperature.The kinetics of dynamic recrystallization was analyzed using the Avrami kinetics equation.A range of apparent activation energy for hot deformation from 303 kJ/mol to 477 kJ/mol is obtained at different deformation regimes.Microscopic characterization confirms that under a certain deformation condition(medium Zener-Hollomon parameter(Z) values),dynamic recrystallization appears at first,but large particles can not inhibit the recrystallization.At low or high Z values,dynamic recrystallization may occur before dynamic precipitation and proceeds faster.In both cases,secondary phase precipitation is observed along prior austenite grain boundaries.Stress relaxation tests at the same deformation temperatures also confirm the possibility of dynamic precipitation.Unexpectedly,the Avrami's exponent value increases with the increase of Z value.It is associated with the priority of dynamic recrystallization to dynamic precipitation at higher Z values.Amir Momeni Shahab Kazemi Golam Ebrahimi Alireza Maldar 2014International Journal of Minerals,Metallurgy and Materials2014,21,1:3
19Evaluation of rabbit antibody response against 8 and 16 kDa recombinant subunits of antigen B from Echinococcus granulosus显示文摘Objective:To immunize rabbits with 12 and 16 kDa recombinant subunits of antigen B from Echinococcus granulosus(E.granulosus) and measuring polyclonal antibody and humoral immune response using ELISA and gel diffusion.Methods:Two mentioned antigens were cloned and expressed in expression vector and purified by affinity chromatography.Four young rabbits were selected and challenged intradermallt with yielded recombinant antigens.Rabbits’ sera were collected post infection and were tested using ELISA and gel diffusion for polyclonal antibody detection 10 days after last injection.Results:The specific antibody against the recombinant peptides was efficiently produced within 4 weeks post infection.Conclusions: Produced recombinants proteins could induce the immune response of the rabbits successfully. 20Responses of Four Rice Varieties to Elevated CO_2 and Different Salinity Levels显示文摘This study was carried out in 2014 at Isfahan University of Technology, Iran, to evaluate the responses of four rice varieties(Neda, Deylamani, Shiroudi and Domsorkh) to ambient(360 ± 50 μmol/mol) and elevated(700 ± 50 μmol/mol) air carbon dioxide(CO_2) concentrations under four salinity levels(0, 30, 60 and 90 mmol/L Na Cl). There was significant variation among rice varieties in response to elevated CO_2 concentration under the four salinity levels. Under non-saline condition, elevated CO_2 increased the dry weight of Neda, Deylamani and Domsorkh by 8%, 50% and 8%, respectively, but reversely decreased that of Shiroudi by 34%. Increasing CO_2 concentration significantly reduced the negative effects of salinity on Shiroudi, but these effects were even increased in Deylamani and Domsorkh under all the salinity levels and in Neda only under 30 and 60 mmol/L Na Cl. Significant correlations were established between plant dry weight, SPAD value and leaf area under both CO_2 levels. However, this trend was observed only at ambient CO_2 concentration in the presence of soluble carbohydrates.
