1Non-viral and viral delivery systems for CRISPR-Cas9 technology in the biomedical field显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)-associated protein 9(CRISPR-Cas9) system provides a novel genome editing technology that can precisely target a genomic site to disrupt or repair a specific gene. Some CRISPR-Cas9 systems from different bacteria or artificial variants have been discovered or constructed by biologists, and Cas9 nucleases and single guide RNAs(sgRNA) are the major components of the CRISPR-Cas9 system. These Cas9 systems have been extensively applied for identifying therapeutic targets, identifying gene functions, generating animal models, and developing gene therapies.Moreover, CRISPR-Cas9 systems have been used to partially or completely alleviate disease symptoms by mutating or correcting related genes. However, the efficient transfer of CRISPR-Cas9 system into cells and target organs remains a challenge that affects the robust and precise genome editing activity. The current review focuses on delivery systems for Cas9 mRNA, Cas9 protein, or vectors encoding the Cas9 gene and corresponding sgRNA. Non-viral delivery of Cas9 appears to help Cas9 maintain its on-target effect and reduce off-target effects, and viral vectors for sgRNA and donor template can improve the efficacy of genome editing and homology-directed repair. 3CRISPR/Cas9-mediated correction of human genetic disease显示文摘The clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated(Cas) protein 9 system(CRISPR/Cas9) provides a powerful tool for targeted genetic editing. Directed by programmable sequence-specific RNAs,this system introduces cleavage and double-stranded breaks at target sites precisely. Compared to previously developed targeted nucleases, the CRISPR/Cas9 system demonstrates several promising advantages, including simplicity, high specificity,and efficiency. Several broad genome-editing studies with the CRISPR/Cas9 system in different species in vivo and ex vivo have indicated its strong potential, raising hopes for therapeutic genome editing in clinical settings. Taking advantage of non-homologous end-joining(NHEJ) and homology directed repair(HDR)-mediated DNA repair, several studies have recently reported the use of CRISPR/Cas9 to successfully correct disease-causing alleles ranging from single base mutations to large insertions. 4Oxidized mitochondrial DNA sensing by STING signaling promotes the antitumor effect of an irradiated immunogenic cancer cell vaccine显示文摘Exposure to ionizing radiation,a physical treatment that inactivates live tumor cells,has been extensively applied to enhance the antitumor responses induced by cancer cell vaccines in both animal research and human clinical trials.However,the mechanisms by which irradiated cells function as immunogenic tumor vaccines and induce effective antitumor responses have not been fully explored.Here,we demonstrate that oxidized mitochondrial DNA(mtDNA)and stimulator of interferon genes(STING)signaling play a key roles in the enhanced antitumor effect achieved with an irradiated tumor cell vaccine.Elevations in ROS and oxidized mtDNA 8-OHG content could be induced in irradiated tumor cells.Oxidized mtDNA derived from irradiated tumor cells gained access to the cytosol of dendritic cells(DCs).Oxidized mtDNA,as a DAMP or adjuvant,activated the STING-TBK1-IRF3-IFN-β pathway in DCs,which subsequently cross-presented irradiated tumor cell-derived antigens to CD8^(+)T cells and elicited antitumor immunity.The results of our study provide insight into the mechanism by which an irradiated cell vaccine mediates antitumor immunity,which may have implications for new strategies to improve the efficacy of irradiated vaccines.Chunju Fang Fei Mo Li Liu Jing Du Min Luo Ke Men Feifei Na Wei Wang Hanshuo Yang Xiawei Wei 2021Cellular & Molecular Immunology2021,18,9:2

6Sustainability of SARS-CoV-2 Induced Humoral Immune Responses in COVID-19 Patients from Hospitalization to Convalescence Over Six Months显示文摘Understanding the persistence of antibody in convalescent COVID-19 patients may help to answer the current major concerns such as the risk of reinfection,the protection period of vaccination and the possibility of building an active herd immunity.This retrospective cohort study included 172 COVID-19 patients who were hospitalized in Wuhan.A total of404 serum samples were obtained over six months from hospitalization to convalescence.Antibodies in the specimens were quantitatively analyzed by the capture chemiluminescence immunoassays(CLIA).All patients were positive for the anti-SARS-Co V-2 Ig M/Ig G at the onset of COVID-19 symptoms,and the Ig G antibody persisted in all the patients during the convalescence.However,only approximately 25%of patients can detect the Ig M antibodies,Ig M against N protein(NIg M)and receptor binding domain of S protein(RBD-Ig M)at the 27 th week.The titers of Ig M,N-Ig M and RBD-Ig M reduced to 16.7%,17.6%and 15.2%of their peak values respectively.In contrast,the titers of Ig G,N-Ig G and RBD-Ig G peaked at 4–5 th week and reduced to 85.9%,62.6%and 87.2%of their peak values respectively at the end of observation.Dynamic behavior of antibodies and their correlation in age,gender and severity groups were investigated.In general,the COVID-19 antibody was sustained at high levels for over six months in most of the convalescent patients.Only a few patients with antibody reducing to an undetectable level which needs further attention.The humoral immune response against SARS-Co V-2 infection in COVID-19 patients exhibits a typical dynamic of acquired immunity.Yang Zheng Qing Zhang Ashaq Ali Ke Li Nan Shao Xiaoli Zhou Zhiqin Ye Xiaomin Chen Shanshan Cao Jing Cui Juan Zhou Dianbing Wang Baidong Hou Min Li Mengmeng Cui Lihua Deng Xinyi Sun Qian Zhang Qinfang Yang Yong li Hui Wang Yake Lei Bo Yu Yegang Cheng Xiaolin Tong Dong Men Xian-En Zhang 2021Virologica Sinica2021,36,5:1
7Effects of Nano-carbon Humic Acid Water-retaining Fertilizer on Citrus Growth and the Soil Bacterial Community in Citrus Field显示文摘[Objective] In order to reveal the effects of reducing the amount of novel nano-carbon humic acid water-retaining fertilizer(CSF) on soil microbial community structure and citrus growth. [Method]In this study,conventional fertilization was as the control(KC1) in Wanzhou citrus orchard of Three Gorges Reservoir area. CSF reductions by 0%(KC2),10%(KC3),20%(KC4),30%(KC5) and 40%(KC6) were used to analyze the changes of soil bacterial community structure,citrus yield and quality. [Result]The results showed that the observed species,Shannon index,Chao1 index and PD_whole_tree of KC6 were higher than those of KC1,and were the same as KC2. The abundance of Xanthomonadaceae was the highest in KC5. Compared with KC1,the Xanthomonadaceae in KC3,KC4 and KC6 was significantly decreased,and the levels of Nitrosomonadaceae and Pseudomonasaceae were higher than that of KC1 after the treatment of KC6. Sphingomonas in different reduction treatments was lower than that of KC1,but Burkholderia and Pseudomonas were significantly higher than those of KC1. It was found that the similarity among treatments was small after bacterial community similarity clustering analysis,and citrus yield increased somewhat after CSF fertilization reduction.When CSF fertilization reduced by 30%,citrus yield increased by 4. 50%. When CSF fertilization reduced by 40%,citrus yield decreased by4. 14%. After CSF fertilization,citrus quality did not change significantly in CSF conventional fertilization and reduction of 10% and 40%,while significantly decreased in 20% and 30% of fertilization reduction. 10Factors affecting rate of stroke-related death in elderly male military population A 23-year cohort study of 1268 cases in Xi'an,China显示文摘The study is the first long-term cohort study examining stroke and its subtypes among a population of Chinese elderly male retired military veterans.We reported on a 23-year cohort study examining stroke in 1 268 elderly male patients living in Xi'an,China since 1987.The stroke-related mortality rate in this cohort was 361.50/1 × 106 per year.Cerebral hemorrhage was the dominant cause of death,with 28 cases of fatal cerebral infarction and 49 cases of fatal cerebral hemorrhage among 77 stroke-related deaths.Independent risk factors for stroke mortality included age,blood pressure,smoking,body mass index,family history of hypertension,past medical history of stroke,hypertension and hyperlipidemia.Among them,ischemic stroke mortality correlated with age,smoking,family history of hypertension and past medical history of stroke,while hemorrhagic stroke was related to blood pressure,body mass index and past medical history of hypertension.Our results indicated that maintaining appropriate levels of blood pressure and body mass,smoking cessation and prevention of hyperlipidemia can reduce the risk of stroke-related death in elderly males who are retired from military service.Yanhua Li Yu Sun Xiaoyong Sai Yao He Qiang Wu Yongping Yan Ke Men Liangshou Li 2011Neural Regeneration Research2011,6,13:0
11Small molecules in the treatment of COVID-19显示文摘The outbreak of COVID-19 has become a global crisis,and brought severe disruptions to societies and economies.Until now,effective therapeutics against COVID-19 are in high demand.Along with our improved understanding of the structure,function,and pathogenic process of SARS-CoV-2,many small molecules with potential anti-COVID-19 effects have been developed.So far,several antiviral strategies were explored.Besides directly inhibition of viral proteins such as RdRp and M^(pro),interference of host enzymes including ACE2 and proteases,and blocking relevant immunoregulatory pathways represented by JAK/STAT,BTK,NF-κB,and NLRP3 pathways,are regarded feasible in drug development.The development of small molecules to treat COVID-19 has been achieved by several strategies,including computer-aided lead compound design and screening,natural product discovery,drug repurposing,and combination therapy.Several small molecules representative by remdesivir and paxlovid have been proved or authorized emergency use in many countries.And many candidates have entered clinical-trial stage.Nevertheless,due to the epidemiological features and variability issues of SARS-CoV-2,it is necessary to continue exploring novel strategies against COVID-19.This review discusses the current findings in the development of small molecules for COVID-19 treatment.Moreover,their detailed mechanism of action,chemical structures,and preclinical and clinical efficacies are discussed.Sibei Lei Xiaohua Chen Jieping Wu Xingmei Duan Ke Men 2023Signal Transduction and Targeted Therapy2023,8,1:0
12Phenanthrene sorption to environmental black carbon in sediments from the Song-Liao watershed(China)显示文摘Black carbon(BC)in ten contaminated sediments from the Song-Liao watershed,NE China,was isolated upon treatments using a combustion method at 375℃,and the isolates’sorption isotherms for phenanthrene(Phen)were determined.All sorption isotherms were nonlinear and fitted well by the Freundlich model.A negative relation was found between Freundlich sorption nonlinearity parameter(n values)and BC/total organic carbon(TOC)content of the original sediments(r^(2)=0.687,p<0.01),indicating the dominance of BC in Phen sorption nonlinearity.The BC isolates from this industrialized region had n values of 0.342 to 0.505 and logKFOC values of 6.02 to 6.42(μg·kg^(–1)·OC^(–1))/(μg·L^(–1))n for Phen.At a given Ce,the BC had higher Koc value than the original sediments,revealing a higher sorption capacity for BC.BC was responsible for 50.0%to 87.3%of the total sorption at Ce=0.05 Sw,clearly indicating the dominance of BC particles in overall sorption of Phen by sediments.Jinghuan ZHANG Mengchang HE Chunye LIN Ke SUN Bin MEN John L.ZHOU 2009Frontiers of Environmental Science & Engineering2009,3,4:0
13Self-powered stimuli responsive material for dual stimulation of heat and guest molecules显示文摘Stimuli-responsive smart materials exhibit reverse chemical/physical changes in response to external stimuli and research on stimuli-responsive smart materials with self-powered properties is still uncultivated ground.Here,we report perovskite crystalline self-powered multiple stimuli-responsive materials triggered by chemical and thermal stimuli.[HMEP]PbI_(3)·(H_(2)O)(1;HMEP is a hydroxytris(1-methylethyl)phosphorus cation)crystallizes in a chiral space group P 21 at 293 K and has the piezoelectric reaction(d_(33)=10 pC/N and output voltage=1 V)of self-powered modes,this value is larger than the value of 3 pC/N for the classical piezoelectric material ZnO.Piezoelectric materials can generate energy due to mechanical deformation,and using thermal heating to lose water,[HMEP]PbI_(3)(2)can be obtained.2 crystallizes in the non-centrosymmetric space group,undergoes two reversible phase transitions at 243/255 K and 315/348 K,and shows second harmonic generation switching.Interestingly,2 can return to the hydrated form 1 after absorbing water.This work will lay the foundation for self-powered stimuli-responsive compounds and contribute to the construction of novel organic-inorganic hybrid materials with second harmonic generation switching.Jintao Men Ke Xu Taiting Sha Lei He Yan Xu Jie Mu Tijian Yin Qiong Ye 2024Chinese Chemical Letters2024,35,2:0

15Pollutant emissions from biomass burning:A review on emission characteristics,environmental impacts,and research perspectives显示文摘Biomass is one most abundant resource on the earth providing important energies in support of so-cioeconomic development in many areas.Burning of biomass fuels comprises to nearly 10%of the total energy from anthropogenic combustion processes:however,as the burning is usually incomplete,this process yields products of incomplete combustion posing consequently significant impacts on air quality,human health,and climate change.Here,we analyzed spatiotemporal characteristics in intentional and unintentional biomass burning from different sectors,discussed impacts of biomass burning emissions on indoor and outdoor air quality,and consequent influences on human health.The global total con-sumption amount of biomass including both natural and anthropogenic sources was approximately 7900 Tg in 2019,with significantly large regional and sectorial discrepancies among regions.Globally,anthropogenic biomass burning amounts increased gradually,but notably in some developing countries like China residential consumption of biomass fuels,as one large sector of biomass use,decreased over time.Uncommercial biomass consumption needs to be accurately quantified.There are relatively rich datasets of pollutant emission factors from biomass burning,including laboratory and field tests,but still large variations exit and contribute substantially to the uncertainty in emission inventory.Global pri-mary PM2.5,black carbon and organic carbon emissions from biomass burning were about 51,4.6,and 29 Tg,respectively,contributing to nearly 70%,55%,and 90%of the total emission from all sources,and emissions from the residential sector and open fires are major sources.Brown carbon emissions from biomass burning attracts growing interests but available studies adopted different methodologies challenging the comparability of those results.Biomass burning emissions polluted not only ambient air but more severely indoor air quality,adversely affecting human health.Future studies that should be emphasized and promoted are suggested.Ke Jiang Ran Xing Zhihan Luo Wenxuan Huang Fan Yi Yatai Men Nan Zhao Zhaofeng Chang jinfeng Zhao Bo Pan Guofeng shen 2024Particuology2024,85,2:0
16Research on error accumulation control of three-dimensional adjustment with offset constraint显示文摘Background Currently,laser tracker is the primary instrument used to carry out three-dimensional position measurement in accelerator alignment.Theoretically,three-dimensional measuring data processed by three-dimensional adjustment are more rigorous,however,error accumulation is found in practice.Purpose In order to control error accumulation and further improve the measurement accuracy of accelerator alignment,this research introduces the laser alignment system into the activity of measurement and data processing.Methods A measurement scheme combining laser tracker and laser alignment system is proposed.To construct the constraint condition,the offset values from the measuring points to the laser straight-line datum were used.To carry out the three-dimensional adjustment with offset constraint,the laser tracker observations were used.Results A three-dimensional adjustment function model of laser tracker observations is given.The construction method of the constraint equation is researched,and the calculation formulas of the three-dimensional adjustment with offset constraint are derived.A 200 m linac tunnel control network is designed,using simulation measurement method,the measuring data of laser tracker and the offset values from the measuring points to the laser straight-line datum were generated.The simulated data are calculated by the method given in this paper and the result is analyzed.Conclusion Simulation result shows introducing the laser alignment system into laser tracker measurement and applying the three-dimensional adjustment with offset constraint can effectively suppress the error accumulation caused by long distance move station measurement.Wang Xiaolong Kang Ling Dong Lan Li Bo Men Lingling Luo Tao Wang Tong Liang Jing He Zhenqiang Ke Zhiyong Ma Na Lu Shang Han Yuanying Yan Luping Zhang Luyan Liu Xiaoyang Yan Haoyue Li Chunhua Wu Lei 2022Radiation Detection Technology and Methods2022,6,4:0
