| 1 | Photosynthesis and growth responses of pea Pisum sativum L. under heavy metals stress显示文摘The present work aimed to study the physiological effects of cadmium (Cd) and copper (Cu) in pea (Pisum sativum). Pea plants were exposed to increasing doses of cadmium chloride (CdCl2) and copper chloride (CuCl2) for 20 d. The examined parameters, namely root and shoot lengths, the concentration of photosynthetic pigments and the rate of photosynthesis were affected by the treatments especially with high metals concentrations. The analysis of heavy metals accumulation shows that leaves significantly accumulate cadmium for all the tested concentrations. However, copper was significantly accumulated only with the highest tested dose. This may explain the higher inhibitory effects of cadmium on photosynthesis and growth in pea plants. | 2 | 蓝舌病毒VP5蛋白抗体的制备及鉴定显示文摘本研究参考GenBank中蓝舌病毒16型(BTV-16)标准株RSAvvvv的序列,首先人工合成编码BTV-16 VP5蛋白的S6全长基因,并克隆于pUC-57载体。设计表达引物,利用PCR方法扩增N末端缺少编码第1~41位氨基酸的截短基因VP5Δ41aa,将其亚克隆到表达载体pET-32a(+)中,构建重组表达质粒pET-VP5Δ41aa。将表达质粒转化大肠杆菌BL21(DE3)后,利用IPTG进行诱导表达,SDS-PAGE结果显示,重组蛋白VP5Δ41aa在大肠杆菌中大量表达。利用组氨酸标签纯化树脂获得了高纯度的VP5Δ41aa蛋白,用其3次免疫新西兰大白兔制备相应的抗血清,Western-blot检测显示,抗血清不仅可与重组表达的VP5Δ41aa蛋白反应,而且可与BTV感染细胞中的VP5蛋白反应。免疫荧光试验表明,该抗体也可与BTV感染C6/36细胞中具有天然构象的VP5蛋白反应。上述结果表明,本试验制备的VP5抗体将为深入研究VP5蛋白的生物学功能奠定基础。 | 康棣 独军政 高闪电 田占成 Darien Kheder ALI MOHAMED 郭艳妮 刘光远 罗建勋 殷宏 | 2018 | 中国兽医科学2018，48，6： | 3 |
| 3 | 蓝舌病毒NS2蛋白的抗体制备及鉴定显示文摘本研究参考GenBank中蓝舌病毒16型(Bluetongue virus serotype 16,BTV-16)标准株RSAvvvv的序列,人工合成编码BTV-16 NS2蛋白的S8基因,将其亚克隆至原核表达载体pPRoEx-HTb中,构建重组表达质粒pPro-NS2,并转化大肠杆菌BL21进行IPTG诱导表达,并进行可溶性分析。SDS-PAGE结果显示,表达的rNS2蛋白主要以包涵体的形式存在于菌体中。利用组氨酸标签纯化树脂获得了高纯度的NS2蛋白,免疫新西兰白兔制备抗NS2蛋白的多克隆抗体。Western blot结果显示,制备的抗NS2蛋白多克隆抗体不仅可与重组表达的rNS2蛋白反应,而且可与BTV感染细胞后的天然NS2蛋白反应。间接免疫荧光结果显示,该抗体也可与BTV感染C6/36细胞中的天然NS2蛋白反应,且发现NS2大量分布于C6/36细胞的胞质中。本研究所制备的NS2蛋白的多克隆抗体具有很好的反应性和特异性,为进一步研究NS2蛋白的功能奠定了基础。 | 张国芮 独军政 高闪电 田占成 Darien Kheder Ali Mohamed 康棣 郭艳妮 薛慧文 殷宏 | 2019 | 中国动物传染病学报2019，27，2： | 3 |
