| 2 | 人肝癌细胞系P2-HCC的建立及体外诱导分化特性的初步研究显示文摘目的:从人肝细胞癌中分离肝癌细胞并对其体外诱导分化特性进行分析,试图得到有关导致肝癌发生的“癌干细胞”的相关资料。方法:首先将人原发性肝癌组织小块在裸鼠皮下过继接种,然后将生成的肿瘤进行原代培养并得到单层生长的肿瘤细胞系,利用体外诱导实验对其形态及分子表型的变化情况进行观察和分析。结果:所分离获得的细胞系具有肝肿瘤细胞的特征,但也表达某些干细胞的分子标志如c met。体外诱导分化实验提示胰岛素/氢化可的松、二甲亚砜可能具有诱导该细胞向成熟肝细胞方向分化的作用,诱导后的细胞又重新表达葡萄糖6 磷酸酶,白蛋白表达明显升高。结论:人P2 HCC细胞具有强大的增殖能力,体外实验提示其具有一定的向成熟肝细胞分化的潜能。这些细胞的发生与肝癌干细胞及肝细胞的关系尚待进一步研究。 | 朱海英 张朵 谢东甫 王新民 周旭宇 Joseph T Lau 胡以平 | 2005 | 第二军医大学学报2005，26，3： | 16 |

| 4 | 人胎肝干细胞在重症联合免疫缺陷小鼠损伤肝脏中的植入显示文摘目的:从孕4.5~6周的人胚胎肝脏中分离培养肝干细胞,并观察其在重症联合免疫缺陷(SCID)小鼠损伤肝脏中的植入.方法:从孕4.5~6周的人胚胎肝脏中分离干细胞并进行体外长期培养,用RT PCR方法检测肝干细胞相关分子标志以及八聚物结合蛋白4(Oct-4)的表达,然后植入经四氯化碳(CCl4)造成急性肝损伤的SCID小鼠体内,在移植后2、l0、17d分别对受体鼠肝脏冰冻切片进行荧光观察和H-E染色.结果:所建立的胎肝细胞系不仅表达二肽水解酶Ⅳ(DPPⅣ)、白蛋白(Alb)、细胞角蛋白(CK)19、γ-谷氨酰转肽酶(γ-GT)、CK18、肝细胞生长因子受体(c-met)、干细胞因子受体(c-kit)等肝干细胞相关标志,还表达干细胞转录因子Oct-4.在细胞移植后2 d,肝脏冰冻切片未见绿色荧光;移植后10 d和17d肝脏冰冻切片见散在的绿色荧光.对同一切片做H-E染色,发现绿色荧光部位为肝板内的多个成熟肝细胞.结论:从人胚胎肝脏中分离的干细胞能够植入SCID小鼠的损伤肝脏. | 苏娟 李文林 陶欣荣 朱海英 王新民 李建秀 田棣 余宏宇 Joseph T Lau 胡以平 | 2005 | 第二军医大学学报2005，26，3： | 4 |
