1Distribution of Prestin on Outer Hair Cell Basolateral Surface显示文摘Prestin has been identified as a motor protein responsible for outer hair cell (OHC) electromotility and is expressed on the OHC surface. Previous studies revealed that OHC electromotility and its associated nonlinear capacitance were mainly located at the OHC lateral wall and absent at the apical cuticular plate and the basal nucleus region. Immunofluorescent staining for prestin also failed to demonstrate prestin expression at the OHC basal ends in whole-mount preparation of the organ of Corti. However, there lacks a definitive demonstration of the pattern of prestin distribution. The OHC lateral wall has a trilaminate organization and is composed of the plasma membrane, cortical lattice, and subsurface cisternae. In this study, the location of prestin proteins in dissociated OHCs was examined using immunofluorescent staining and confocal microscopy. We found that prestin was uniformly expressed on the basolateral surface, including the basal pole. No staining was seen on the cuticular plate and stereocilia. When co-stained with a membrane marker di-8-ANEPPS, prestin-labeling was found to be in the outer layer of the OHC lateral wall. After separating the plasma membrane from the underlying subsurface cisternae using a hypotonic extracellular solution, prestin-labeling was found to be in the plasma membrane, not the subsurface cisternae. 4Plasma-Jet Forming of Sheet Metal Shapes显示文摘Plasma-jet forming is a newly proposed flexible sheet metal forming process. A non-transferred arc plasma torch is used as a controllable heat source to produce internal stress in sheet metals, causing plastic deformation without the necessity of hard tooling. This method has potential for rapid prototyping of sheet metal parts by reducing development costs and lead times. A robotic system has been used to perform simple linear bends in several different alloys. In order to develop a controllable process and to improve the forming accuracy, the effects of various process parameters on the obtained shape changes and on the resulting structure and properties have been studied. 6NUMERICAL SIMULATIONS OF THE EMISSION-LINE REGIONS OF QUASARS显示文摘The luminous quasars are the most distant objects we can directly observe. Once understood, their emission lines will measure the quasar’s luminosity and the composition of the interstellar medium of the host galaxy. Unfortunately the emitting plasma is far from equilibrium, and its conditions are set by a host of microphysical processes. The equations of statistical and thermal equilibrium must be solved to determine the ionization distribution, level populations, and kinetic temperature as a function of depth. Simultaneously the line and continuum radiative transfer problems are solved to predict the observed spectrum. A complete simulation involves many hundreds of stages of ionization, many thousands of levels, with populations determined by a vast sea of atomic and molecular processes, many with accurate cross sections and rate coefficients only now becoming available. This is a problem at the very forefront of atomic and computational physics. Once complete, we will be able to map out the first generations of stellar processing in the cores of massive galaxies, and directly chart the expansion of the universe when it had an age under a billion years.Gary J. 7Comparison study of trapped water in human erythrocytes by EPC and DSC method and its application显示文摘The novel differential scanning calorimetry method for determining trapped water volume of human red blood cell during freezing process has been reexamined. Results show that the final erythrocyte volume is 53% of its isotonic volume after freezing to -40℃. An electronic particle counter (MultisizerTM III, Beckman Coulter Inc., USA) was used to measure cell volume changes in response to hypertonic solution. Using this approach, when extracellular solution was 3186 mOsm, the equilibrium cell volume was found to be 57% of its isotonic value. Both results indicate that 34%—40% of intracellular water is trapped and cannot respond to osmotic difference between intra- and extracellular solution. These findings are consistent with the published data: at least 20%—32% of the isotonic cell water volume is retained within RBCs during freezing.
