3Antileishmanial assessment of leaf extracts from Pluchea carolinensis,Pluchea odorata and Pluchea rosea显示文摘Objective:To evaluate the antileishmanial activity of different extracts from three Cuban Pluchea species.Methods:In in vitro assays the IC50was calculated in the promastigotes and amastigotes forms as cytotoxicity in murine macrophages.In leishmaniasis cutanea experiment,mortality, weight loss,lesion size and burden parasite were measured.Results:Extracts evaluated showed inhibitive effect on growing of promastigote form;however,active extracts caused a high toxicity. Ethanol and n-hexane extracts demonstrated specific antileishmanial activity.Ethanol and n-hexane extracts from Pluchea carolinensis(P.carolinensis) caused similar inhibition against amastigote form.The intraperitoneal administration of the ethanol extract of P.carolinensis at 100 mg/kg prevented lesion development compared with control groups.Conclusions:The antileishmanial experiment suggests that ethanol extracts from P.carolinensis is the most promising.Further studies are still needed to evaluate the polential of this plant as a source of new antileishmanial agents.Marley Garcia Wilmer H Perera Ramon Scull Lianet Monzote 2011Asian Pacific Journal of Tropical Medicine2011,4,10:0
4Evaluation of Cuban Bacillus thuringiensis(Berliner,1911)(Bacillales:Bacillacea)isolates with larvicidal activity against Aedes aegypti(Linnaeus,1762)(Diptera:Culicidae)显示文摘Objective:To evaluate 11 Cuban native Bacillus(B.)thuringiensis isolates in order to select one with the best larvicidal activity against Aedes(Ae.)aegypti and low cytotoxicity.Methods:The cry and cyt genes of the isolates(A21,A51,L95,L910,M29,R84,R85,R87,R89,U81 and X48)were amplified by PCR.The influence of organic matter and NaCl on the larvicidal activity was tested by bioassays.Cytotoxicity was assayed on peritoneal macrophages of BALB/c mice.Results:The cyt1(Aa,Ab,Ba),cyt2,cry4aA,cry4Ba,cry11(Aa,Ba,Bb)and cry10 genes were identified in all native Cuban isolates.The larvicidal activity(LC_(90))of seven isolates was affected by the presence of organic matter in the water,while A21,A51,L910,R84,U81 and X48 had better LC_(50),LC_(90),LC_(95) than the 266/29-Ⅶ-98 control strain.The LC_(50) of two isolates was affected by the presence of NaCl and A21,A51,R85 isolate had better larvicidal activity than the 266/29-Ⅶ-98 control strain.In terms of toxicity against macrophages,the extracts of nine isolates were less cytotoxic than the control strains.Conclusions:Native isolate A21 had the main virulence factors against Ae.aegypti larvae,displayed a good larvicidal activity in presence of different factors related with Ae.aegypti breeding sites,and had low citotoxicity against macrophages.These results can contribute to the improvement of existing biological control strategies and the development of new biolarvicides.Aileen Gonzalez Rizo Camilo E Castañet Martinez Celeste Ramírez Cardentey Ariamys Companioni Ibañez Zulema Menéndez Díaz Lianet Monzote Fidalgo Hilda M HernandezÁlvarez 2022Asian Pacific Journal of Tropical Medicine2022,15,2:0
5HT-SuperSAGE of the gut tissue of a Vip3Aa-resistant Heliothis virescens (Lepidoptera: Noctuidae) strain provides insights into the basis of resistance显示文摘Multitoxin Bt-crops expressing insecticidal toxins with different modes of action, for example, Cry and Vip, are expected to improve resistance management in target pests. While Cry1A resistance has been relatively well characterized in some insect species, this is not the case for Vip3A, for which no mechanism of resistance has yet been identified. Here we applied HT-SuperSAGE to analyze the transcriptome of the gut tissue of tobacco budworm Heliothis virescens (F.) laboratory-selected for Vip3Aa resistance. From a total of 1 324 252 sequence reads, 5 895 126-bp tags were obtained representing 17 751 nonsingleton unique transcripts (UniTags) from genetically similar Vip3Aa-resistant (Vip- Sel) and susceptible control (Vip-Unsel) strains. Differential expression was significant (≥2.5 fold or ≤0.4;P < 0.05) for 1989 sequences (11.2% of total UniTags), where 420 represented overexpressed (OE) and 1569 underexpressed (UE) genes in Vip-Sel. BLASTN searches mapped 419 UniTags to H. virescens sequence contigs, of which, 416 (106 OE and 310 UE) were unambiguously annotated to proteins in NCBI nonredundant protein databases. Gene Ontology distributed 345 of annotated UniTags in 14 functional categories with metabolism (including serine-type hydrolases) and translation/ribosome biogenesis being the most prevalent. A UniTag homologous to a particular member of the REsponse to PAThogen (REPAT) family was found among most overexpressed, while UniTags related to the putative Vip3Aa-binding ribosomal protein S2 (RpS2) were underexpressed. qRT-PCR of a subset of UniTags validated the HT-SuperSAGE data. This study is the first providing lepidopteran gut transcriptome associated with Vip3Aa resistance and a foundation for future attempts to elucidate the resistance mechanism.Camilo Ayra-Pardo Maria E.
