2The allosteric modulation effects of doxycycline, minocycline, and their derivatives on the neuropeptide receptor PAC1-R显示文摘Class B G-protein coupled receptors (GPCR) PAC1-R is a neuropeptide pituitary adenylate cyclase activating polypeptide(PACAP)-preferring receptor that mediates the effective neuroprotective activity.Based on our previous data showing that doxycycline and minocycline work as the positive allosteric modulator (PAM) of RAC1-R,we used computer molecular docking and isothermal titration calorimetry assay to further determine the bindings of doxycycline/minocycline's derivatives including tetracycline/tigecycline with the N-terminal extracellular domain of PAC1-R (PAC1-EC1).Then the cAMP assay combined with the PAC1-R natural agonist PACAP27 was used to confirm the possible PAM roles of the small-molecule antibiotics. The results showed that tetracycline/tigecycline had significant lower affinity to PAC1-EC1 than doxycycline/minocycline, which was con sistent with their no n-positive allosteric modulation activity on PAC1-R.Furthermore, by comparing the key residues contributing to the PAM binding with the predicted allosteric site in PAC1-EC1,we characterized four motifs contributing to PAM binding in PAC1-EC1.The site-directed mutation results showed that ASN60 played the most important role in the PAM binding of the small-molecule antibiotics, while ASP116 played a sensitive marginal role in the PAM binding.These results not only help to explain the clinical and experimental neuroprotective effects of doxycycline/minocycline, but also help to characterize the PAM binding site in PAC1-EC1,which will promote the screening and characterization of novel small-molecule PAMs targeting PAC1-EC1 with drug development potency in nerve system disease.Suqin Song Like Wang Junfeng Li Xiaoling Huang Rongjie Yu 2019Acta Biochimica et Biophysica Sinica2019,51,6:1
3A universal tactic of using Lewis-base polymer-CNTs composites as additives for high performance cm2-sized and flexible perovskite solar cells显示文摘Lewis-base polymers have been widely utilized as additives to act as a template for the perovskite nucleation/crystal growth and passivate the under-coordinated Pb2+ sites.However,it is uncovered in this work that the polymer on the perovskite grain boundaries would significantly hinder the charge transport due to its low conductivity,which brings about free carrier recombination and photocurrent losses.To circumvent this issue while fully exploiting the benefits of polymers in passivating the trap states in perovskite,we incorporate highly conductive multiwall carbon nanotubes(CNTs) with Lewis-base polymers as coadditives in the perovskite film.Functionalizing the CNTs with-COOH group enables a selective hole-extraction and charge transport from perovskite to the hole transporting materials(HTM).By studying the charge transporting and recombination dynamics,we revealed the individual role of the polymer and CNTs in passivating the trap states and facilitating the charge transport,respectively.As a result,the perovskite solar cells(PSCs) with polymer-CNTs composites exhibit an impressive PCE of 21.7% for a small-area device(0.16 cm2) and 20.7% for a large-area device(1.0 cm2).Moreover,due to the superior mechanical flexibility of both polymer and CNTs,the polymer-CNTs composites incorporation in the perovskite film encourages the fabrication of flexible PSCs(f-PSCs) with an impressive PCE of 18.3%,and a strong mechanical durability by retaining 80%of the initial PCE after 1,000 times bending.In addition,we proved that the selection criteria of the polymers can be extended to other long-chain Lewis-base polymers,which opens new possibilities in design and synthesis of inexpensive material for this tactic towards the fabrication of high performance large-area PSCs and f-PSCs.Chang Liu Jingsong Sun Xiao-Fang Jiang Like Huang Qiang Lou Yi-Bing Cheng Shulin Song Ziyi Ge 2021Science China Chemistry2021,64,2:0
4Tocilizumab in rheumatoid arthritis-associated peripheral ulcerative keratitis:A case report显示文摘We report a case of rheumatoid arthritis-associated peripheral ulcerative keratitis(PUK)successfully treated with tocilizumab.Despite having been treated with systemic corticosteroids and several disease-modifying antirheumatic drugs and having,received corneal transplantation,the patient lost sight in the left eye.Tocilizumab resulted in a good response,with her systemic illness controlled and the progression of corneal lesions prevented.Tocilizumab may be an additional tool for patients with rheumatoid arthritis associated PUK that is refractory to other drugs.Jia Huang Zhen Chen Like Zhao Yongjing Cheng Ming Gao Cibo Huang Zheng Wang Fang Wang 2022Rheumatology & Autoimmunity2022,2,1:0
