1No apparent lock-in depth of the Laschamp geomagnetic excursion: Evidence from the Malan loess显示文摘The first-order chronological framework of the long-term Chinese loess/paleosol sequences was based mainly on magnetostratigraphy. However, there remain arguments on the lock-in ages of the major geomagnetic reversals. This study systematically compared the stratigraphic locations of the Laschamp excursion and Henrich events (H4 at~39 ka and H5 at~48 ka) recorded by the Luochuan loess profile in the central Chinese Loess Plateau. Results show that the Lascchamp excursion is bracketed well by these two cold events, and the corresponding estimated age is between about 41.7-43.7 ka, which is close to the widely accepted age of ~40-41 ka for the excursion.Therefore, our new results do not suggest apparent lock-in effects in the last glacial Malan loess.ZHU Rixiang1, LIU Qingsong1,2, PAN Yongxin1, DENG Chenglong1, ZHANG Rui1 & WANG Xianfeng3 1. 2Discovery and analysis of ultra-micro grinding grain texture in slipping lamellae of ductile-brittle zone显示文摘Based on the observation of the deep bore cores sampled from the State of Cali-fornia, USA, preferred arranged particulate concentration belts were found in the ultra-slipping lamellae. And the same phenomenon was found again later in the bore cores from the petroleum basin in the Northern Shaanxi Province. It may be the product of the dynamic processes involv-ing tumbling, grinding and compresso-shearing functions, so the spherical particulates with ul-tra-size of nm and mm are called grinding grain. The formation of the grinding grains is related with the ductile-brittle deformation, the lateral pressure of the slipping, and the very low grade of dynamic metamorphism of the rocks. Based on the systematic analysis, it is indicated that some elements like Nb and Ta are enriched in the grinding grains in the granitic rocks because of the chemical differentiation, and that the slipping lamellae bearing grinding grains in mudstone with smear efficiency prove a perfect seal for oil and gas reservoirs. 4Regulatory mechanisms for abnormal expression of the human breast cancer specific gene 1 in breast cancer cells显示文摘Breast cancer-specific gene 1 (BCSG1), also referred as synuclein γ, was originally iso-lated from a human breast cancer cDNA library and the protein is mainly localized to presynaptic terminals in the nervous system. BCSG1 is not expressed in normal or benign breast lesions, but expressed at an extremely high level in the vast majority of the advanced staged breast carcinomas and ovarian carcinomas. Overexpression of BCSG1 in cancer cells led to significant increase in cell proliferation, motility and invasiveness, and metastasis. To elucidate the molecular mechanism and regulation for abnormal transcription of BCSG1, a variety of BCSG1 promoter luciferase reporters were constructed including 3′ end deleted sequences, Sp1 deleted, and activator protein-1 (AP1) domains mutated. Transient transfection assay was used to detect the transcriptional activation of BCSG1 promoters. Results showed that the Sp1 sequence in 5′-flanking region was involved in the basal transcriptional activities of BCSG1 without cell-type specificity. In comparison to pGL3-1249, the reporter activities of pGL3-1553 in BCSG1-negative MCF-7 cells and pGL3-1759 in HepG2 cells were notably decreased. Mutations at AP1 sites in BCSG1 intron 1 significantly reduced the promoter ac-tivity in all cell lines. Transcription factors, c-jun, c-fos and cyclin AMP-responsive element binding (CREB) protein, could markedly enhance the promoter activities. Thus, our results suggest that the abnormal expression of BCSG1 in breast cancer cells is likely regulated by multiple mechanisms. The 5′ flanking region of BCSG1 provides the basal transcriptional activity without cell type specificity. A critical promoter element involved in abnormal expression of BCSG1 presents in the first exon. The cell type specificity of BCSG1 transcription is probably affected through intronic cis-regulatory se-quences. 5microRNAs-powerful repression comes from small RNAs显示文摘microRNAs (miRNAs) encode a novel class of small, non-coding RNAs that regulate gene expression post-trancriptionally. miRNAs comprise one of the major non-coding RNA families, whose diverse bio- logical functions and unusual capacity for gene regulation have attracted enormous interests in the RNA world. Over the past 16 years, genetic, biochemical and computational approaches have greatly shaped the growth of the field, leading to the identification of thousands of miRNA genes in nearly all metazoans. The key molecular machinery for miRNA biogenesis and silencing has been identified, yet the precise biochemical and regulatory mechanisms still remain elusive. However, recent findings have shed new light on how miRNAs are generated and how they function to repress gene expression. miRNAs provide a paradigm for endogenous small RNAs that mediate gene silencing at a genome-wide level. The gene silencing mediated by these small RNAs constitutes a major component of gene regu- lation during various developmental and physiological processes. 6Kinetics of early methane generation from Green River shale显示文摘During oil/gas exploration in an area, there is a question as to whether methane in the strata was generated \%in situ\% mainly by thermal degradation of local organic matter. The key to the question is whether the kerogen could produce methane of more than 80 mL/g TOC, when its \%R\%\-o increased from initial (0.36%) to current value of 0.70% during thermal evolution. The authors have made efforts to answer this question by the kinetics method. First, a series of pyrolysis experiments were carried out to obtain data of methane yield and \%R\%\-o evolution, from which kinetic parameters of methane and \%R\%\-o were derived; then these parameters were extrapolated to the geological condition and finally, a methane yield, 96 mL/g TOC, was predicted. 7Posterior gut development in Drosophila:a model system for identifying genes controlling epithelial morphogenesis显示文摘The posterior gut of the Drosophila embryo, consisting of hindgut and Malpighian tubules, provides a simple,well-defined system where it is possible to use a genetic approach to define components essential for epithelial morphogenesis.We review here the advantages of Drosophila as a model genetic organism, the morphogenesis of the epithelial structures of the posterior gut, and what is known about the genetic requirements to form these structures.In overview, primordia are patterned by expression of hierarchies of transcription factors; this leads to localized expression of cell signaling molecules, and finally, to the least understood step: modulation of cell adhesion and cell shape. 8Macroscopic relationship for preferential flow in the vadose zone:Theory and validation显示文摘Preferential flow commonly observed in unsaturated soils allows rapid movement of solute from the ground surface or vadose zone to the groundwater, bypassing a significant volume of unsaturated soil and increasing the risk of groundwater contamination. A variety of evidence indicates that complex preferential flow patterns observed from fields are fractals. This paper discusses a macroscopic relationship for modeling preferential flow in the vadose zone. Conceptually, the flow domain can be divided into active and inactive regions. Flow occurs preferentially in the active region (characterized by fractals), and inactive region is simply bypassed. The portion of the active region was found to be a power function of saturation.
