1大疱性类天疱疮和妊娠疱疹患者血清抗BP180NC16A抗体的纯化和鉴定显示文摘目的 探讨大疱性类天疱疮(BP)和妊娠疱疹(HG)患者血清抗BP180 NC16A抗体的纯化和鉴定方法.方法 原核表达系统pGEX-2TBP180NC16A表达GST/NC16A融合蛋白,将融合蛋白与谷胱甘肽琼脂糖凝聚微珠进行共价偶联.微珠亲和层析法纯化BP和HG患者血清中抗BP180 NC16A抗体,并用ELISA、免疫荧光、及Western印迹进行鉴定.结果 原核表达系统pGEX-2TBP180NC16A表达37 000GST/NC16A融合蛋白,微珠亲和层析法纯化后得单一抗BP180 NC16A抗体.经ELISA方法定量后确定其含量为2.4 mg/ml;该抗体能与人皮肤基底膜带结合,证明抗体活性;免疫印迹可见单一片段,显示抗体纯度.结论 微珠亲和层析法纯化的BP和HG患者血清中抗BP180 NC16A自身抗体活性高、特异性强.左亚刚 LIUZhi 2012中华皮肤科杂志2012,45,1:2

8Identification of Expression Pattern and Clinical Significance of the Small Cajal Body-specific RNA SCARNA16 in Hepatocellular Carcinoma显示文摘Background and Aims:For high morbidity and mortality,hepatocellular carcinoma(HCC)becomes a major health issue worldwide.Nowadays,numerous non-coding RNAs(ncRNAs)are known to regulate the occurrence and patho-genesis of tumors.Some ncRNAs have also been developed as tumor biomarkers and therapeutic targets.However,the potential function of the small Cajal body-specific RNA(scaRNA)SCARNA16,a newly identified ncRNA,remains to be explored in HCC.Methods:In both HCC cell lines and specimens from 120 enrolled patients,the expression val-ues of SCARNA16 were detected.We divided patients into SCARNA16 high and low expression subgroups,and then analyzed the difference of various clinical characteristics and prognosis data between subgroups.Results:Compared to paired controls,SCARNA16 was significantly down-regulated in HCC cell lines and clinical specimens(p<0.01).Besides,HCC patients with lower SCARNA16 expression commonly presented with larger and more tumor lesions,more ves-sel carcinoma emboli,more capsular invasion and higher TNM stages(p<0.05).Moreover,SCARNA16 expression was negatively correlated with postoperative prognosis of HCC patients in 5-year follow-up,including tumor-free survival(TFS)(median time of low vs.high subgroups:14 vs.48 months,p=0.006)and overall survival(OS)(median time of low vs.high subgroups:39 vs.52 months,p=0.001).Besides,SCARNA16 acted as an independent prognostic bio-marker in TFS(hazard ratio[HR]:0.578,95%CI:0.345-0.969,p=0.038)and OS(HR:0.366,95%CI:0.178-0.752,p=0.006).Conclusions:Low expression patterns of SCAR-NA16 remarkably associated with severe clinical status and poor survival of patients,suggesting that SCARNA16 pos-sesses potency as a novel biomarker for HCC.Sitong Zhang Yuan Ding Zhongquan Sun Yao Ge Yanjie Li Xin Han Qianhui Xu Liuzhi Zhou Yining Chen Hao Xu Yang Bai Chang Xu Hao Ding Sheng Yan Weilin Wang 2022Journal of Clinical and Translational Hepatology2022,10,1:0

10Compensatory Growth of Juvenile Brown Flounder Paralichthys olivaceus Following Low Temperature Treatment for Different Periods显示文摘We investigated the effects of low temperature(8.5℃) on the growth and feeding rates and feed conversion efficiency of juvenile P.olivaceus with an average initial weight of 3.87 ± 0.06 g(mean ± SE).Fish were exposed to 8.5℃ for 0(control),1,2,3 and 4 weeks,and then to 20℃ for 10,9,8,7 and 6 weeks,respectively.Low temperature clearly led to growth depression.The weight of fish exposed to low temperature for 1 week was restored to that of control,while that of fish exposed to low temperature longer was significantly decreased(P < 0.05).During the entire low-temperature period,specific growth rate,feeding rate and feed conversion efficiency of the fish were significantly lower(P < 0.05) than those of control,while in the whole recovery period,specific growth and average feeding rate were markedly higher(P < 0.05) than those of control.At the end of experiment,only the feeding rate of the fish exposed to low temperature for 1 week was not significantly different from that of control(P > 0.05).Feeding rate and feed conversion efficiency were reduced at low temperature in juvenile P.olivaceus.The compensatory growth of juvenile P.olivaceus may therefore be attributed to the improvement of feeding rate.Our results suggested that growth depression occurs when juvenile P.olivaceus are exposed to low temperature for more than one week.PENG Yinhui LIU Xiujia HUANG Guoqiang WEI Liuzhi ZHANG Xiumei 2017Journal of Ocean University of China2017,16,2:0
11miR-204 ameliorates osteoarthritis pain by inhibiting SP1-LRP1 signaling and blocking neuro-cartilage interaction显示文摘Osteoarthritis(OA)is a painful degenerative joint disease and is the leading cause of chronic disability among elderly individuals.To improve the quality of life for patients with OA,the primary goal for OA treatment is to relieve the pain.During OA progression,nerve ingrowth was observed in synovial tissue and articular cartilage.These abnormal neonatal nerves act as nociceptors to detect OA pain signals.The molecular mechanisms for transmitting OA pain in the joint tissues to the central nerve system(CNS)is currently unknown.MicroRNA miR-204 has been demonstrated to maintain the homeostasis of joint tissues and have chondro-protective effect on OA pathogenesis.However,the role of miR-204 in OA pain has not been determined.In this study,we investigated interactions between chondrocytes and neural cells and evaluated the effect and mechanism of miR-204 delivered by exosome in the treatment of OA pain in an experimental OA mouse model.Our findings demonstrated that miR-204 could protect OA pain by inhibition of SP1-LDL Receptor Related Protein 1(LRP1)signaling and blocking neuro-cartilage interaction in the joint.Our studies defined novel molecular targets for the treatment of OA pain.Ke Lu Qingyun Wang Liuzhi Hao Guizheng Wei Tingyu Wang William WLu Guozhi Xiao Liping Tong Xiaoli Zhao Di Chen 2023Bioactive Materials2023,,8:0
