| 1 | 3D printed silk-gelatin hydrogel scaffold with different porous structure and cell seeding strategy for cartilage regeneration显示文摘Hydrogel scaffolds are attractive for tissue defect repair and reorganization because of their human tissue-like characteristics.However,most hydrogels offer limited cell growth and tissue formation ability due to their submicron-or nano-sized gel networks,which restrict the supply of oxygen,nutrients and inhibit the proliferation and differentiation of encapsulated cells.In recent years,3D printed hydrogels have shown great potential to overcome this problem by introducing macro-pores within scaffolds.In this study,we fabricated a macroporous hydrogel scaffold through horseradish peroxidase(HRP)-mediated crosslinking of silk fibroin(SF)and tyramine-substituted gelatin(GT)by extrusion-based low-temperature 3D printing.Through physicochemical characterization,we found that this hydrogel has excellent structural stability,suitable mechanical properties,and an adjustable degradation rate,thus satisfying the requirements for cartilage reconstruction.Cell suspension and aggregate seeding methods were developed to assess the inoculation efficiency of the hydrogel.Moreover,the chondrogenic differentiation of stem cells was explored.Stem cells in the hydrogel differentiated into hyaline cartilage when the cell aggregate seeding method was used and into fibrocartilage when the cell suspension was used.Finally,the effect of the hydrogel and stem cells were investigated in a rabbit cartilage defect model.After implantation for 12 and 16 weeks,histological evaluation of the sections was performed.We found that the enzymatic cross-linked and methanol treatment SF5GT15 hydrogel combined with cell aggregates promoted articular cartilage regeneration.In summary,this 3D printed macroporous SF-GT hydrogel combined with stem cell aggregates possesses excellent potential for application in cartilage tissue repair and regeneration. | Qingtao Li Sheng Xu Qi Feng Qiyuan Dai Longtao Yao Yichen Zhang Huichang Gao Hua Dong Dafu Chen Xiaodong Cao | 2021 | Bioactive Materials2021,6,10: | 6 |
| 2 | Piceatannol enhances Beclin-1 activity to suppress tumor progression and its combination therapy strategy with everolimus in gastric cancer显示文摘The effects and regulation of Beclin-1-an autophagy-related protein-have not been fully defined, however, a negative correlation has been reported between Beclin-1 expression and carcinogenesis. Meanwhile, no compound has been shown to directly inhibit its activity. Here, we evaluate piceatannol, a naturally occurring polyphenolic compound, as a potential targeting agonist of Beclin-1, to assess its efficacy as an antitumor agent against gastric cancer. More specifically, we determine the effects of piceatannol treatment on cell viability using a monitoring system and colony forming assay. Piceatannol was found to efficiently inhibit the proliferation of several human gastric cancer cell lines. Autophagic flux is increased by piceatannol treatment, and correlates with inhibition of cell proliferation and colony formation. Additionally, microscale thermophoresis and surface plasmon resonance results show a direct interaction between piceatannol and Beclin-1, which reduces the phosphorylation activity of Beclin-1 at the Ser-295 site. Notably, piceatannol impairs the binding of Beclin-1 to Bcl-2 and enhances the recruitment of binding of UV radiation resistance-associated gene protein, which further triggers Beclin-1-dependent autophagy signaling. An increase in autophagic activity via treatment with the mTOR inhibitor, everolimus, effectively sensitizes piceatannol-induced antitumor effects. Xenograft models confirmed that piceatannol inhibits tumor development and elicits a potent synergistic effect with everolimus in vivo. | 3 | Upconversion nanoparticles regulated drug&gas dual-effective nanoplatform for the targeting cooperated therapy of thrombus and anticoagulation显示文摘Thromboembolism is the leading cause of cardiovascular mortality.Currently,for the lack of targeting,short half-life,low bioavailability and high bleeding risk of the classical thrombolytic drugs,pharmacological thrombolysis is usually a slow process based on micro-pumping.In addition,frequently monitoring and regulating coagulation functions are also required during(and after)the process of thrombolysis.To address these issues,a targeted thrombolytic and anticoagulation nanoplatform(UCATS-UK)is developed based on upconversion nanoparticles(UCNPs)that can convert 808 or 980 nm near-infrared(NIR)light into UV/blue light.This nanoplatform can target and enrich in the thrombus site.Synergistic thrombolysis and anticoagulation therapy thus could be realized through the controlled release of urokinase(UK)and nitric oxide(NO).Both in vitro and in vivo experiments have confirmed the excellent thrombolytic and anticoagulative capabilities of this multifunctional nanoplatform.Combined with the unique fluorescent imaging capability of UCNPs,this work is expected to contribute to the development of clinical thrombolysis therapy towards an integrated system of imaging,diagnosis and treatment. | Shichen Liu Yao Sun Teng Zhang Longtao Cao Zhiwei Zhong Haoxin Cheng Qingqing Wang Zhuang Qiu Weimin Zhou Xiaolei Wang | 2022 | Bioactive Materials2022,7,12: | 1 |
| 4 | Crustal activities recorded incoral reefs in the northwestern South China Sea显示文摘Coral reefs in the northwest of South China Sea have recorded the information from not only the environmental variation but also the crustal activities there during their development. The main crustal activities correlated with the coral reef development include fault, seismic, and volcano activities, etc. The high-resolution spark seismic profiles in the northwestern South China Sea show that the fault activities in the coral reef region have been clearly recorded, and appear as neonatal faults incising reefs. Earthquakes in the coral reef region are rather intense, especially the two occurring on December, 31, 1994, and January, 10, 1995, around the southwest of Leizhou Peninsula, with the magnitude of 6.1 and 6.2, respectively. They have great influence on the growth of the local coral reefs. Quaternary volcanos are active in the northwestern South China Sea, especially around the southwest of Leizhou Peninsula, and they have obvious control of the coral reef development.
