| 3 | Application of single and combination therapy of clarithromycin and tamoxifen to suppress breast cancer cell proliferation and metabolism显示文摘Objective This study compares the anti-tumor effects of single and combination use of clarithromycin and tamoxifen on estrogen receptor (ER) positive breast cancer cell lines,BT-483 and MCF-7 as well as triple negative cell line,MBA-MD-231,which acts as a negative control.The effect of solid breast tumor inhibition by clarithromycin is also studied.Method BT-483,MCF-7 and MBA-MD-231 were cultured in 6-well plates in a 37 ℃ humidified incubator without CO2 for 24 h prior to the addition of the test drugs.The test groups were clarithromycin (Group 1),tamoxifen (Group 2),clarithromycin and tamoxifen (Group 3),and control (Group 4).Group 3 was prepared in 1 to 1 ratio at a concentration of 1.5 mmol/L clarithromycin and 25 μmol/L tamoxifen.On the other hand,1 mm3 solid breast tumors were submerged into various groups as above for 24 h.On the harvest day,the proliferation of cancer cells and solid breast tumor samples were measured by WST-1 proliferation reagent while ATP bioluminescence assay was employed to measure the metabolic rate of the three cell lines.Results The proliferation of BT-483 and MCF-7 was suppressed most by combination use of clarithromycin and tamoxifen with statistical significance.The two drugs did not have an inhibitory effect on the hormonal negative cancer cells.For solid breast tumor samples,all the test groups showed reduced metabolic rate as compared with the control group (P<0.05).Conclusion Combination use of tamoxifen and clarithromycin are effective in suppressing cell proliferation and metabolism rate of breast cancer cells while single use of clarithromycin effectively inhibits the proliferation of solid breast tumor. | Wings T.Y.Loo Louis W.C.Chow Adrian Y.S Yip Mary N.B.Cheung | 2012 | 中华乳腺病杂志(电子版)2012,6,1: | 1 |

| 9 | Additive manufacturing of antibacterial PLA-ZnO nanocomposites:Benefits,limitations and open challenges显示文摘Polymeric biomaterials such as polylactic acid(PLA)play a prominent role in the advancement of biomedical additive manufacturing(AM).PLA offers indeed a very advantageous combination of thermomechanical properties and functional attributes,as it is biobased,biodegradable,biocompatible and easy to print.However,PLA can be damaged by common sterilization methods and is sensitive to most chemical disinfectants,and this may impair its widespread usage.One of the most promising ways to overcome this shortcoming is to provide PLA with embedded antibacterial activity by the addition of appropriate fillers such as zinc oxide(Zn O)nanoparticles.After a detailed introduction to the basic properties of PLA and ZnO nanoparticles,the present review analyzes the main variables that govern the antibacterial activity of PLA-ZnO nanocomposites.Current applications and related manufacturing processes are also presented to showcase the importance of having embedded antibacterial functions in demanding applications such as food packaging and wound dressing.Emphasis is then placed on the emerging literature of the AM of PLA-ZnO nanocomposites,with a focus on fused filament fabrication(also known as fused deposition modeling).Existing gaps and hurdles related to the development and 3D printing of such composites is critically discussed.It is envisioned that a deeper understanding of the processability,thermo-mechanical behavior,biocompatibility and antibacterial efficacy of additively manufactured PLAZnO nanocomposites will foster their adoption in the biomedical field and,ultimately,in all circumstances where it is crucial to limit infection transmission. | Wei Juene Chong Shirley Shen Yuncang Li Adrian Trinchi Dejana Pejak Ilias(Louis)Kyratzis Antonella Sola Cuie Wen | 2022 | Journal of Materials Science & Technology2022,,16: | 1 |

| 11 | Application of biomarkers in sentinel lymph node biopsy for the management of breast cancer显示文摘Objective Sentinel lymph node in breast cancer is a predictor of the tumor's metastatic potential.Cytokines emerging from antitumor immune response might also target sentinel lymph node (SLN).In this study,we evaluated the cytokine profile including interleukin-8 (IL-8),interleukin-12 (IL-12),interferon-gamma (IFN-γ),and tumor necrosis factor alpha (TNF-α) for the T-cell response.Methods From January 2008 to August 2010,35 patients with breast mass underwent SLN biopsy in UNIMED Medical Institute.The specimens were formalin-fixed and paraffin-embedded for hematoxylin and eosin staining and immunohistochemistry with following markers:IL-8,IL-12,IFN-γ and TNF-α.The staining intensity was scored according to four grades:negative (-),weak expression (+),moderately positive expression (+ +),strong positive expression (+ + +).The correlation between the expression of cytokines and pathological characteristics were also analyzed using the Spearman Rank Correlation test.Results Out of 35 patients,SLN metastases were observed in 20 patients and among them,75% (15 /20) were strongly stained (+ + +) with IL-8 and moderately stained (+ +) with IL-12,but only 25% (5 /20) were strongly stained (+ + +) with IFN-γ and TNF-α.The expression of IL-8 was significantly associated with all tumor grades (r=0.639,P=0.00),estrogen receptor (r=0.778,P=0.01),progesterone receptor (r=0.759 P=0.00) and HER-2 /neu receptor (r=0.863 P=0.00).Conclusion The strong association between IL-8 expression and SLN metastases might highlight the prognostic significance of IL-8 in SLN. | Qing Liu Wings Tjing Yung Loo Adrian Yun San Yip Louis Wing Cheong Chow Elizabeth Lam Yan Ng Mary Ngan Bing Cheung | 2012 | 中华乳腺病杂志(电子版)2012,6,6: | 1 |

| 14 | Potential of a COX-2 inhibitor in lowering chemotherapy-induced neutropenia显示文摘Objective This study was initially designed to evaluate the effect of celecoxib on the regimen of 5-fluorouracil,epirubicin,and cyclophosphamide(FEC)combination,followed by docetaxel(T)in neoadjuvant setting.An unplanned preliminary review on safety was conducted after a halt of the study due to the concerned potential cardiovascular risk of using COX-2 inhibitors.Methods We studied 23 consecutive cases of operable breast cancer having received four cycles of FEC(500 mg/m2,100 mg/m2,500 mg/m2)followed by four cycles of T(100 mg/m2)with concurrent celecoxib(400 mg twice daily)(group A)or same chemotherapy regimen but without concurrent celecoxib(group B).These combined chemotherapies were administered every 3 weeks.The Chi-square test or Fisher's exact test were used to assess the difference in incidence of limiting hematological toxicites between groups.Results 23 patients(group A:n=12;group B,n=11)received a total of 183 out of 184 planned treatment cycles;one(4%,1/23)of them omitted the fourth cycle of FEC owing to repeated incidences of febrile neutropenia.Received dose intensity(RDI)for FEC in group A(90%±11%)was higher than that in group B(80%±8%)while RDI for T was similar between group A(93%±8%)and group B(96%±9%).Of the first 91 treatment cycles of FEC,limiting hematological toxicity,severe neutropenia including febrile neutropenia,was significantly different between group A and B [(10.4%,5/48)vs.(32.6%,14/43),P=0.009].Other toxicities commonly observed in chemotherapy receiving patients were manageable.Conclusions Neoadjuvant use of FEC followed by T with concurrent celecoxib appeared to be safe for treatment of operable invasive breast cancer.The observed lower incidence of chemotherapy-induced neutropenia is possibly contributed by the administration of COX-inhibitor.We believe that further investigation might provide more evidence on the use of COX-2 inhibitors in breast cancer. | Louis Wing-Cheong Chow Adrian Yun-San Yip Eleanor Yuen-Yuen Ong Chi-Kei Lam Masakazu Toi | 2010 | 中华乳腺病杂志(电子版)2010,4,5: | 0 |

| 16 | Effect of Tai Chi Chuan on serotonin and cortisol for monitoring stress and quality of life in post-treatment breast cancer patients显示文摘Objective Immediate physical exercise has been recommended for patients in the recovery phase to improve survival and quality of life(QOL)and reduce recurrence of disease.The new NCCN Guidelines for Survivorship also highlighted the role of exercise in post-cancer health,encouraging patients to perform light physical activity following treatment.The aim of our study is to effect of Tai Chi Chuan(TCC)on serotonin and cortisol for monitoring stress and QOL in post-treatment breast cancer patients.Methods Totally85 post-treatment breast cancer patients were enrolled in this study to observe the effects of practicing TCC on recovery,as well as stress and happiness which are indicators of QOL of in patients.Peripheral blood was drawn from study subjects to analyze the levels of serotonin,cortisol and high sensitive C-reactive protein(HSCRP)at baseline,and at 3,6 and 12 months of TCC practice.Blood was drawn from healthy subjects only at baseline.A QOL questionnaire was administered to study subjects at three time points throughout the study,and once for healthy controls.The data were processed by analysis of variance of repeated measurement.Results At 3,6 and 12 months time points following regular TCC exercise,WBC,RBC,hemoglobin in blood samples showed a statistically significant difference(F=161.55,172.14,289.73;all P=0.00);the level of serotonin(biomarker for well-being),cortisol(indicator of stress)and HS-CRP(biomarker for inflammation)showed a statistical improvement(F=307.46,182.85,102.23;all P=0.00).After 3,6 and 12 months of regular TCC exercise,according to the results of QOL questionnaire,the indicators including quality of sleep,perceived hunger,fatigue,contentment,stress and social interaction presented a significant difference(F=312.98,222.64,543.90,46.05,28.10,78.92,all P<0.05),while there was no statistical difference in life dissatisfaction(F=56.61,P=0.166)Conclusions TCC physical activity for post-treatment breast cancer patients improved QOL and overall well-being,leading to improved mental,physical and psychological functioning.Regulated levels of serotonin and cortisol mediated by TCC exercises are proved to be vital for continued good health. | William Wai Nam Tsang Wings Tjing Yung Loo Louis Wing Cheong Chow Elizabeth Lam Yan Ng Adrian Yun San Yip Jianping Chen | 2014 | 中华乳腺病杂志(电子版)2014,8,4: | 0 |
