1The Role and Function of Ras-association domain family in Cancer:A Review显示文摘Ras gene mutation has been observed in more than 30%of cancers,and 90%of pancreatic,lung and colon cancers.Ras proteins(K-Ras,H-Ras,N-Ras)act as molecular switches which are activated by binding to GTP.They play a role in the cascade of cell process control(proliferation and cell division).In the inactive state,transforming GTP to GDP leads to the activation of GTpase in Ras gene.However,the mutation in Ras leads to the loss of internal GTPase activity and permanent activation of the protein.The activated Ras can promote the cell death or stop cell growth,which are facilitated by Ras-association domain family.Various studies have been conducted to determine the importance of losing RASSF proteins in Rasinduced tumors.This paper examines the role of Ras and RASSF proteins.In general,RASSF proteins can be used as a suitable means for targeting a large group of Ras-induced tumors.Mohammad Reza Zinatizadeh Seyed Ali Momeni Peyman Kheirandish Zarandi Ghanbar Mahmoodi Chalbatani Hassan Dana Hamid Reza Mirzaei Mohammad Esmaeil Akbari Seyed Rouhollah Miri 2019Genes & Diseases2019,6,4:10
2The Nuclear Factor KappaB(NF-kB)signaling in cancer development and immune diseases显示文摘The nuclear factor kappa B(NF-kB)family of transcription factors plays an essential role as stressors in the cellular environment,and controls the expression of important regula-tory genes such as immunity,inflammation,death,and cell proliferation.NF-kB protein is located in the cytoplasm,and can be activated by various cellular stimuli.There are two path-ways for NF-kB activation,as the canonical and non-canonical pathways,which require com-plex molecular interactions with adapter proteins and phosphorylation and ubiquitinase enzymes.Accordingly,this increases NF-kB translocation in the nucleus and regulates gene expression.In this study,the concepts that emerge in different cellular systems allow the design of NF-kB function in humans.This would not only allow the development for rare dis-eases associated with NF-kB,but would also be used as a source of useful information to elim-inate widespread consequences such as cancer or inflammatory/immune diseases.Mohammad Reza Zinatizadeh Bettina Schock Ghanbar Mahmoodi Chalbatani Peyman Kheirandish Zarandi Seyed Amir Jalali Seyed Rouhollah Miri 2021Genes & Diseases2021,8,3:8
3Weathering and soils formation on different parent materials in Golestan Province,Northern Iran显示文摘Geochemical,mineralogical,and micromorphological characteristics of soils and their relevant parent rocks including loess,ignimbrite,sandstone and limestone were investigated to identify the soil-parent material uniformity and the weathering degree of soils in Golestan Province,northern Iran.Highly developed Calcixerolls and moderately developed Haploxerepts were formed on loess and limestone,respectively.In contrast,the soils formed on ignimbrite and sandstone were nondeveloped Entisols.Illite was the dominant clay mineral found in ignimbrite and sandstone in both the A horizon and parent material.In loess derived soils however,smectite was dominant especially in the Bt horizon compared to its parent material indicating partly to its pedogenic formation.In limestone,illite and vermiculite were dominant both in the A and C horizons.Ti/Zr ratio proved that the studied soils were closely related to their underlying parent materials geochemically.Chemical index of alteration(CIA),micromorphological index of soil development(MISECA),smectite/illite+chlorite ratio and magnetic susceptibility were applied to investigate the degree of soil development.Results showed that the most and the least developed soils were those formed on loess deposits and limestone,respectively.Application of the different geochemical and pedogenetic approaches was proved to be useful in identifying the relevance of soils to their underlying parent materials and also their degree of development.Maryam MAHMOODI Farhad KI-IORMALI Arash AMINI Shamsollah AYOUBI 2016Journal of Mountain Science2016,13,5:6
4Effect of crocin carotenoid on BDNF and CREB gene expression in brain ventral tegmental area of morphine treated rats显示文摘Objective: To investigate the effect of crocin carotenoid on BNDF and CREB gene expression in the brain ventral tegmental area(VTA) and the serum level of BDNF in morphine-treated rats compared to control. Methods: In this study, 40 male Wistar rats(200-250 g) were used in 5 experimental groups: 1) non morphine treat rats(control); 2) non morphine-treated rats with 25 mg/kg crocin carotenoid(i.p., for 21 d); 3) morphine treated rats(10 mg/kg twice a day, s.c., 21 d); 4 and 5) morphine-treated rats with 12.5 and 25 mg/kg crocin carotenoid, respectively. By the end of research, BDNF and CREB expression was determined by real-time-PCR method. ELISA analysis was also applied for assessing the serum BDNF level. Results: The data indicated that morphine treatment could cause a significant decrease in BDNF and CREB gene expression(P<0.01 and P<0.001, respectively) in brain VTA as well as serum level of BDNF(P<0.01) in comparison to control group. Treatment with 25 mg/kg crocin carotenoid caused a significant enhancement in BDNF and CREF gene expression(P<0.01 and P<0.05, respectively) and serum level of BDNF(P<0.01) in morphine-treated rats in comparison to morphine-treated group. 5The Hippo Tumor Suppressor Pathway(YAP/TAZ/TEAD/MST/LATS)and EGFR-RAS-RAF-MEK in cancer metastasis显示文摘Hippo Tumor Suppressor Pathway is the main pathway for cell growth that regulates tissue enlargement and organ size by limiting cell growth.This pathway is activated in response to cell cycle arrest signals(cell polarity,transduction,and DNA damage)and limited by growth factors or mitogens associated with EGF and LPA.The major pathway consists of the central kinase of Ste20 MAPK(Saccharomyces cerevisiae),Hpo(Drosophila melanogaster)or MST kinases(mammalian)that activates the mammalian AGC kinase dmWts or LATS effector(MST and LATS).YAP in the nucleus work as a cofactor for a wide range of transcription factors involved in proliferation(TEA domain family,TEAD1-4),stem cells(Oct4 mononuclear factor and SMAD-related TGFb effector),differentiation(RUNX1),and Cell cycle/apoptosis control(p53,p63,and p73 family members).This is due to the diverse roles of YAP and may limit tumor progression and establishment.TEAD also coordinates various signal transduction pathways such as Hippo,WNT,TGFb and EGFR,and effects on lack of regulation of TEAD cancerous genes,such as KRAS,BRAF,LKB1,NF2 and MYC,which play essential roles in tumor progression,metastasis,cancer metabolism,immunity,and drug resistance.However,RAS signaling is a pivotal factor in the inactivation of Hippo,which controls EGFR-RAS-RAF-MEK-ERKmediated interaction of Hippo signaling.Thus,the loss of the Hippo pathway may have significant consequences on the targets of RAS-RAF mutations in cancer.Mohammad Reza Zinatizadeh Seyed Rouhollah Miri Peyman Kheirandish Zarandi Ghanbar Mahmoodi Chalbatani Catarina Raposo Hamid Reza Mirzaei Mohammad Esmaeil Akbari Habibollah Mahmoodzadeh 2021Genes & Diseases2021,8,1:5
6Pentoxifylline in hepatopulmonary syndrome显示文摘AIM:To determine the effects of pentoxifylline(PTX) on clinical manifestations and evaluate arterial blood gas data in hepatopulmonary syndrome(HPS) in children.METHODS:In a pilot study of 10 children with chronic liver disease,who had HPS,20 mg/kg/d PTX was administered for 3 mo.Clinical data and arterial blood gas parameters were evaluated at baseline,the end of the treatment period,and 3 mo after drug discontinuation.RESULTS:Six patients could tolerate PTX,while four patients experienced complications.Among patients who could tolerate PTX,there was a significant increase in arterial oxygen pressure(PaO 2)(P = 0.02) and oxygen saturation(SaO 2)(P = 0.04) and alveolar-arterial oxygen gradient(P = 0.02) after 3 mo of treatment.Significant decreases in PaO 2(P = 0.02) and alveolararterial oxygen gradient(P = 0.02) were also seen after drug discontinuation.CONCLUSION:PTX may improve PaO 2,SaO 2 and alveolar-arterial oxygen gradient in the early stage of HPS.Hamid Reza Kianifar Maryam Khalesi Eftekhar Mahmoodi Monavar Afzal Aghaei 2012World Journal of Gastroenterology2012,18,35:4
7Anti-cancer effects of hydro-alcoholic extract of pericarp of pistachio fruits显示文摘Objective:To investigate the cytotoxicity and anti-cancer effects of hydro-alcoholic extract of pistachio pericarp on hepatocellular carcinoma cells(HepG2)and mouse fibroblast L929 cells as normal and control group cell.Methods:MTT assay was performed to investigate the cytotoxicity effects of the extract at 0-4 000μg/mL on the cells after 24 and48 h.The expressions of some genes involved in apoptosis including Bax,Bcl-2 and P53were investigated by real time PCR.Results:Our results showed that after 24 and 48 hours of treatment of cells with this extract,the viability of HepG2 and L929 cells was reduced.Therefore,this extract had the cytotoxicity effect on both cells.The IC_(50) concentration of extract for HepG2 cells after 24 and 48 hours of treatment was 1 500 and 1 000μg/mL and for L929 cells was 2 000 and 1 500μg/mL,respectively.The expressions of Bax and P53 genes were up-regulated after treatment in the HepG2 and L929 cells and the expression of Bcl-2gene was down-regulated after treatment of extract in HepG2 cell.Conclusions:According to the results of MTT assay and real time PCR,this extract can be considered as a potential candidate for use in the production of anti-cancer drugs for the treatment of patients with liver cancer in future.Hamidreza Harandi Ahmad Majd Soudeh Khanamani Falahati-pour Mehdi Mahmoodi 2018Asian Pacific Journal of Tropical Biomedicine2018,8,12:2
8Experimental investigation of flow and distortion mitigation by mechanical vortex generators in a coupled serpentine inlet-turbofan engine system显示文摘The performance of compact,aggressive ducts in advanced propulsion systems is limited by the internal flow separation coupled with the formation of secondary counter-rotating vortices that give rise to intensive flow distortions at the duct exit.An experimental investigation was conducted to study the flow field and passive suppression of flow separation and Aerodynamic Interface Plane (AIP) distortion in a serpentine air inlet duct.Tests were performed by a turbofan engine at several Engine Operating Points (EOPs) from 56%(idle) to 100%(max).A large total pressure deficit region arose at the upper part of the AIP,which was associated with the upper surface flow separation.Using the new mechanical S-type vortex generators in two longitudinal positions (VGl and VG2),separation and loss were diminished at the upper part of the duct and AIP.The VG2 arrangement attained the maximum reductions in distortion coefficients which were 73.72%,60.7% and 37.8% in DC(90°),DC(60°) and△P_C/P metrics,respectively.In the next step of the study,some unsteady aspects of the flow field were analyzed inside the duct.The separation onset and reattachment points were determined by the standard deviation of static pressure on the upper surface.The AIP spectral distribution showed that the boundary of low pressure and high pressure recovery regions was dominated by the maximum fluctuations.Furthermore,the PSD diagram of several probes at AIP revealed the vortex shedding frequency and its higher harmonics at separation region.The energy content of distinct unsteady spectral features in the bare configuration was significantly reduced using VGs,which showed the improvement of flow at the duct exit.Iman MAGHSOUDI Mohammad Ali VAZIRY Mostafa MAHMOODI 2020Chinese Journal of Aeronautics2020,33,5:2
9Micromorphological Evidences of Climatic Change in Yazd Region,Iran显示文摘The fact of, present is the key of the past, will help us to use paleosols properties as indicators of the ecological characteristics of past time, particularly the paleoclimate. In this respect the micromorphological properties showed to be a very good indicator. Therefore, for investigating of climate change in Ardakan-Yazd plain, Central Iran 9 pedons were digged and described. Yazd has an arid climate with less than 100 mm annual precipitation and more than 22°C mean annual temperature (Aridic-hyper thermic soil moisture and temperature regions, respectively). Based on the morphological and physicochemical analysis Arglic, Calcic and Gypsic diagnostic horizons have been distinguished in these soils. Thin section studied showed that the illuviated form of clay includes, infillings on channel, coating on pendant, on nodules and on grains, at lower depths and also juxtaposed calcite needles on void argillan at upper part of the profiles. Mineralogical result showed fine clay in arglic horizon, too. Considering depth and forms of these pedofeatures, we concluded that, the observed illuviated clays at lower depth must be the result of the more humid climate of the past, where the carbonates have been removed completely as pendant, nodules or coating to considerable depth, following processes, clay has been dispersed and also trans located to these depths. 10CAR-T cells: Early successes in blood cancer and challenges in solid tumors显示文摘New approaches to cancer immunotherapy have been developed, showing the ability to harness the immune system to treat and eliminate cancer. For many solid tumors, therapy with checkpoint inhibitors has shown promise. For hematologic malignancies, adoptive and engineered cell therapies are being widely developed, using cells such as T lymphocytes, as well as natural killer(NK) cells, dendritic cells, and potentially others. Among these adoptive cell therapies, the most active and advanced therapy involves chimeric antigen receptor(CAR)-T cells, which are T cells in which a chimeric antigen receptor is used to redirect specificity and allow T cell recognition, activation and killing of cancers, such as leukemia and lymphoma. Two autologous CAR-T products have been approved by several health authorities,starting with the U.S. Food and Drug Administration(FDA) in 2017. These products have shown powerful, inducing, long-lasting effects against B cell cancers in many cases. In distinction to the results seen in hematologic malignancies, the field of using CAR-T products against solid tumors is in its infancy. Targeting solid tumors and trafficking CAR-T cells into an immunosuppressive microenvironment are both significant challenges.
