| 1 | Urinary nuclear magnetic resonance spectroscopy of a Bangladeshi cohort with hepatitis-B hepatocellular carcinoma: A biomarker corroboration study显示文摘AIM: To establish if a distinct urinary metabolic profile could be identified in Bangladeshi hepatitis-B hepatocellular carcinoma(HCC) patients compared to cirrhosis patients and controls. METHODS: Urine samples from 42 Bangladeshi patients with HCC(39 patients with hepatitis-B HCC), 47 with cirrhosis on a background of hepatitis B, 46 with chronic hepatitis B, and seven ethnically-matched healthy controls were analyzed using nuclear magnetic resonance(NMR) spectroscopy. A full dietary and medication history was recorded for each subject. The urinary NMR data were analyzed using principal component analysis(PCA) and orthogonal partial leastsquared discriminant analysis(OPLS-DA) techniques. Differences in relative signal levels of the most discriminatory metabolites identified by PCA and OPLSDA were compared between subject groups using an independent samples Kruskal-Wallis one-way analysis of variance(ANOVA) test with all pairwise multiple comparisons. Within the patient subgroups, the MannWhitney U test was used to compare metabolite levels depending on hepatitis B e-antigen(HBe Ag) status and treatment with anti-viral therapy. A BenjaminiHochberg adjustment was applied to acquire the level of significance for multiple testing, with a declared level of statistical significance of P < 0.05.RESULTS: There were significant differences in age(P < 0.001), weight(P < 0.001), and body mass index(P < 0.001) across the four clinical subgroups. Serum alanine aminotransferase(ALT) was significantly higher in the HCC group compared to controls(P < 0.001); serum α-fetoprotein was generally markedly elevated in HCC compared to controls; and serum creatinine levels were significantly reduced in the HCC group compared to the cirrhosis group(P = 0.004). A threefactor PCA scores plot showed clustering of the urinary NMR spectra from the four subgroups. Metabolites that contributed to the discrimination between the subgroups included acetate, creatine, creatinine, dimethyamine(DMA), formate, glycine, hippurate, and trimethylamine-N-oxide(TMAO). A comparison of relative metabolite levels confirmed that carnitine was significantly increased in HCC; and creatinine, hippurate, and TMAO were significantly reduced in HCC compared to the other subgroups. HBe Ag negative patients showed a significant increase in creatinine(P = 0.001) compared to HBe Ag positive patients in the chronic hepatitis B subgroup, whilst HBe Ag negative patients showed a significant decrease in DMA(P = 0.004) in the cirrhosis subgroup compared to HBe Ag positive patients. There were no differences in metabolite levels in HCC patients who did or did not receive antiviral treatment. CONCLUSION: Urinary NMR changes in Bangladeshi HCC were identified, corroborating previous findings from Egypt and West Africa. By setting the specific norms and weight parameters, we present the expressions of the structured normwise, mixed, componentwise condition numbers and the corresponding results for unstructured ones. In addition, we investigate the statistical estimation of condition numbers of LDU factorization using the probabilistic spectral norm estimator and the small-sample statistical condition estimation method, and devise three algorithms. | 6 | Epidemiology and Control of Congo Fever in Sacrificial Animals of Pakistan显示文摘The cases and deaths due to Crimean-Congo haemorrhagic fever(CCHF)virus commonly known as Congo virus(fatality rate 15%)have been reported throughout Pakistan from the last few years especially during religious occasion,Eid-ul-Azha.The annual increase in death rates due to CCHF demonstrate the importance of awareness of Congo fever at academia as well as public level.The symptoms of Congo fever which appear one to nine days after tick bite,include sudden high fever,muscle aches,abdominal pain,headache,dizziness,sore eyes,jaundice,mood swings,confusion,aggression,and sensitivity to light.The other signs include sore throat,joint pain,vomiting,diarrhea,hemorrhages,and bleeding from skin and large intestine.The Infection has been reported in many species of wild as well as domestic animals including hares,cattle,sheep,goats,dogs,mice and hedgehogs.At least 31 species of Hyalomma,Boophilus,Rhipicephalus,Dermacentor(Ixodidae:hard ticks)act as vector of CCHF in which transovarial,transstadial and venereal transmission occurs.The virus attacks the immune system of the host and influences the immune cells.The Congo fever virus can be isolated from blood,plasma and many body tissues(kidneys,liver,spleen,lungs,brain and bone marrow).Mice inoculation,enzyme-linked immunosorbent assay(ELISA),reverse transcription polymerase chain reaction(RT-PCR)can be used for detection of the infection.Furthermore,IgM and IgG antibodies against CCHFV can also be detected and quantified.Education of general public,tick control with acaricides,use of anti-CCHFV immunoglobulin,usage of approved repellents to prevent tick bites,wearing neutral-coloured garments,application of a permethrin spray to the clothing,avoiding tall grasses and shrubs,applying sunscreen,avoiding direct contact with the blood or tissues of animals are the factors for successful prevention of the infection. | Hafiz Muhammad Rizwan Muhammad Sohail Sajid Haider Abbas Muhammad Fiaz Qamar Qaiser Akram Mahvish Maqbool | 2019 | Veterinary Science Research2019,1,2: | 0 |

In vivo engraftment of human peripheral blood stem cells cryopreserved for up to 18 years-implications for clinical transplantation and stability programs显示文摘BACKGROUND Peripheral blood stem cells(PBSC)are commonly cryopreserved awaiting clinical use for hematopoietic stem cell transplant.Long term cryopreservation is commonly defined as five years or longer,and limited data exists regarding how long PBSC can be cryopreserved and retain the ability to successfully engraft.Clinical programs,stem cell banks,and regulatory and accrediting agencies interested in product stability would benefit from such data.Thus,we assessed recovery and colony forming ability of PBSC following long-term cryopreservation as well as their ability to engraft in NOD/SCID/IL-2 Rγnull(NSG)mice.AIM To investigate the in vivo engraftment potential of long-term cryopreserved PBSC units.METHODS PBSC units which were collected and frozen using validated clinical protocols were obtained for research use from the Cellular Therapy Laboratory at Indiana University Health.These units were thawed in the Cellular Therapy Laboratory using clinical standards of practice,and the pre-freeze and post-thaw characteristics of the units were compared.Progenitor function was assessed using standard colony-forming assays.CD34-selected cells were transplanted into immunodeficient mice to assess stem cell function.RESULTS Ten PBSC units with mean of 17 years in cryopreservation(range 13.6-18.3 years)demonstrated a mean total cell recovery of 88%±12%(range 68%-110%)and post-thaw viability of 69%±17%(range 34%-86%).BFU-E growth was shown in 9 of 10 units and CFU-GM growth in 7 of 10 units post-thaw.Immunodeficient mice were transplanted with CD34-selected cells from four randomly chosen PBSC units.All mice demonstrated long-term engraftment at 12 wk with mean34%±24%human CD45+cells,and differentiation with presence of human CD19+,CD3+and CD33+cells.Harvested bone marrow from all mice demonstrated growth of erythroid and myeloid colonies.CONCLUSION We demonstrated engraftment of clinically-collected and thawed PBSC following cryopreservation up to 18 years in NSG mice,signifying likely successful clinical transplantation of PBSC following long-term cryopreservation. | John Underwood Mahvish Rahim Carijo West Rebecca Britton Elaine Skipworth Vicki Graves Steven Sexton Hillary Harris Dave Schwering Anthony Sinn Karen E Pollok Kent A Robertson W Scott Goebel Kerry M Hege | 2020 | World Journal of Stem Cells2020,12,5: | 0 |
