| 1 | Liver venous deprivation versus portal vein embolization before major hepatectomy:future liver remnant volumetric and functional changes显示文摘Background:We previously showed that embolization of portal inflow and hepatic vein(HV)outflow(liver venous deprivation,LVD)promotes future liver remnant(FLR)volume(FLR-V)and function(FLR-F)gain.Here,we compared FLR-V and FLR-F changes after portal vein embolization(PVE)and LVD.Methods:This study included all patients referred for liver preparation before major hepatectomy over 26 months.Exclusion criteria were:unavailable baseline/follow-up imaging,cirrhosis,Klatskin tumor,two-stage hepatectomy.99mTc-mebrofenin SPECT-CT was performed at baseline and at day 7,14 and 21 after PVE or LVD.FLR-V and FLR-F variations were compared using multivariate generalized linear mixed models(joint modelling)with/without missing data imputation.Results:Baseline FLR-F was lower in the LVD(n=29)than PVE group(n=22)(P<0.001).Technical success was 100%in both groups without any major complication.Changes in FLR-V at day 14 and 21(+14.2%vs.+50%,P=0.002;and+18.6%vs.+52.6%,P=0.001),and in FLR-F at day 7,14 and 21(+23.1%vs.+54.3%,P=0.02;+17.6%vs.+56.1%,P=0.006;and+29.8%vs.+63.9%,P<0.001)differed between PVE and LVD group.LVD(P=0.009),age(P=0.027)and baseline FLR-V(P=0.001)independently predicted FLR-V variations,whereas only LVD(P=0.01)predicted FLR-F changes.After missing data handling,LVD remained an independent predictor of FLR-V and FLR-F variations.Conclusions:LVD is safe and provides greater FLR-V and FLR-F increase than PVE.These results are now evaluated in the HYPERLIV-01 multicenter randomized trial. | Boris Guiu François Quenet Fabrizio Panaro Lauranne Piron Christophe Cassinotto Astrid Herrerro François-Régis Souche Margaux Hermida Marie-Ange Pierredon-Foulongne Ali Belgour Serge Aho-Glele Emmanuel Deshayes | 2020 | Hepatobiliary Surgery and Nutrition2020，9，5： | 10 |

| 5 | Loss of Notch signaling in skeletal stem cells enhances bone formation with aging显示文摘Skeletal stem and progenitor cells(SSPCs) perform bone maintenance and repair. With age, they produce fewer osteoblasts and more adipocytes leading to a loss of skeletal integrity. The molecular mechanisms that underlie this detrimental transformation are largely unknown. Single-cell RNA sequencing revealed that Notch signaling becomes elevated in SSPCs during aging. To examine the role of increased Notch activity, we deleted Nicastrin, an essential Notch pathway component, in SSPCs in vivo. Middle-aged conditional knockout mice displayed elevated SSPC osteo-lineage gene expression, increased trabecular bone mass, reduced bone marrow adiposity, and enhanced bone repair. Thus, Notch regulates SSPC cell fate decisions, and moderating Notch signaling ameliorates the skeletal aging phenotype, increasing bone mass even beyond that of young mice. | 10 | Soil chemical fertility change over four decades in the Morvan Mountains and influence of tree species (Burgundy, France)显示文摘Background: Intensive silvicultural practices and the planting of monospecific forests of coniferous, that are more productive compared to hardwoods may threaten over the mid to long-term the sustainability of soil chemical fertility of forest ecosystems, and are a major concern for forest managers and policy.Methods: We investigated the tree species effect(Quercus sessiliflora Smith, Fagus sylvatica L., Picea abies Karst.,Pseudotsuga menziesii Mirb. Franco., Abies nordmanniana Spach. and Pinus nigra Arn. ssp. laricio Poiret var. corsicana) on the change over time of soil chemical properties and nutrient pool sizes in the mineral and organic layers of the soil during the 45 years after the plantation of the Breuil-Chenue common garden experiment(Burgundy,France). The organic and mineral soil layers down to 70-cm depth were sampled in the different monospecific plots in 1974, 2001 and 2019.Results: Exchangeable Ca and Mg pools and soil pH increased on average over time in the 0–70 cm soil profile in most stands. However, in the topsoil layers(0–15 cm), the decrease of pH, the increase of exchangeable acidity over time under the coniferous stands and the decrease of exchangeable K pools in most stands highlighted that soil acidification is still on-going at Breuil-Chenue site but the intensity of this process depends on the tree species.Indeed, three groups of species could be distinguished: i) Nordmann fir(Abies nordmanniana Spach.)/Norway spruce(Picea abies Karst.) where acidolysis and chelation occurred, resulting in the most pronounced pH decrease in the topsoil, ii) Douglas fir(Pseudotsuga menziesii Mirb. Franco.)/Laricio pine(Pinus nigra Arn. ssp. laricio Poiret var. corsicana) where acidification caused by elevated nitrification rates is probably currently compensated by larger weathering and/or atmospheric depositions fluxes, and iii) oak(Quercus sessiliflora Smith)/beech(Fagus sylvatica L.) where soil acidification was less intense. Counterintuitively, soil acidification at Breuil-Chenue site resulted in an increase in soil CEC which limited the loss of nutrient cations. | 16 | Added value of molecular karyotype in childhood acute lymphoblastic leukemia显示文摘Background:Thanks to an improved therapeutic regimen in childhood B-cell precursor acute lymphoblastic leukemia(BCP-ALL),5 year-overall survival now exceeds 90%.Unfortunately,the 25%of children who relapse have an initial poor prognosis,potentially driven by pre-existing or emerging molecular anomalies.The latter are initially and essentially identified by cytogenetics.However,some subtle alterations are not visible through karyotyping.Methods:Single nucleotide polymorphisms(SNP)array is an alternative way of chromosomal analysis allowing for a more in-depth evaluation of chromosomal modifications such as the assessment of copy number alterations(CNA)and loss of heterozygosity(LOH).This method was applied here in retrospective diagnosis/relapse paired samples from seven children with BCP-ALL and in a prospective cohort of 38 newly diagnosed childhood cases.Results:In the matched study,compared to the initial karyotype,SNP array analysis reclassified two patients as poor prognosis cases.Modulation during relapse was seen for 4 CNA and 0.9 LOH.In the prospective study,SNP reclassified the 10 patients with intermediate karyotype as 7 good prognosis and 3 poor prognosis.Ultimately,in all the children tested,SNP array allowed to identify additional anomalies compared to conventional karyotype,refine its prognostic value and identify some druggable anomalies that could be used for precision medicine.Overall,the anomalies detected could be segregated in four groups respectively involved in B-cell development,cell proliferation,transcription and molecular pathways.Conclusion:SNP therefore appears to be a method of choice in the integrated diagnosis of BCP ALL,especially for patients initially classified as intermediate prognosis.This complementary method of both cytogenetics and high throughput sequencing allows to obtain further classified information and can be useful in case of failure of these techniques. | Margaux Camuset Baptiste Le Calvez Olivier Theisen Catherine Godon Audrey Grain Caroline Thomas Marie-Laure Couec Marie C.Béné Fanny Rialland Marion Eveillard | 2023 | Cancer Innovation2023，2，6： | 0 |
