1Animal models of acute gastric mucosal injury: Macroscopic and microscopic evaluation显示文摘Acute gastric mucosal injury is a common disorder of the gastrointestinal tract and the search for new therapeutics is ongoing. The aim of this study is to update and expand the information related to the most widely used rat models of acute gastric ulcer, the ethanol‐induced ulcer and the indomethacin‐induced ulcer. These two models are compared in terms of macroscopic and microscopic features. Experimentally, ethanol was given orally in a single dose and indomethacin was subcutaneously injected into male Wistar rats. After ulcerative challenges, the stomachs were removed and visually inspected. Anti‐ulcerative drugs were used to validate the models. Histological analysis of the stomachs determined the microscopic score. The methodology used for model evaluation applied to macroscopic and microscopic gastric lesions. With these methods it was possible to induce lesions in the gastric mucosa. Microscopic evaluation permitted assessment of the inflammatory and apoptotic impact in the mucosa not observable by macroscopic evaluation. Groups of animals were treated with two standard drugs: sulcralfate suspension or lansoprazole solution. Both drugs reduced macroscopic and microscopic lesions, particularly the hemorrhagic ones. Both models induced acute gastric mucosal injury and no single evaluation method can address all the aspects of the pathology of gastric lesions. As a complement to macroscopic evaluation, microscopy appears to be a relevant tool to selectively identify specific aspects of the development of mucosal injury, quantify the extent of lesions, and contribute to an appropriate interpretation of results. 2Quantitative Study on Nonmetallic Inclusion Particles in Steels by Automatic Image Analysis With Extreme Values Method显示文摘The presence of nonmetallic inclusion particles which appear during steelmaking process is harmful to the properties of steels,which is mainly as a function of some aspects such as size,volume fraction,shape,and distribution of these particles.The automatic image analysis technique is one of the most important tools for the quantitative determination of these parameters.The classical Student approach and the Extreme Values Method(EVM) were used for the inclusion size and shape determination and the evaluation of distance between the inclusion particles.The results thus obtained indicated that there were significant differences in the characteristics of the inclusion particles in the analyzed products.Both methods achieved results with some differences,indicating that EVM could be used as a faster and more reliable statistical methodology.Cássio Barbosa José Brant de Campos Jneo Lopes do Nascimento Iêda Maria Vieira Caminha 2009Journal of Iron and Steel Research(International)2009,16,4:7
3Helicobacter pylori and EBV in gastric carcinomas:Methylation status and microsatellite instability显示文摘AIM:To verify the methylation status of CDH1, DAPK, COX2, hMLH1 and CDKN2A genes and to evaluate their association with Helicobacter pylori (H. pylori)-cagA+ and Epstein Barr virus (EBV) infections in gastric adenocarcinomas.METHODS: Methylation-specific PCR (MSP) assay was performed in 89 primary gastric carcinomas (intestinal and diffuse types). Microsatellite instability (MSI) analysis was performed using the BAT26 primer set and PCR products were analyzed with the ABI PRISM 3100 Genetic Analyzer using Genescan 3.7 software (Applied Biosystems). Detection of H. pylori and genotyping were performed by PCR, using specifi c primers for ureaseC and cagA genes. The presence of EBV was assessed by in situ hybridization. Statistical analyses were performed using the χ2 or Fisher's exact test.RESULTS: The most frequent hypermethylated gene was COX-2 (63.5%) followed by DAPK (55.7%), CDH1 (51%), CDKN2A (36%) and hMLH1 (30.3%). Intestinal and diffuse adenocarcinomas showed different methylation profiles and there was an association between methylation of E-CDH1 and H. pylori-cagA+ in the intestinal adenocarcinoma type. MSI was correlated with hMLH1 methylation. 4Brazilian biosafety law and the new breeding technologies显示文摘Globally,the area of land cultivated with genetically modified(GM)crops has increased a thousand-fold over the last two decades.Although this technology has become important for food production,the regulatory frameworks that underpin these outcomes are based on a list of requirements for a risk assessment that differ from country to country.In recent years,policymakers have had the opportunity to learn from the controversies over transgenics to create effective regulatory milestones for emerging technologies,allowing them to reach their potential for a more sustainable agriculture,ensuring food security.In Brazil,Law No.11.105 of 24 March 2005 established a framework with four main organizations responsible for risk assessment and management.However,most of new breeding technologies did not exist at that time and were not considered in this law.In2016,Normative Resolution No.16 of the National Biosafety Technical Commission(CTNBio)was established to address this gap based on the evaluation of the products obtained through these techniques(termed Innovative Precision Improvement Techniques in the resolution),in a case-by-case consultation system.Briefly,if the product is designated to be a GM,the developer will have to go through the biosafety requirements and will be approved only after CTNBio risk assessment.If the product is designated not to be GM(for the purposes of the legislation),then it can be registered using the existing procedures.Currently,152 GM products are commercially approved in Brazil.In 2018,CTNBio assessed the first consultation on commercial release of plants generated using the new breeding technologies and has subsequently approved six products.It is expected that many institutions would be able to participate in Brazilian and world markets,developing and introducing new biotechnological solutions and products through a more sustainable approach and without facing public disapproval,a common issue for GM crops.Alexandre Lima NEPOMUCENO Renata FUGANTI-PAGLIARINI Maria Sueli Soares FELIPE Hugo Bruno Correa MOLINARI Edivaldo Domingues VELINI Eduardo Romano de Campos PINTO Maria Lucia Zaidan DAGLI Galdino ANDRADE FILHO Patricia Machado Bueno FERNANDES 2020Frontiers of Agricultural Science and Engineering2020,7,2:5
5Colonic duplication in an adult mimicking a tumor of pancreas显示文摘Duplications of the alimentary tract are uncommon congenital malformations that can present diagnostic difficulties. We report a rare case of a cystic colonic duplication in a female adult. Preoperative investigations were suggestive of pancreatic tumor. The diagnosis was established based on the histopathological examination of the resected specimen. 7Somatic Embryogenesis in Iberian Grapevine (Vitis vinifera) Cultivars Using Carpels as Initial Explants: Protocol Establishment and Histological Evaluation显示文摘Trincadeira and Aragonez are two important grapevine cultivars in the Iberian Peninsula, used for high quality red wines production. Both cultivars are strongly affected by fungal diseases, with consequent high loses on plant productivity and fruit quality. A successful protocol for plant regeneration via somatic embryogenesis (SE) was established for both cultivars allowing further plant improvement based on gene transfer technology. Several factors were evaluated during the three different phases which characterize an SE plant regeneration protocol. The culture room temperature during the induction phase, a parameter usually accepted as standard by most researchers, proved in these trials to significantly affect the embryo induction rates. Concerning embryo conversion, it was specially affected by the embryo developmental stage, by the intensity and duration of the chilling treatment and by the supplementation of conversion culture medium with activated charcoal. The responses obtained, both for induction and conversion, proved to be highly genotype dependent. 8Characterization of goat whey proteins and their bioactivity and toxicity assay显示文摘Goat whey is an industry-discarded dairy by-product that has interesting nutritional value and a nutrient composition with important functional potential,where proteins have a high biological value and play an important role in the organism’s physiology.Therefore,the aim of this study was to identify,quantify,concentrate and investigate the capacity of goat whey proteins to perform different biological activities and to evaluate their acute toxicity.Concentrated proteins were quantified and identified by Bradford,Kjehdal and SDS-PAGE methods.Antibacterial activity was performed against Listeria monocytogenes,Salmonella spp.,Staphylococcus aureus,Pseudomonas aeruginosa and Escherichia coli microorganisms and showed MIC ranging from 15 to 120μg mL-1,with the highest inhibition for L.monocytogenes growth.Antiproliferative activity was performed against human malignant melanoma cells and their proliferation inhibition was up to 66.8%within 72 h of exposure.The highest antioxidant capacity was obtained at concentration 2 mg that showed 29.69%of inhibition of DPPH radical oxidation.The acute toxicity test was performed using Artemia franciscana larvae in which whey proteins were able to preserve larval survival throughout exposure.Thus,we can conclude that goat’s whey has proteins that can play a positive effect on health,with antimicrobial activity and without toxicity.The present study highlights the great potential of goat’s whey to be developed as a safe biopreservative or as a functional ingredient in the food industry.Maria Isabel Ferreira Campos Paula Perazzo de Souza Barbosa Laura Junqueira Camargo Luciano Da Silva Pinto Bianca Mataribu Catarina Serr˜ao Luis Fernando Marques-Santos Jos´e Hon´orio Lopes Julia Mariano Caju de Oliveira Carlos Alberto de Almeida Gadelha Tatiane Santi-Gadelha 2022Food Bioscience2022,46,2:2

10Parathyroid ultrasonography and bone metabolic profile of patients on dialysis with hyperparathyroidism显示文摘AIM To evaluate the parathyroid ultrasonography and define parameters that can predict poor response to treatment in patients with secondary hyperparathyroidism due to renal failure.METHODS This cohort study evaluated 85 patients with chronic kidney disease stage V with parathyroid hormone levels above 800 pg/mL. All patients underwent ultrasonography of the parathyroids and the following parameters were analyzed: Demographic characteristics(etiology of chronic kidney disease, gender, age, dialysis vintage, vascular access, use of vitamin D), laboratory(calcium, phosphorus, parathyroid hormone, alkaline phosphatase, bone alkaline phosphatase), and the occurrence of bone changes, cardiovascular events and death. The χ~2 test were used to compare proportions or the Fisher exact test for small sample frequencies. Student t-test was used to detect differences between the two groups regarding continuous variables.RESULTS Fifty-three patients(66.4%) had parathyroid nodules with higher levels of parathyroid hormone, calcium and phosphorus. Sixteen patients underwent parathyroidectomy and had higher levels of phosphorus and calcium × phosphorus product(P = 0.03 and P = 0.006, respectively). They also had lower mortality(32% vs 68%, P = 0.01) and lower incidence of cardiovascular or cerebrovascular events(27% vs 73%, P = 0.02). Calcium × phosphorus product above 55 mg^2/dL^2 [RR 1.48(1.06, 2.08), P = 0.03], presence of vascular calcification [1.33(1.01, 1.76), P = 0.015] and previous occurrence of vascular events [RR 2.25(1.27, 3.98), P < 0.001] were risk factors for mortality in this population. 11Association between the CYBA and NOX4 genes of NADPH oxidase and its relationship with metabolic syndrome in non-alcoholic fatty liver disease in Brazilian population显示文摘Background: Oxidative stress has been implicated in the progression of severe forms of non-alcoholic fatty liver disease(NAFLD). NADPH oxidase produces reactive oxygen species. In the present study, we investigated for the first time two single nucleotide polymorphisms(SNPs) in the regulatory region of genes encoding NADPH oxidase 4(NOX4) and p22 phox(CYBA) in NAFLD.Methods: A total of 207 biopsy-proven NAFLD patients [simple steatosis(n = 27); nonalcoholic steatohepatitis(NASH)(n = 180)] were evaluated. Genomic DNA was extracted from peripheral blood cells, and polymorphisms in CYBA(unregistered) and NOX4(rs3017887) were determined by direct sequencing of PCR.Results: Associations of CYBA-675 T/A with high-density lipoprotein(HDL)(TT vs TA vs AA; P < 0.01) and triglycerides(TGL)(TT vs XA; P < 0.01) were observed only in NASH patients. For polymorphisms in the NOX4 gene, NOX4(rs3017887) CA + AA genotypes was significant associated with alanine aminotransferase(ALT)(CA + AA vs CC; P = 0.02). However, there was no association of SNPs in the CYBA and NOX4 genes encoding the NADPH oxidase system proteins and the presence of NASH. Regarding the clinical results, it was observed that the most advanced degrees of fibrosis occurred in patients diagnosed with type 2 diabetes mellitus(66.9% vs 37.5%, P < 0.01) and those who were more obese(32.2 vs 29.0 kg/m2,P < 0.01). In addition, serum glucose and insulin levels increased significantly in the presence of NASH.Conclusions: There were associations between the presence of the allele A in the NOX4 SNP and a higher concentration of ALT in the NAFLD population; between the presence of the AA genotype in the polymorphism of the CYBA-675 T/A CYBA gene and a higher level of TGL and lower HDL in NASH patients. 13One-week dual therapy with ranitidine bismuth citrate and clarithromycin for the treatment of Helicobacter pylori infection in Brazilian patients with peptic ulcer显示文摘AIM: To assess the efficacy and safety of ranitidine bismuth citrate plus clarithromycin given for 1 wk in Brazilian patients with peptic ulcer.METHODS: One hundred and twenty patients with peptic ulcer were randomized in two treatment groups: (1) 1-wk regimen consisting of ranitidine bismuth citrate 400 mg b.i.d. with clarithromycin 500 mg b.i.d. or (2) 2-wk regimen of the same treatment. Eradication of the infection was considered when both the histologic examination and the urease test were negative for the infection 3 mo after treatment.RESULTS: By intention to treat analysis, Helicobacter pylori (H pylori) was eradicated in 73% and 76% of patients, respectively treated for 1 or 2 wk (P>0.05). By per protocol analysis, the eradication rates were 80% and 83%,respectively, in patients treated for 1 or 2 wk (P>0.05). Nine patients (8.2%) reported minor side effects. 18High throughput sequencing unravels tomato-pathogen interactions towards a sustainable plant breeding显示文摘Tomato(Solanum lycopersicum)is one of the most economically important vegetables throughout the world.It is one of the best studied cultivated dicotyledonous plants,often used as a model system for plant research into classical genetics,cytogenetics,molecular genetics,and molecular biology.Tomato plants are affected by different pathogens such as viruses,viroids,fungi,oomycetes,bacteria,and nematodes,that reduce yield and affect product quality.The study of tomato as a plant-pathogen system helps to accelerate the discovery and understanding of the molecular mechanisms underlying disease resistance and offers the opportunity of improving the yield and quality of their edible products.The use of functional genomics has contributed to this purpose through both traditional and recently developed techniques,that allow the identification of plant key functional genes in susceptible and resistant responses,and the understanding of the molecular basis of compatible interactions during pathogen attack.Next-generation sequencing technologies(NGS),which produce massive quantities of sequencing data,have greatly accelerated research in biological sciences and offer great opportunities to better understand the molecular networks of plant–pathogen interactions.In this review,we summarize important research that used high-throughput RNA-seq technology to obtain transcriptome changes in tomato plants in response to a wide range of pathogens such as viruses,fungi,bacteria,oomycetes,and nematodes.These findings will facilitate genetic engineering efforts to incorporate new sources of resistance in tomato for protection against pathogens and are of major importance for sustainable plant-disease management,namely the ones relying on the plant’s innate immune mechanisms in view of plant breeding.Maria Doroteia Campos Maria do Rosario Felix Mariana Patanita Patrick Materatski Carla Varanda 2021Horticulture Research2021,8,1:1

20Serum levels of the novel adipokine isthmin-1 are associated with obesity in pubertal boys显示文摘Objectives To evaluate whether there is an association between the serum levels of the novel insulin-like adipokine isthmin-1(ISM1)and obesity-related phenotypes in a population of Spanish children and to investigate the plausible molecular alterations behind the alteration of the serum levels of this protein in children with obesity.Methods The study population is a sub-cohort of the PUBMEP research project,consisting of a cross-sectional population of 119 pubertal children with overweight(17 boys,19 girls),obesity(20 boys,25 girls),and normal weight(17 boys,21 girls).All subjects were classified into experimental groups according to their sex,obesity,and insulin resistance(IR)status.They were counted anthropometry,glucose and lipid metabolism,inflammation and cardiovascular biomarkers as well as isthmin-1(ISM1)serum levels.This population was intended as a discovery population to elucidate the relationship between obesity and ISM1 levels in children.Furthermore,the study population had blood whole-genome DNA methylation examined,allowing deepening into the obesity–ISM1 molecular relationship.Results Higher serum ISM1 levels were observed in boys with obesity than in normal weight(P=0.004)and overweight(P=0.007)boys.ISM1 serum levels were positively associated with body mass index(BMI)Z-score(P=0.005)and fat mass(P=0.058)and negatively associated with myeloperoxidase(MPO)(P=0.043)in boys.Although we did not find associations between ISM1 serum levels and metabolic outcomes in girls,which may indicate a putative sexual dimorphism,fat mass was positively associated in all children,including boys and girls(P=0.011).DNA methylation levels in two-enhancer-related CpG sites of ISM1(cg03304641 and cg14269097)were associated with serum levels of ISM1 in children.Conclusions ISM1 is associated with obesity in boys at the pubertal stage,elucidating how this protein might be of special relevance as a new biomarker of obesity in children.Further studies including a longitudinal design during puberty are needed.Francisco Javier Ruiz‑Ojeda Augusto Anguita‑Ruiz Maria C.Rico Rosaura Leis Gloria Bueno Luis A.Moreno Mercedes Gil‑Campos Ángel Gil Concepción M.Aguilera 2023World Journal of Pediatrics2023,19,9:1
