6Association between homeobox protein transcript antisense intergenic ribonucleic acid genetic polymorphisms and cholangiocarcinoma显示文摘BACKGROUND Cholangiocarcinoma(CCA)represents a rare but highly aggressive malignancy that is often challenging to diagnose,especially in early stages.The role of existing tumor biomarkers for CCA diagnosis,remains controversial due to their low sensitivity and specificity.Increasing evidence has implicated long non-coding ribonucleic acid polymorphisms with cancer susceptibility in a variety of tumor types.The association between long non-coding ribonucleic acid homeobox protein transcript antisense intergenic ribonucleic acid(HOTAIR)polymorphisms and CCA risk has not been reported yet.AIM To investigate the influence of HOTAIR variants on the risk of CCA development.METHODS We conducted a case-control study in which three HOTAIR single nucleotide polymorphisms(rs920778,rs4759314 and rs7958904)were genotyped in a Greek cohort.Our study population included 122 CCA patients(80 males and 42 females)and 165 healthy controls.The polymorphisms under investigation were examined in peripheral blood samples.RESULTS HOTAIR rs4759314 AG and GG genotypes were associated with a significantly increased CCA risk[P=0.004,odds ratio:3.13;95%confidence interval:1.65-5.91 and P=0.005,odds ratio:12.31;95%confidence interval:1.48-101.87,respectively].However,no significant associations of HOTAIR rs920778,and rs7958904 were detected.Similarly,we found no significant associations between rs4759314 AA genotype and CCA susceptibility.CONCLUSION HOTAIR rs4759314 AG and GG genotypes may be implicated with CCA development and may serve as a potential diagnostic biomarker.Dimitra Ioanna Lampropoulou Konstantinos Laschos Gerasimos Aravantinos Konstantinos Georgiou Konstantinos Papiris George Theodoropoulos Maria Gazouli Dimitrios Filippou 2021World Journal of Clinical Cases2021,9,8:0

8Parents of Children with Autistic Spectrum Disorder (ASD) as Co-therapists: The Therapists’ View显示文摘Studies on the evaluation by therapists of parental behavior towards their children with Autistic Spectrum Disorder(ASD)and towards the therapists of their children are scarce.They are necessary,however,for enabling parents to become co-therapists.The present study’s purpose was the evaluation by therapists of the behavior of parents towards their children,of their relationship to therapists,and therapy outcome.The sample consisted of 178 parents of 89 children(72 boys)with ASD,who underwent intensive early intervention at a day centre for developmental disabilities.The professional team completed a questionnaire,separately for the mother and father,evaluating the parental attitude towards their children and towards the therapists.The behavior of parents was less satisfactory than expected.The fathers had difficulties in understanding their child’s problems,and had unrealistic expectations;mothers’behavior towards their children and therapists was better than the fathers’.Mothers had difficulty mostly in the management of the child’s behavior,and did not do well with feeding.It might be difficult for every parent to become co-therapist.Understanding the child’s difficulties by the mother,adequate handling of feeding and homework,were statistically significant in the good outcome of therapy.Angelique Kotsopoulou Ermioni Papadaki Irene Florou Antigoni Troupou Dimitra Kolosioni Anastasia Georgiou Maria Gyftogianni Aikaterini Gyftogianni Athina Iliopoulou Eleonora Paleodimou Marigo Sakellari Efthimia Koumanioti 2021Psychology Research2021,11,2:0
9Gut microbiome:Linking together obesity,bariatric surgery and associated clinical outcomes under a single focus显示文摘Obesity is increasingly prevalent in the post-industrial era,with increased mortality rates.The gut microbiota has a central role in immunological,nutritional and metabolism mediated functions,and due to its multiplexity,it is considered an independent organ.Modern high-throughput sequencing techniques have allowed phylogenetic exploration and quantitative analyses of gut microbiome and improved our current understanding of the gut microbiota in health and disease.Its role in obesity and its changes following bariatric surgery have been highlighted in several studies.According to current literature,obesity is linked to a particular microbiota profile that grants the host an augmented potential for calorie release,while limited diversity of gut microbiome has also been observed.Moreover,bariatric surgery procedures represent effective interventions for sustained weight loss and restore a healthier microbiota,contributing to the observed fat mass reduction and lean mass increase.However,newer evidence has shown that gut microbiota is only partially recovered following bariatric surgery.Moreover,several targets including FGF15/19(a gutderived peptide),could be responsible for the favorable metabolic changes of bariatric surgery.More randomized controlled trials and larger prospective studies that include well-defined cohorts are required to better identify associations between gut microbiota,obesity,and bariatric surgery.Konstantinos Georgiou Nikolay A Belev Tilemachos Koutouratsas Hector Katifelis Maria Gazouli 2022World Journal of Gastrointestinal Pathophysiology2022,13,3:0
10Mesenchymal stromal cell delivery as a potential therapeutic strategy against COVID-19:Promising evidence from in vitro results显示文摘BACKGROUND Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)is responsible for the coronavirus disease 2019(COVID-19)pandemic,which was initiated in December 2019.COVID-19 is characterized by a low mortality rate(<6%);however,this percentage is higher in elderly people and patients with underlying disorders.COVID-19 is characterized by mild to severe outcomes.Currently,several therapeutic strategies are evaluated,such as the use of anti-viral drugs,prophylactic treatment,monoclonal antibodies,and vaccination.Advanced cellular therapies are also investigated,thus representing an additional therapeutic tool for clinicians.Mesenchymal stromal cells(MSCs),which are known for their immunoregulatory properties,may halt the induced cytokine release syndrome mediated by SARS-CoV-2,and can be considered as a potential stem cell therapy.AIM To evaluate the immunoregulatory properties of MSCs,upon stimulation with COVID-19 patient serum.METHODS MSCs derived from the human Wharton’s Jelly(WJ)tissue and bone marrow(BM)were isolated,cryopreserved,expanded,and defined according to the criteria outlined by the International Society for Cellular Therapies.Then,WJ and BM-MSCs were stimulated with a culture medium containing 15%COVID-19 patient serum,1%penicillin-streptomycin,and 1%L-glutamine for 48 h.The quantification of interleukin(IL)-1 receptor a(Ra),IL-6,IL-10,IL-13,transforming growth factor(TGF)-β1,vascular endothelial growth factor(VEGF)-a,fibroblast growth factor(FGF),platelet-derived growth factor(PDGF),and indoleamine-2,3-dioxygenase(IDO)was performed using commercial ELISA kits.The expression of HLA-G1,G5,and G7 was evaluated in unstimulated and stimulated WJ and BMMSCs.Finally,the interactions between MSCs and patients’macrophages were established using co-culture experiments.RESULTS Thawed WJ and BM-MSCs exhibited a spindle-shaped morphology,successfully differentiated to“osteocytes”,“adipocytes”,and“chondrocytes”,and in flow cytometric analysis were characterized by positivity for CD73,CD90,and CD105(>95%)and negativity for CD34,CD45,and HLA-DR(<2%).Moreover,stimulated WJ and BM-MSCs were characterized by increased cytoplasmic granulation,in comparison to unstimulated cells.The HLA-G isoforms(G1,G5,and G7)were successfully expressed by the unstimulated and stimulated WJ-MSCs.On the other hand,only weak expression of HLA-G1 was identified in BM-MSCs.Stimulated MSCs secreted high levels of IL-1Ra,IL-6,IL-10,IL-13,TGF-β1,FGF,VEGF,PDGF,and IDO in comparison to unstimulated cells(P<0.05)after 12 and 24 h.Finally,macrophages derived from COVID-19 patients successfully adapted the M2 phenotype after co-culturing with stimulated WJ and BM-MSCs.CONCLUSION WJ and BM-MSCs successfully produced high levels of immunoregulatory agents,which may efficiently modulate the over-activated immune responses of critically ill COVID-19 patients.Panagiotis Mallis Theofanis Chatzistamatiou Zetta Dimou Eirini-Faidra Sarri Eleni Georgiou Maria Salagianni Vasiliki Triantafyllia Evangelos Andreakos Catherine Stavropoulos-Giokas Efstathios Michalopoulos 2022World Journal of Biological Chemistry2022,13,2:0
