| 1 | Seasonal Rainfall Forecasts for the Yangtze River Basin of China in Summer 2019 from an Improved Climate Service显示文摘Rainfall forecasts for the summer monsoon season in the Yangtze River basin(YRB) allow decision-makers to plan for possible flooding, which can affect the lives and livelihoods of millions of people. A trial climate service was developed in 2016, producing a prototype seasonal forecast product for use by stakeholders in the region, based on rainfall forecasts directly from a dynamical model. Here, we describe an improved service based on a simple statistical downscaling approach. Through using dynamical forecast of an East Asian summer monsoon(EASM) index, seasonal mean rainfall for the upper and middle/lower reaches of YRB can be forecast separately by use of the statistical downscaling, with significant skills for lead times of up to at least three months. The skill in different sub-basin regions of YRB varies with the target season. The rainfall forecast skill in the middle/lower reaches of YRB is significant in May–June–July(MJJ), and the forecast skill for rainfall in the upper reaches of YRB is significant in June–July–August(JJA). The mean rainfall for the basin as a whole can be skillfully forecast in both MJJ and JJA. The forecasts issued in 2019 gave good guidance for the enhanced rainfall in the MJJ period and the near-average conditions in JJA. | 3 | Colorectal liver metastases: Current management and future perspectives显示文摘The liver is the commonest site of metastatic disease for patients with colorectal cancer,with at least 25%developing colorectal liver metastases(CRLM)during the course of their illness.The management of CRLM has evolved into a complex field requiring input from experienced members of a multi-disciplinary team involving radiology(cross sectional,nuclear medicine and interventional),Oncology,Liver surgery,Colorectal surgery,and Histopathology.Patient management is based on assessment of sophisticated clinical,radiological and biomarker information.Despite incomplete evidence in this very heterogeneous patient group,maximising resection of CRLM using all available techniques remains a key objective and provides the best chance of long-term survival and cure.To this end,liver resection is maximised by the use of downsizing chemotherapy,optimisation of liver remnant by portal vein embolization,associating liver partition and portal vein ligation for staged hepatectomy,and combining resection with ablation,in the context of improvements in the functional assessment of the future remnant liver.Liver resection may safely be carried out laparoscopically or open,and synchronously with,or before,colorectal surgery in selected patients.For unresectable patients,treatment options including systemic chemotherapy,targeted biological agents,intraarterial infusion or bead delivered chemotherapy,tumour ablation,stereotactic radiotherapy,and selective internal radiotherapy contribute to improve survival and may convert initially unresectable patients to operability.Currently evolving areas include biomarker characterisation of tumours,the development of novel systemic agents targeting specific oncogenic pathways,and the potential reemergence of radical surgical options such as liver transplantation. | Jack Martin Angelica Petrillo Elizabeth C Smyth Nadeem Shaida Samir Khwaja HK Cheow Adam Duckworth Paula Heister Raaj Praseedom Asif Jah Anita Balakrishnan Simon Harper Siong Liau Vasilis Kosmoliaptsis Emmanuel Huguet | 2020 | World Journal of Clinical Oncology2020,11,10: | 5 |

| 9 | The heterodimeric structure of heterogeneous nuclear ribonucleoprotein C1/C2 dictates 1,25-dihydroxyvitamin D-directed transcriptional events in osteoblasts显示文摘Heterogeneous nuclear ribonucleoprotein(hnRNP) C plays a key role in RNA processing but also exerts a dominant negative effect on responses to 1,25-dihydroxyvitamin D(1,25(OH)2D) by functioning as a vitamin D response element-binding protein(VDRE-BP). hnRNPC acts a tetramer of hnRNPC1(huC1) and hnRNPC2(huC2), and organization of these subunits is critical to in vivo nucleic acid-binding. Overexpression of either huC1 or huC2 in human osteoblasts is sufficient to confer VDRE-BP suppression of 1,25(OH)2D-mediated transcription. However, huC1 or huC2 alone did not suppress 1,25(OH)2D-induced transcription in mouse osteoblastic cells. By contrast, overexpression of huC1 and huC2 in combination or transfection with a bone-specific polycistronic vector using a ‘‘self-cleaving'' 2A peptide to co-express huC1/C2 suppressed1,25D-mediated induction of osteoblast target gene expression. Structural diversity of hnRNPC between human/NWPs and mouse/rat/rabbit/dog was investigated by analysis of sequence variations within the hnRNP CLZ domain. The predicted loss of distal helical function in hnRNPC from lower species provides an explanation for the altered interaction between huC1/C2 and their mouse counterparts.
