| 1 | Random forest machine learning models for interpretable X-ray absorption near-edge structure spectrum-property relationships显示文摘X-ray absorption spectroscopy(XAS)produces a wealth of information about the local structure of materials,but interpretation of spectra often relies on easily accessible trends and prior assumptions about the structure.Recently,researchers have demonstrated that machine learning models can automate this process to predict the coordinating environments of absorbing atoms from their XAS spectra.However,machine learning models are often difficult to interpret,making it challenging to determine when they are valid and whether they are consistent with physical theories.In this work,we present three main advances to the data-driven analysis of XAS spectra:we demonstrate the efficacy of random forests in solving two new property determination tasks(predicting Bader charge and mean nearest neighbor distance),we address how choices in data representation affect model interpretability and accuracy,and we show that multiscale featurization can elucidate the regions and trends in spectra that encode various local properties. | Steven B.Torrisi Matthew R.Carbone Brian A.Rohr Joseph H.Montoya Yang Ha Junko Yano Santosh K.Suram Linda Hung | 2020 | npj Computational Materials2020,,1: | 3 |

| 8 | MFSD7c functions as a transporter of choline at the blood–brain barrier显示文摘Mutations in the orphan transporter MFSD7c(also known as Flvcr2),are linked to Fowler syndrome.Here,we used Mfsd7c knockout(Mfsd7c–/–)mice and cell-based assays to reveal that MFSD7c is a choline transporter at the blood–brain barrier(BBB).We performed comprehensive metabolomics analysis and detected differential changes of metabolites in the brains and livers of Mfsd7c–/–embryos.Particularly,we found that choline-related metabolites were altered in the brains but not in the livers of Mfsd7c–/–embryos.Thus,we hypothesized that MFSD7c regulates the level of choline in the brain.Indeed,expression of human MFSD7c in cells significantly increased choline uptake.Interestingly,we showed that choline uptake by MFSD7c is greatly increased by choline-metabolizing enzymes,leading us to demonstrate that MFSD7c is a facilitative transporter of choline.Furthermore,single-cell patch clamp analysis showed that the import of choline by MFSD7c is electrogenic.Choline transport function of MFSD7c was shown to be conserved in vertebrates,but not in yeasts.We demonstrated that human MFSD7c is a functional ortholog of HNM1,the yeast choline importer.We also showed that several missense mutations identified in patients exhibiting Fowler syndrome had abolished or reduced choline transport activity.Mice lacking Mfsd7c in endothelial cells of the central nervous system suppressed the import of exogenous choline from blood but unexpectedly had increased choline levels in the brain.Stable-isotope tracing study revealed that MFSD7c was required for exporting choline derived from lysophosphatidylcholine in the brain.Collectively,our work identifies MFSD7c as a choline exporter at the BBB and provides a foundation for future work to reveal the disease mechanisms of Fowler syndrome. | Xuan Thi Anh Nguyen Thanh Nha Uyen Le Toan Q.Nguyen Hoa Thi Thuy Ha Anna Artati Nancy C.P.Leong Dat T.Nguyen Pei Yen Lim Adelia Vicanatalita Susanto Qianhui Huang Ling Fam Lo Ngah Leong Isabelle Bonne Angela Lee Jorge L.Granadillo Catherine Gooch Dejie Yu Hua Huang Tuck Wah Soong Matthew Wook Chang Markus R.Wenk Jerzy Adamski Amaury Cazenave-Gassiot Long N.Nguyen | 2024 | Cell Research2024,34,3: | 0 |
| 9 | Simultaneous ramp right heart catheterization and echocardiography in a Reliant Heart left ventricular assist device显示文摘Many clinicians caring for patients with continuous flow left ventricular assist devices(CF-LVAD) use ramp right heart catheterization(RHC) studies to optimize pump speed and also to troubleshoot CF-LVAD malfunction. An investigational device,the Reliant Heart Heart Assist 5(Houston,TX),provides the added benefit of an ultrasonic flow probe on the outflow graft that directly measures flow through the CF-LVAD. We performed a simultaneous ramp RHC and echocardiogram on a patient who received the above CF-LVAD to optimize pump parameters and investigate elevated flow through the CF-LVAD as measured by the flow probe. We found that the patient's hemodynamics were optimized at their baseline pump speed,and that the measured cardiac output via the Fick principle was lower than that measured by the flow probe. Right heart catheterization may be useful to investigate discrepancies between flow measured by a CF-LVAD and a patient's clinical presentation,particularly in investigational devices where little clinical experience exists. | 10 | 母亲颗粒物空气污染暴露与足月出生体重:效应与异质性的多国评估显示文摘[背景]越来越多的证据显示,母亲空气污染暴露与对胎儿生长的不良影响之间存在关联;但是,现有的文献并不一致。[目的]研究9个国家14个中心的数据,量化母亲颗粒物空气污染暴露与足月出生体重和低出生体重(LBW)之间的关联,并探讨此关联中地点特征和暴露评估方法对中心之间异质性的影响。[方法]空气污染与妊娠结局国际合作(ICAPPO)的各中心采用共同的分析方案,对PM10和PM2.5(颗粒物≤10和2.5μm)与足月LBW和连续出生体重进行效应评估。采用meta分析将各个中心(~300万例出生)的效应评估结合起来,并采用meta回归分析在报告的效应评估中,中心特征和暴露评估方法对中心之间异质性的影响。[结果]随机效应meta分析显示,调整母亲的社会经济地位后,在整个妊娠期间,足月LBW与PM10[比值比(OR)=1.03;95%CI:1.01~1.05]和PM2.5[OR=1.10;95%CI:1.03~1.18]每增加10μg/m3呈正相关。在充分调整随机效应的meta分析中,作为一个连续性的结果,PM10暴露每增加10μg/m3与足月出生体重呈负相关(-8.9g;95%CI:-13.2^-4.6 g)。meta回归分析显示,具有较高平均PM2.5水平和PM2.5∶PM10比率的中心,以及采用时间暴露评估(与时空相比)的中心,所报告的关联更强。[结论]在研究人群中,母亲颗粒物污染暴露与足月LBW相关联。有3个地点特征和暴露评估方法学方面的不同导致各个中心报告的关联的差异。 | Payam Dadvand Jennifer Parker Michelle L.
