| 1 | Clostridium difficile infections in China显示文摘Clostridium difficile (C. difficile) infection has become one of the major hospital-associated infections in West-ern countries in the last two decades. However, there is limited information on the status of C. difficile infection inChinese healthcare settings. Given the large and increasing elderly population and the well-recognized problem ofover-prescribing of broad spectrum antibiotics in China, it is critical to understand the epidemiology and potentialrisk factors that may contribute to C. difficile infection in China. A literature review of available published studies,including those in Chinese language-based journals, was conducted. A review of the currently available literaturesuggested the presence of C. difficile infections in China, but also suggested that these infections were not particu-larly endemic. This finding should lead to better designed and greatly expanded studies to provide a more reliableepidemiologically-based conclusion on the actual status of C. difficile infection in China, including the identifica-tion of any associated risk factors. | 3 | Effects of disinhibition on spatiotemporal pattern of neuronal first recruitment in neuronal networks显示文摘The propagation of neuronal activities is a key feature to understanding information processing in networks. The analysis based on first-spikes of bursts in turn plays an important role in the research of neuronal activity propagation. Our focus here is to investigate how spatiotemporal patterns of neuronal first-spikes are affected by disinhibition. Multi-electrode arrays were used to record stimulation-evoked bursts of multiple neurons in randomly cultured neuronal networks. Both the precise timing of and the rank relationships between first-spikes were analyzed. Compared with evoked bursts in the network's native state, the precise first-spike latencies in its dis-inhibited state are more consistent and the propagation of its bursting activities is much faster. | 4 | High performance liquid chromatographic separation of thirteen drugs collected in Chinese Pharmacopoeia 2010(Ch.P2010) on cellulose ramification chiral stationary phase显示文摘The enantiomers separation of thirteen drugs collected in Ch.P2010 was performed on chiral stationary phase of cellulose ramification (chiralpak OD and chiralpak OJ) by high performance liquid chromatographic (HPLC) methods, which included ibuprofen (C1), ketoprofen (C2), nitrendipine (C3), nimodipine (C4), felodipine (C5), omeprazole (C6), praziquantel (C7), propranolol hydrochloride (C8), atenolol (C9), sulpiride (C10), clenbuterol hydrochloride (C11), verapamil hydrochloride (C12), and chlorphenamine maleate (C13). The mobile phase consisted of isopropanol and n-hexane. The detection wavelength was set at 254 nm and the fiow rate was 0.7 mL/min. The enantiomers separation of these thirteen racemates on chiralpak OD column and chiralpak OJ column was studied, while the effects of proportion of organic additives, alcohol displacer and temperature on the separation were studied. And the mechanism of some of racemates was discussed. The results indicated that thirteen chiral drugs could be separated onchiral stationary phase of cellulose ramification in normal phase chromatographic system. The chromatographic retention and resolution of enantiomers could be adjusted by factors including column temperature and the concentration of alcohol displacer and organic alkaline modifier in mobile phase. It was shown that the resolution was improved with reducing concentration of alcohol displacer. When concentration of organic alkaline modifier was 0.2% (v/v), the resolution and the peak shape were fairly good. Most racemates mentioned above had better resolution at column temperature of 25 1C. | 5 | Epstein-Barr virus-encoded latent membrane protein 1 modulates cyclin D1 by c-Jun/Jun B heterodimers显示文摘In our recent studies, we found that LMP1 encoded by Epstein-Barr virus could accelerate the formation of active c-Jun/Jun B heterodimer. We studied the regulation of cyclinD1 by c-Jun/Jun B heterodimers by laser scanning confocal influorescence microscopy, Western blot, luciferase activity assay, super-EMSA and flow cytometry in the Tet-on-LMP1 HNE2 cell line, in which LMP1 expression was regulated by Tet-on system. c-Jun/Jun B heterodimers induced by LMP1 could up regulate cyclin D1 promoter activity and expression. | 12 | Role of Notch expression in premature senescence of murine bone marrow stromal cells显示文摘The aim of the present study was to investigate the role of the Notch signaling pathway in premature senescence of murine bone marrow stromal cells in vitro. The intracellular domain of Notch 1 (ICN) was transfected into cultured murine bone marrow stromal cells by lipofectamine transfection. After three days, the proliferation of transfected cells was measured by MTT assay. Cell cycle distribution was analyzed by flow cytometry. Senescence-associated beta-galactosidase (SA-beta-gal) was measured, and the percentage of positive cells was evaluated by assessing 1000 cells in random fields of view. The expressions of p53 and p21Cip1/Waf1 were analyzed by both RT-PCR and Western blot analysis. The results showed that activation of Notch signaling inhibited proliferation of murine bone marrow stromal cells with induction of G1 arrest, increased the percentage of SA-beta-gal positive cells, and upregulated p53 and p21Cip1/Waf1 mRNA and protein expression levels. | 19 | Mapping and characterization of two relevance networks from SNP and gene levels显示文摘Variations of gene expression and DNA sequence are genetically associated. The goal of this study was to build genetic networks to map from SNPs to gene expressions and to characterize the two different kinds of networks. We employed mutual information to evaluate the strength of SNP-SNP and gene-gene associations based on SNPs identity by descent (IBD) data and differences of gene expressions. We applied the approach to one dataset of Genetics of Gene Expression in Humans, and discovered that both the SNP relevance network and the gene relevance network approximated the scale-free topology. We also found that 12.09% of SNP-SNP interactions matched 24.49% of gene-gene interactions, which was consistent with that of the previous studies.
