1The Genomes of the Allohexaploid Echinochloa crus-galli and Its Progenitors Provide Insights into Polyploidization-Driven Adaptation显示文摘The hexaploid species Echinochloa crus-galli is one of the most detrimental weeds in crop fields,especially in rice paddies.Its evolutionary history is similar to that of bread wheat,arising through polyploidization after hybridization between a tetraploid and a diploid species.In this study,we generated and analyzed high-quality genome sequences of diploid(E.haploclada),tetraploid(E.oryzicola),and hexaploid(E.crus-galli)Echinochloa species.Gene family analysis showed a significant loss of disease-resistance genes such as those encoding NB-ARC domain-containing proteins during Echinochloa polyploidization,contrary to their significant expansionduring wheat polyploidization,suggesting that natural selection might favor reduced investment in resistance in this weed to maximize its growth and reproduction.In contrast to the asymmetric patterns of genome evolution observed in wheat and other crops,no significant differences in selection pressure were detected between the subgenomes in E.oryzicola and E.crus-galli.In addition,distinctive differences in subgenome transcriptome dynamics during hexaploidization were observed between E.crus-galli and bread wheat.Collectively,our study documents genomic mechanisms underlying the adaptation of a major agricultural weed during polyploidization.The genomic and transcriptomic resources of three Echinochloa species and new insights into the polyploidization-driven adaptive evolution would be useful for future breeding cereal crops.Chu-Yu Ye Dongya Wu Lingfeng Mao Lei Jia Jie Qiu Sangting Lao Meihong Chen Bowen Jiang Wei Tang Qiong Peng Lang Pan Lifeng Wang Xiaoxiao Feng Longbiao Guo Chulong Zhang Elizabeth A.Kellogg Kenneth M.Olsen Lianyang Bai Longjiang Fan 2020Molecular Plant2020,13,9:4
2Noninvasive Ultrasound Stimulation of Ventral Tegmental Area Induces Reanimation from General Anaesthesia in Mice显示文摘Evidence in animals suggests that deep brain stimulation or optogenetics can be used for recovery from disorders of consciousness(DOC).However,these treatments require invasive procedures.This report presents a noninvasive strategy to stimulate central nervous system neurons selectively for recovery from DOC in mice.Through the delivery of ultrasound energy to the ventral tegmental area,mice were aroused from an unconscious,anaesthetized state in this study,and this process was controlled by adjusting the ultrasound parameters.The mice in the sham group under isoflurane-induced,continuous,steady-state general anaesthesia did not regain their righting reflex.On insonation,the emergence time from inhaled isoflurane anaesthesia decreased(sham.13.63±0.53 min,ultrasound.1.5±0.19 min,p<0.001).Further,the induction time(sham.12.0±0.6 min,ultrasound.17.88±0.64 min,p<0.001)and the concentration for 50%of the maximal effect(EC50)of isoflurane(sham.0.6%,ultrasound.0.7%)increased.In addition,ultrasound stimulation reduced the recovery time in mice with traumatic brain injury(sham.30.38±1.9 min,ultrasound.7.38±1.02 min,p<0.01).This noninvasive strategy could be used on demand to promote emergence from DOC and may be a potential treatment for such disorders.Tianyuan Bian Wen Meng Meihong Qiu Zhigang Zhong Zhengrong Lin Junjie Zou Yibo Wang Xiaowei Huang Lisheng Xu Tifei Yuan Zhili Huang Lili Niu Long Meng Hairong Zheng 2021Research2021,,1:2
3Long acting carmustine loaded natural extracellular matrix hydrogel for inhibition of glioblastoma recurrence after tumor resection显示文摘Many scientific efforts have been made to penetrate the blood-brain barrier and target glioblastoma cells,but the outcomes have been limited.More attention should be given to local inhibition of recurrence after glioblastoma resection to meet real medical needs.A biodegradable wafer containing the chemotherapeutics carmustine(1,3-bis(2-chloroethyl)-1-nitrosourea,BCNU)was the only local drug delivery system approved for clinical glioblastoma treatment,but with a prolonged survival time of only two months and frequent side effects.In this study,to improve the sustained release and prolonged therapeutic effect of drugs for inhibiting tumor recurrence after tumor resection,both free BCNU and BCNU-poly(lactic-co-glycolic acid)(the ratio of lactic acid groups to glycolic acid groups is 75/25)nanoparticles were simultaneously loaded into natural extracellular matrix hydrogel from pigskin to prepare BCNU gels.The hydrogel was injected into the resection cavity of a glioblastoma tumor immediately after tumor removal in a fully characterized resection rat model.Free drugs were released instantly to kill the residual tumor cells,while drugs in nanoparticles were continuously released to achieve a continuous and effective inhibition of the residual tumor cells for 30 days.These combined actions effectively restricted tumor growth in rats.Thus,this strategy of local drug implantation and delivery may provide a reliable method to inhibit the recurrence of glioblastoma after tumor resection in vivo.Sunhui Chen Qiujun Qiu Dongdong Wang Dejun She Bo Yin Meihong Chai Huining He Dong Nyoung Heo Jianxin Wang 2022Frontiers of Chemical Science and Engineering2022,16,4:1
