| 1 | Matrix metalloproteinase gene expressions might be oxidative stress targets in gastric cancer cell lines显示文摘Objective: Oxidative stress is linked to increased risk of gastric cancer and matrix metalloproteinases (MMPs) are important in the invasion and metastasis of gastric cancer. We aimed to analyze the effect of the accumulation of oxidative stress in the gastric cancer MKN-45 and 23132/87 cells following hydrogen peroxide (H 2 O 2 ) exposure on the expression patterns of MMP-1, MMP-3, MMP-7, MMP-9, MMP-10, MMP- 11, MMP-12, MMP-14, MMP-15, MMP-17, MMP-23, MMP-28, and β-catenin genes. Methods: The mRNA transcripts in the cells were determined by RT-PCR. Following H 2 O 2 exposure, oxidative stress in the viable cells was analyzed by 2',7'-dichlorofluorescein diacetate (DCFH-DA). Caffeic acid phenethyl ester (CAPE) was used to eliminate oxidative stress and the consequence of H 2 O 2 exposure and its removal on the expressions of the genes were evaluated by quantitative real-time PCR. Results: The expressions of MMP-1, MMP-7, MMP-14, MMP-15, MMP-17 and β-catenin in MKN-45 cells and only the expression of MMP-15 in 23132/87 cells were increased. Removal of the oxidative stress resulted in decrease in the expressions of MMP genes of which the expressions were increased after H 2 O 2 exposure. β-catenin, a transcription factor for many genes including MMPs, also displayed decreased levels of expression in both of the cell lines following CAPE treatment. | 3 | Development of Chromium(Ⅲ)-selective Potentiometric Sensor by Using Synthesized Pyrazole Derivative as an lonophore in PVC Matrix and its Applications显示文摘A novel poly(vinyl chloride)membrane potentiometric sensor for chromium(Ⅲ)ions based on the use of 5,5′-(1,4-phenylene)bis(3-(naphthalen-1-yl)-4,5-dihydro-1H-pyrazole-1-carbothioamide)as a neutral ionophore was developed.The optimum composition of the best performing membrane contained ionophore,potassium tetrakis(p-chlorophenyl)borate(KTpClPB),dibutyl phthalate(DBP),and poly(vinyl chloride)(PVC)in the ratio of 5.5:1.5:55:38(mg).The sensor exhibits a working concentration range of 1.0×10^(-5)-1.0×10^(-1 )mol L^(−1) and a detection limit of 1.7×10^(-6) mol L^(−1).The sensor shows good selectivity for chromium(III)ions over a number of cations including alkali,alkaline earth,heavy and transition metals.The response time of the sensor is 8 s.In addition,the developed sensor shows good reusability and stability.The sensor operates in the wide pH range of 5.0-11.0.The sensor could be used as an indicator electrode in the quantification of Cr^(3+)ions by potentiometric titration against ethylenediaminetetraacetic acid(EDTA).Finally,this sensor was successfully used for the determination of chromium(III)in commercial water,purification water and wastewater. | Ömer Isildak OguzÖzbek Meliha Burcu Gürdere | 2020 | Journal of Analysis and Testing2020,4,4: | 2 |

| 15 | Benefit period using alendronate to increase bone mineral density in women with osteoporosis?显示文摘Background Alendronate, a nitrogen-containing bisphosphonate is a specific inhibitor of bone resorption and now in the forefront of treatment of osteoporosis. In this study, we reported a significant increase in bone mineral density (BMD) of the spine and the hip in postmenopausal women taking alendronate at 10 mg/d for 1, 2 and 3 years. Methods Participants had received daily, oral, 10 mg dose of alendronate for one to three years and placed into one of three groups according to alendronate treatment duration: 41 women received alendronate for 1 year (group Ⅰ), 46 received alendronate for 2 years (group Ⅱ), and 30 received alendronate for 3 years (group Ⅲ). Measurements of bone density had been made by dual energy X-ray absorbtiometry once each year. Results The differences in L2-L4, L2, L4, femoral neck and trochanter BMD values before and after treatment for first group were significantly different. In second group, significant differences between initial and after treatment were found at the other sites except at the Ward’s triangle. In the third group, only a significant increase in the L2-L4, L2, L3, L4, trochanter BMD values between before treatment and at the end of third year was found. Comparisons between groups were performed with Student’s t test. ANOVA was used to test the age, menopause age, menopause duration and initial BMD values between the three groups. Calculated P values of less than 0.05 were considered statistically significant. Conclusions Alendronate had increased BMD significantly at the spine and hip in postmenopausal women over three years. Increases of BMD in third group were significant during the first and second years.
