| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Overexpression of p27^(KIP1)induced cell cycle arrest in G_1 phase and subsequent apoptosis in HCC-9204 cell line显示文摘AIM We have previously reported that inducibleover-expression of Bak may prolong cell cycle inG1 phase and lead to apoptosis in HCC-9204 cells.This study is to investigate whether p27KIP1playsan important role in this process.METHODS In order to elucidate the exactfunction of p27KIP1in this process,a zinc induciblep27KIP1stable transfectant and transient p27KIP1-GFP fusion transfectant were constructed.Theeffects of inducible,p27KIP1on cell growth,cellcycle arrest and apoptosis were examined in themock,control pMD vector,and pMD-KIP1transfected HCC-9204 cells.RESULTS This p27KIP1-GFP transfectant maytransiently express the fusion gene.The cellgrowth was reduced by 35% at 48 h of p27KIP1induction with zinc treatment as determined bytrypan blue exclusion assay.These differencesremained the same after 72 h of p27KIP1expression,p27KIP1caused cell cycle arrest after24 h of induction,with 40% increase in G1population.Prolonged p27KIP1expression in thiscell line induced apoptotic cell death reflected byTUNEL assay.Fourty-eight h and 72 h of p27KIP1expression showed a characteristic DNA ladder onagarose gel electrophoresis. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 4 | Drawdown mechanism of light particles in baffled stirred tank for the KR desulphurization process显示文摘To improve the efficiency of the desulfurization process, the drawdown mechanism of light particles in stirred tank is studied in this paper. For both up and down pumping modes, the just drawdown speeds(Njd) of floating particles in transformative Kanbara Reactor(KR) are measured in one and four baffled stirred tanks experimentally. Then numerical simulations with standard k-ε model coupled with volume of fluid model(VOF) and discrete phase model(DPM) are conducted to analyze the flow field at the just drawdown speed Njd. The torques on the impeller obtained from experiments and simulations agree well with each other, which indicates the validity of our numerical simulations. Based on the simulations, three main drawdown mechanisms for floating particles, the axial circulation, turbulent fluctuation and largescale eddies, are analyzed. It's found that the axial circulation dominates the drawdown process at small submergence(S = 1/4 T and 1/3 T) and the large-scale eddies play a major role at large submergence(S = 2/3 T and 3/4 T). Besides, the turbulent fluctuation affects the drawdown process significantly for up pumping mode at small submergence(S = 1/4 T and 1/3 T) and for down pumping mode at large submergence(S = 2/3 T and 3/4 T). | 6 | Compression-related EMIC waves drive relativistic electron precipitation显示文摘With coordinated observations of the NOAA 15 satellite and OUL magnetometer station in Finland, we report that the electromagnetic ion cyclotron(EMIC) waves which were stimulated by the compression of the magnetosphere drive relativistic electron precipitation in geoquiescence on 1 Jan 2007. After an enhancement of solar wind dynamic pressure(SWDP), a dayside Pc1 pulsation was observed by the OUL station. Such a Pc1 pulsation is caused by an EMIC wave which propagates from the generation source to lower altitudes. Simultaneously, the NOAA 15 satellite registered an enhancement of precipitating electron count rates with energies >3 Me V within the anisotropic zone of protons. This phenomenon is coincident with the quasi-linear theoretical calculation presented in this paper. | 8 | VSITA, an Improved Approach of Target Amplification in the Identification of Viral Pathogens显示文摘Objective Unbiased next generation sequencing(NGS) is susceptible to interference from host or environmental sequences. Consequently, background depletion and virome enrichment techniques are usually needed for clinical samples where viral load is much lower than background sequences. Methods A viral Sequence Independent Targeted Amplification(VSITA) approach using a set of non-ribosomal and virus-enriched octamers(V8) was developed and compared with traditionally used random hexamers(N6). Forty-five archived clinical samples of different types were used in parallel to compare the V8 and N6 enrichment performance of viral sequences and removal performance of ribosomal sequences in the step of reverse transcription followed by quantitative PCR(qP CR). Ten sera samples from patients with fever of unknown origin and 10 feces samples from patients with diarrhea of unknown origin were used in comparison of V8 and N6 enrichment performance following NGS analysis. Results A minimum 30 hexamers matching to viral reference sequences(sense and antisense) were selected from a dataset of random 4,096(4~6) hexamers(N6). Two random nucleotides were added to the 5' end of the selected hexamers, and 480(30 × 4~2) octamers(V8) were obtained. In general, VSITA approach showed higher enrichment of virus-targeted c DNA and enhanced ability to remove unwanted ribosomal sequences in the majorities of 45 predefined clinical samples. Moreover, VSITA combined with NGS enabled to detect not only more viruses but also achieve more viral reads hit and higher viral genome coverage in 20 clinical samples with diarrhea or fever of unknown origin. | 10 | Meiotic nuclear divisions 1(MND1)fuels cell cycle progression by activating a KLF6/E2F1 positive feedback loop in lung adenocarcinoma显示文摘Background:Considering the increase in the proportion of lung adenocarcinoma(LUAD)cases among all lung cancers and its considerable contribution to cancer-related deaths worldwide,we sought to identify novel oncogenes to provide potential targets and facilitate a better understanding of the malignant progression of LUAD.Methods:The results from the screening of transcriptome and survival analyses according to the integrated Gene Expression Omnibus(GEO)datasets and The Cancer Genome Atlas(TCGA)data were combined,and a promising risk biomarker called meiotic nuclear divisions 1(MND1)was selectively acquired.Cell viability assays and subcutaneous xenograftmodelswere used to validate the oncogenic role ofMND1 in LUADcell proliferation and tumor growth.Aseries of assays,including mass spectrometry,co-immunoprecipitation(Co-IP),and chromatin immunoprecipitation(ChIP),were performed to explore the underlying mechanism.Results:MND1 up-regulation was identified to be an independent risk factor for overall survival in LUAD patients evaluated by both tissue microarray staining and third party data analysis.In vivo and in vitro assays showed that MND1 promoted LUAD cell proliferation by regulating cell cycle.The results of the Co-IP,ChIP and dual-luciferase reporter assays validated that MND1 competitively bound to tumor suppressor Kruppel-like factor 6(KLF6),and thereby protecting E2F transcription factor 1(E2F1)from KLF6-induced transcriptional repression.Luciferase reporter and ChIP assays found that E2F1 activated MND1 transcription by binding to its promoter in a feedback manner.Conclusions:MND1,KLF6,and E2F1 form a positive feedback loop to regulate cell cycle and confer DDP resistance in LUAD.MND1 is crucial for malignant progression and may be a potential therapeutic target in LUAD patients. | Quanli Zhang Run Shi Yongkang Bai Lijuan Meng Jingwen Hu Hongyu Zhu Tongyan Liu Xiaomeng De Siwei Wang Jie Wang Lin Xu Guoren Zhou Rong Yin | 2021 | Cancer Communications2021,41,6: | 2 |

| 15 | Statistical characteristics of the polar ionospheric scale height around the peak height of F_2 layer with observations of the ESR radar: Quiet days显示文摘The ionospheric scale height around the peak height of F2 layer(HmF2)is a very important parameter defining the profile of topside ionosphere.Based on observations of the EISCAT ESR radar,we statistically study the HmF2 at high latitudes.In order to derive the HmF2,a least square method is adopted to fit the electron density profile above the peak height of F2 layer(hmF2)from observations of the ESR radar.The result shows that it is in well agreement between the topside profiles deduced from the fitted HmF2 and the actual measuring topside profile with a height range from hmF2 to 300 km above the hmF2.Therefore,HmF2 can be considered as a constant in this range.With observations of the ESR between 1997 and 2008,we analyze the statistical characteristics of polar HmF2 with local time,season,and solar activity under quiet geomagnetic conditions(Kp≤2),respectively.Variations of HmF2 in quiet days show a diurnal trend with a maximum in the morning and a minimum in the afternoon.The HmF2 always has the highest seasonal amplitude in summer under three kinds of solar activities.The seasonal magnitude of the HmF2 in winter remains between that in spring and that in autumn under low solar activity,while it is the lowest under moderate and high solar activities.We also compare measured HmF2 with those derived by IRI2007,which indicates that IRI2007 model can only approximately provide the average values of polar HmF2 in quiet days but is limited in presenting diurnal and solar activity variations of HmF2. | QIAO Zheng YUAN Zhi Gang QI Feng HUANG Shi Yong LI Hai Meng LI Hui Min LI Ming WANG De Dong | 2015 | Science China(Technological Sciences)2015,58,4: | 1 |

| 20 | Assembly of plant communities in coastal wetlands--the role of saltcedar Tamarix chinensis during early succession显示文摘Aims The mechanisms of plant community assembly are hypothesized to vary at different stages of succession.Here,we explore the local assemblage structure of a herbaceous plant community at its early stage of succession in a supratidal wetland.specifically,we assess the role of Chinese saltcedar(Tamarix chinensis),the lone dominant shrub species,in shaping the spatial structure and species composi-tion in the local plant community,after landscape alteration.Methods We used the multivariate trend-surface analysis for analyzing the spatial structure of the community composition.a null model was also used to detect potential biotic interactions between species.statistical significance was derived from a permutation test by randomizing the presence-absence matrix and functional traits independently.sensitivity analysis by randomly selecting 50 subplots and repeating the null model tests was also done.Finally,rank correlation analysis was used to study the relationship between effect sizes and distance to nearest T.chinensis individuals.Important Findings The herbaceous plant community was highly structured and shaped by the presence of T.chinensis.at local scale,two functional traits,plant height and leaf area,were found to be significantly convergent.Dispersal,environmental stress and interspecific competition played a trivial effect on the local community assembly.The facilitating effect of T.chinensis on the pioneering herbaceous plants,through acting as a wind shelter,was put forward as the dominant community assembly process. | Meng Gao Xinxiu Wang Cang Hui Huapeng Yi Chuanqi Zhang Xiaoqing Wu Xiaoli Bi Yong Wang Luxiang Xiao De Wang | 2015 | Journal of Plant Ecology2015,8,5: | 1 |
