| 1 | Single-cell transcriptomics reveals regulators underlying immune cell diversity and immune subtypes associated with prognosis in nasopharyngeal carcinoma显示文摘Nasopharyngeal carcinoma(NPC)is an aggressive malignancy with extremely skewed ethnic and geographic distributions.Increasing evidence indicates that targeting the tumor microenvironment(TME)represents a promising therapeutic approach in NPC,highlighting an urgent need to deepen the understanding of the complex NPC TME.Here,we generated single-cell transcriptome profiles for 7581 malignant cells and 40,285 immune cells from fifteen primary NPC tumors and one normal sample.We revealed malignant signatures capturing intratumoral transcriptional heterogeneity and predicting aggressiveness of malignant cells.Diverse immune cell subtypes were identified,including novel subtypes such as CLEC9A^+dendritic cells(DCs).We further revealed transcriptional regulators underlying immune cell diversity,and cell–cell interaction analyses highlighted promising immunotherapeutic targets in NPC.Moreover,we established the immune subtype-specific signatures,and demonstrated that the signatures of macrophages,plasmacytoid dendritic cells(pDCs),CLEC9A^+DCs,natural killer(NK)cells,and plasma cells were significantly associated with improved survival outcomes in NPC.Taken together,our findings represent a unique resource providing in-depth insights into the cellular heterogeneity of NPC TME and highlight potential biomarkers for anticancer treatment and risk stratification,laying a new foundation for precision therapies in NPC. | Yu-Pei Chen Jian-Hua Yin Wen-Fei Li Han-Jie Li Dong-Ping Chen Cui-Juan Zhang Jia-Wei Lv Ya-Qin Wang Xiao-Min Li Jun-Yan Li Pan-Pan Zhang Ying-Qin Li Qing-Mei He Xiao-Jing Yang Yuan Lei Ling-Long Tang Guan-Qun Zhou Yan-Ping Mao Chen Wei Ke-Xu Xiong Hong-Bo Zhang Shi-Da Zhu Yong Hou Ying Sun Michael Dean Ido Amit Kui Wu Dong-Ming Kuang Gui-Bo Li Na Liu Jun Ma | 2020 | Cell Research2020,30,11: | 19 |

| 3 | Hurricane disturbances, tree diversity, and succession in North Carolina Piedmont forests, USA显示文摘Windthrow plays a critical role in maintaining species diversity in temperate forests. Do large-scale strong wind events(i.e., tropical cyclones, including hurricanes,typhoons and severe cyclonic storms) increase tree diversity in severely damaged forest areas? Do hurricanes(tropical cyclones that occurs in the Atlantic Ocean and northeastern Pacific Ocean) lead to altered relative abundance of shade-tolerant and shade-intolerant species? Did historic hurricanes alter the succession trajectory of the damaged forests? We used nearly 70-year tree demographic data to assess the effects of two major hurricanes on woody species diversity in Piedmont forests, North Carolina, USA. Species richness(S) and Shannon–Wiener's diversity index(H') were used to evaluate the changes in tree diversity. The changes in composition were assessed with Nonmetric Multidimensional Scaling. The pre-hurricane successional phase can strongly influence both the damage severity and subsequent responses. Although there is often an immediate drop in diversity following a hurricane, understory tree diversity quickly increases to levels that exceed those prior to the disturbance. This leads to an increase in diversity in stands that were substantially damaged. Hurricanes significantly decrease the dominance of shade-intolerant canopy species while increasing preestablished, more shade-tolerant species. | 4 | Strong prognostic value of nodal and bone marrow micro-involvement in patients with pancreatic ductal carcinoma receiving no adjuvant chemotherapy显示文摘AIM: To study the prognostic value of adjuvant chemo-therapy in patients with pancreatic, ductal adenocar-cinoma.METHODS: Lymph nodes from 106 patients with resectable pancreatic ductal adenocarcinoma were systematically sampled. A total of 318 lymph nodes classified histopathologically as tumor-free were examined using sensitive immunohistochemical assays. Forty-three (41%) of the 106 patients were staged as pT1/2, 63 (59%) as pT3/4, 51 (48%) as pN0, and 55 (52%) as pN1. The study population included 59 (56%) patients exhibiting G1/2, and 47 (44%) patients with G3 tumors. Patients received no adjuvant chemo- or radiation therapy and were followed up for a median of 12 (range: 3.5 to 139) mo.RESULTS: Immunostaining with Ber-EP4 revealed nodal microinvolvement in lymph nodes classified as “tumor free” by conventional histopathology in 73 (69%) out of the 106 patients. Twenty-nine (57%)of 51 patients staged histopathologically as pN0 had nodal microinvolvement. The five-year survival probability for pN0-patients was 54% for those without nodal microinvolvement and 0% for those with nodal microinvolvement. Cox-regression modeling revealed the independent prognostic effect of nodal microinvolvement on recurrence-free (relative risk 2.92, P = 0.005) and overall (relative risk 2.49, P = 0.009) survival.CONCLUSION: The study reveals strong and independent prognostic significance of nodal microinvolvement in patients with pancreatic ductal adenocarcinoma who have received no adjuvant therapy.
