| 3 | Genetic mapping of folate QTLs using a segregated population in maizeFA显示文摘As essential B vitamin for humans, folates accumulation in edible parts of crops, such as maize kernels, Is of great importance for human health. But its breeding is always limited by the prohibitive cost of folate profiling. The molecular breeding is a more executable and efficient way for folate fortification, but is limited by the molecular knowledge of folate regulation. Here we report the genetic mapping of folate quantitative trait loci (QTLs) using a segregated population crossed by two maize lines, one high in folate (GEMS31) and the other low in folate (DAN3130). Two folate QTLs on chromosome 5 were obtained by the combination of F2 whole-exome sequencing and F3 kernel-folate profiling. These two QTLs had been confirmed by bulk segregant analysis using F6 pooled DNA and F7 kernel-folate profiling, and were overlapped with QTLs identified by another segregated population. These two QTLs contributed 41.6% of phenotypic variation of 5-formyltetrahydrofolate, the most abundant storage form among folate derivatives in dry maize grains, in the GEMS31×DAN3130 population. | 6 | CD4-positive diffuse large B-cell lymphoma:A variant with aggressive clinical potential显示文摘CD4 expression is rare in diffuse large B-cell lymphoma(DLBCL), with 4 previously reported cases. Its significance is uncertain. We report five patients with CD4+ DLBCL and one CD4+ primary mediastinal large B-cell lymphoma. Cases were identified by searching the electronic database of the department; each was reviewed. Average age was 56 years. Neoplastic cells expressed CD20(5/6 tested cases). BCL2/BCL6 expression were seen in 3/3 tested cases, suggesting a germinal center origin. Additionally, expression of T-cell antigens CD2 and CD5 was noted in 2/2 and CD7 in 1/1 tested case. CD3 was negative in all. Lymph nodes were commonly involved(67%). Patients received chemotherapy +/- radiation(6/6) and bone marrow transplant(2/6). Average survival was 44.2 mo. CD4 expression in DLBCL raises questions of lineage commitment. CD4+ DLBCL is rare; care should be exercised not to diagnose these as T-cell lymphomas. | 7 | Comparative analysis of complete plastid genome reveals powerful barcode regions for identifying wood of Dalbergia odorifera and D. tonkinensis (Leguminosae)显示文摘Dalbergia odorifera T.C.Chen(Leguminosae),a rare and endangered tree species endemic to Hainan Island of China,produces the most expensive and rarest wood in China.The wood characteristics of D.odorifera are remarkably similar to those of D.tonkinensis(a much less sought-after species from Vietnam),and the DNA from wood is often highly degraded,making it very difficult to identify the two species using anatomical features or DNA barcoding based on regular DNA markers.To solve the confusion of identifying wood reliably from the two species,we built and analyzed the plastome library of 26 samples from 18 Dalbergia species,of which 12 samples from eight closely related species of D.odorifera are newly sequenced in this study.Phylogenomic analysis suggested that the relationships among the 26 samples are mostly well resolved,and conspecific individuals from different populations of D.odorifera and D.tonkinensis clustered together.Between the plastid genomes of the two species,we identified 129 indels and 114 single nucleotide polymorphisms.By assessing a subset of 20 nucleotide polymorphisms and 10 indels using 37 population-level samples(20 samples of D.odorifera and 17 samples of D.tonkinensis),we recovered eight species-specific barcode regions that could be suitable for identifying the wood D.odorifera and D.tonkinensis.To examine their utility in wood identification,we amplified the eight DNA barcodes using six wood samples and recovered an amplification success rate of 83.3%,demonstrating a reliable method for precise wood identification of the two species. | Ming Qin Cheng-Jie Zhu Jun-Bo Yang Mohammad Vatanparast Rowan Schley Qiang Lai Dan-Yan Zhang Tie-Yao Tu Bente B.Klitgard Shi-Jin Li Dian-Xiang Zhang | 2022 | Journal of Systematics and Evolution2022,60,1: | 0 |
