| 1 | Paeonol attenuates inflammation-mediated neurotoxicity and microglial activation显示文摘Chronic activation of microglial cells endangers neuronal survival through the release of various proinflammatory and neurotoxic factors. The root of Paeonia lactiflora Pall has been considered useful for the treatment of various disorders in traditional oriental medicine. Paeonol, found in the root of Paeonia lactiflora Pall, has a wide range of pharmacological functions, including anti-oxidative, anti-inflammatory and neuroprotective activities. The objective of this study was to examine the efficacy of paeonol in the repression of inflammation-induced neurotoxicity and microglial cell activation. Organotypic hippocampal slice cultures and primary microglial cells from rat brain were stimulated with bacterial lipopolysaccharide. Paeonol pretreatment was performed for 30 minutes prior to lipopolysaccharide addition. Cell viability and nitrite (the production of nitric oxide), tumor necrosis factor-alpha and interleukin-1beta products were measured after lipopolysaccharide treatment. In organotypic hippocampal slice cultures, paeonol blocked lipopolysaccharide-related hippocampal cell death and inhibited the release of nitrite and interleukin-1beta. Paeonol was effective in inhibiting nitric oxide release from primary microglial cells. It also reduced the lipopolysaccharide-stimulated release of tumor necrosis factor-alpha and interleukin-1β from microglial cells. | 2 | Berberine inhibits inflammatory activation of rat brain microglia显示文摘Chronic activation of microglial cells endangers neuronal survival through the release of various proinflammatory and neurotoxic factors.Berberine,the effective ingredient of Coptidis Rhizoma and Cortex Phellodendri,has a wide range of pharmacological functions,including anti-inflammatory,anti-atherosclerotic and anti-cancer effects.The neuroprotective potential of berberine has previously been demonstrated.The present study aimed to examine whether berberine could repress microglial activation and can be considered a potential therapeutic candidate to target neurodegenerative diseases.Primary microglial cells and BV2 microglial cells were cultured and stimulated with bacterial lipopolysaccharide (LPS).Berberine chloride was treated prior to LPS or simultaneously with LPS stimulation.Results revealed that berberine was effective at inhibiting nitric oxide release from primary microglial cells when cells were exposed to the compound prior to LPS or simultaneously with LPS.It also reduced the LPS-stimulated production of tumor necrosis factor-α,interleukin-1β,prostaglandin E2,and intracellular reactive oxygen species and nuclear factor-kappa activation.Additionally,berberine reduced nitric oxide release from microglia stimulated with interferon-γ and amyloid β.These results suggest that berberine provides neuroprotection by reducing the production of various neurotoxic molecules from activated microglia. | Kyong Nyon Nam Jae-Hong Kim Hoon-Ji Jung Jung-Mi Park Sang-Kwan Moon Young-Suk Kim Sun Yeou Kim Eunjoo H. | 3 | Magnetic resonance imaging of the pancreas in streptozotocin-induced diabetic rats: Gadofluorine P and Gd-DOTA显示文摘AIM: To investigate the performance of Gadofluorine P-enhanced magnetic resonance imaging(MRI) on the diagnosis of diabetes in a streptozotocin(STZ)-induced diabetic rat model.METHODS: Fischer 344 rats were treated with STZ.Rats not treated with STZ served as controls. T1-weighted MRI was performed using a 3T scanner before and after the injection of Gd-DOTA or Gadofluorine P(6 diabetic rats, 5 controls). The normalized signal intensity(SI) and the enhancement ratio(ER) of the pancreas were measured at each time point, and the values were compared between the normal and diabetic rats using the Mann-Whitney test. In addition,the values were correlated with the mean islet number.Optimal cut-off values were calculated using a positive test based on receiver operating characteristics.Intrapancreatic Gd concentration after the injection of each contrast media was measured using laser ablation-inductively coupled plasma-mass spectrometry in a separate set of rats(4 diabetic rats, 4 controls for Gadofluorine P; 2, 2 for Gd-DOTA).RESULTS: The normalized SI and ER of the pancreas using Gd-DOTA were not significantly different between diabetic rats and controls. With Gadofluorine P, the values were significantly higher in the diabetic rats than in the control rats 30 min after injection(P < 0.05). The area under the receiver operating characteristic curve that differentiated diabetic rats from the control group was greater for Gadofluorine P than for Gd-DOTA(0.967 vs 0.667, P = 0.085). An increase in normalized SI 30 min after Gadofluorine P was correlated with a decrease in the mean number of islets(r 2 = 0.510, P = 0.014). Intra-pancreatic Gd was higher in rats with Gadofluorine P injection than Gd-DOTA injection(Gadofluorine P vs Gd-DOTA, 7.37 vs 0.00, P < 0.01). | 16 | Development of large acceptance multi-purpose spectrometer in Korea for symmetry energy显示文摘The Rare Isotope Accelerator complex for ONline experiments(RAON) is a new radioactive ion beam accelerator facility under construction in Korea. The large acceptance multi-purpose spectrometer(LAMPS) is one of the experimental devices for nuclear physics at RAON. It focuses on the nuclear symmetry energy at supra-saturation densities. The LAMPS Collaboration has developed and constructed various detector elements, including a time projection chamber(TPC) and a forward neutron detector array. From the positron beam test, the drift velocity of the secondary electrons in the TPC is 5:3±0:2 cm/ls with P10 gas mixture, and the position resolution for pads with dimensions of 4×15 mm^2 is in the range of$ 0.6–0.8 mm, depending on the beam position. | 17 | Health risks of pest control and disinfection workers after the COVID-19 outbreak in South Korea显示文摘The exposure patterns of pest control and disinfection workers have changed after the coronavirus disease 2019(COVID-19)outbreak,but the health risks of chemical exposure have not been assessed.We identified these workers’chemical exposure patterns and risks before and after the COVID-19 outbreak.We used data conducted between 2018(pre-pandemic)and 2021(post-pandemic)from three-year cross-sectional surveys on pest control and disinfection workers.Inhalation and dermal exposure concentrations were estimated using equations based on a biocidal product risk assessment model from the Korean National Institute of Environmental Research.The non-carcinogenic and carcinogenic risks of chemicals were calculated using the United States Environmental Protection Agency risk assessment model.We found that the annual work frequency(50^(th)percentile)of foggers using disinfectants increased the most among all the work types,from 140 uses/year to 176 uses/year after the COVID-19 outbreak.Moreover,all chemicals’non-carcinogenic and carcinogenic risks increased regardless of exposure routes.In the worst scenario(95^(th)percentile),the margin of exposure for citric acid,benzethonium chloride,benzyl-C12-16-alkyldimethyl chlorides,and sodium chlorite of inhalation exposure,and isopropyl alcohol and benzyl-C12-16-alkyldimethyl chlorides of dermal exposure were acceptable(>100)before the COVID-19outbreak but became unacceptable(<100)after the COVID-19 outbreak.Carcinogenic risks of dichlorvos from inhalation and dermal exposure were above acceptable levels(>10^(-6))before and after the COVID-19 outbreak but comparatively high after the COVID-19 outbreak.Additionally,significantly more workers experienced health symptoms after the COVID-19outbreak(p<0.05),with the most common being muscle lethargy(31%),skin/face stinging(28.7%),and breath shortness/neck pain(24.1%). | Yun-Hee Choi Da-An Huh Lita Kim Sang ji Lee Kyong Whan Moon | 2024 | Journal of Environmental Sciences2024,,5: | 0 |
| 18 | Cholecystectomy increases the risk of metabolic syndrome in the Korean population:a longitudinal cohort study显示文摘Background:Cholecystectomy is a common surgical procedure to treat symptomatic gallstones;however,the long-term outcomes after cholecystectomy are unknown.Therefore,we aimed to investigate whether incident metabolic syndrome(MetS)is associated with cholecystectomy through a large,population-based,longitudinal study.Methods:Subjects aged≥20 years who underwent cholecystectomy from 2010 to 2014(n=76,485)and controls(n=76,485),matched for age and sex,were identified from the Korean National Health Insurance Corporation.Cox proportional hazards analyses were performed to evaluate the association between cases and incident MetS,and hazard ratios and 95% confidence intervals(CIs)were calculated.Results:A total of 152,970 patients were included.Mean age was 52.47±12.76 years,and 50.65% of participants were male.During the follow-up period,there were 38,979(25.48%)newly diagnosed MetS cases in the study participants.The risk of MetS in the cholecystectomy group was approximately 20% higher than that in the control group[adjusted odds ratio(OR),1.20;95%CI:1.17-1.23].In the fully adjusted models,the corresponding ORs for new-onset high waist circumference(WC),low high-density lipoprotein cholesterol(HDL-C)levels,high triglycerides(TG)levels,high blood pressure(BP),and high blood glucose levels were 1.16(1.13-1.19),1.19(1.16-1.22),1.25(1.22-1.28),1.27(1.23-1.31),and 1.21(1.18-1.24),respectively.Cholecystectomy was an independent risk factor of incident MetS,after adjusting for potential confounding factors.In the subgroup analyses,the cholecystectomy group had a higher risk of MetS than the control group in subjects without hypertension or dyslipidemia,respectively.Conclusions:In this large,population-based study,cholecystectomy was associated with an increased risk of developing MetS,independent of other confounding factors.Therefore,careful monitoring of metabolic variables and long-term follow-up are required to evaluate MetS risk after cholecystectomy. | Ji Hye Huh Kyong Joo Lee Yun Kyung Cho Shinje Moon Yoon Jung Kim Kyung-Do Han Jun Goo Kang Seong Jin Lee Sung-Hee Ihm | 2023 | Hepatobiliary Surgery and Nutrition2023,12,4: | 0 |
