2Detection and sequencing of plasmid encoded tetracycline resistance determinants(tetA and tetB) from food-borne Bacillus cereus isolates显示文摘Objective:To investigate the detection and sequencing of plasmid encoded tetracycline resistance genes(tet A and tet B) from food-borne and standard strains of Bacillus cereus(B. cereus).Methods:PCR was carried out to detect the tetracycline resistance genes(tet A and tet B) in food-borne B.cereus strains and the amplified products were sequenced.Results:The phenotypic resistance against tetracycline was observed in 39 of the 118 food-borne isolates and two reference strains(MTCC 430 and MTCC 1307) of B.cereus.Among the phenotypically resistant isolates,tet A was detected in 36 food-borne isolates and two reference strains(MTCC 430 and MTCC 1307).whereas,tel B was detected in 12 food-bome isolates and MTCC 1307 strain. Conclusions:A close association was therefore found between phenotypic resistance against tetracycline and presence of tetracycline resistance genes.The tet A and tet B gene fragments were amplified,purified and sequenced.The gene sequences of the isolates studied herein were found similar to tetA and tetB gene sequences of other bacteria available in NCBI.The occurrence of tetA and tetB genes in B.cereus indicate the horizontal transfer of antibiotic resistance determinants from other bacteria into B.cereus.The transfer of these resistant determinants to other potentially pathogenic bacteria may be a matter of great concern.Mudasir Ali Rather Rabinder Singh Aulakh Jatinder Paul Singh Gill Abdul Qayoom Mir Mir Nadeem Hassan 2012Asian Pacific Journal of Tropical Medicine2012,5,9:6
3DCC/DMAP mediated esterification of hydroxy and non-hydroxy olefinic fatty acids with β-sitosterol:In vitro antimicrobial activity显示文摘A new series of fatty alkenoates were synthesized using an appropriate synthetic route involving DCC and DMAP as catalysts. Compounds were characterized by their spectral data. All the synthesized compounds were evaluated for their in vitro antimicrobial activity. The minimum inhibitory concentration (MIC), minimum bacterial concentration (MBC) and minimum fungicidal concentration (MFC) were determined for test compounds as well as for reference standards. 4Histone deacetylase inhibitor pre-treatment enhances the efficacy of DNA-interacting chemotherapeutic drugs in gastric cancer显示文摘BACKGROUND The prognosis of gastric cancer continues to remain poor,and epigenetic drugs like histone deacetylase inhibitors(HDACi)have been envisaged as potential therapeutic agents.Nevertheless,clinical trials are facing issues with toxicity and efficacy against solid tumors,which may be partly due to the lack of patient stratification for effective treatments.To study the need of patient stratification before HDACi treatment,and the efficacy of pre-treatment of HDACi as a chemotherapeutic drug sensitizer.METHODS The expression activity of class 1 HDACs and histone acetylation was examined in human gastric cancer cells and tissues.The potential combinatorial regime of HDACi and chemotherapy drugs was defined on the basis of observed drug binding assays,chromatin remodeling and cell death.RESULTS In the present study,the data suggest that the differential increase in HDAC activity and the expression of class 1 HDACs are associated with hypoacetylation of histone proteins in tumors compared to normal adjacent mucosa tissue samples of gastric cancer.The data highlights for the first time that pretreatment of HDACi results in an increased amount of DNA-bound drugs associated with enhanced histone acetylation,chromatin relaxation and cell cycle arrest.Fraction-affected plots and combination index-based analysis show that pre-HDACi chemo drug combinatorial regimes,including valproic acid with cisplatin or oxaliplatin and trichostatin A with epirubicin,exhibit synergism with maximum cytotoxic potential due to higher cell death at low combined doses in gastric cancer cell lines.CONCLUSION Expression or activity of class 1 HDACs among gastric cancer patients present an effective approach for patient stratification.Furthermore,HDACi therapy in pretreatment regimes is more effective with chemotherapy drugs,and may aid in predicting individual patient prognosis.Ramchandra Vigay Amnekar Shafqat Ali Khan Mudasir Rashid Bharat Khade Rahul Thorat Poonam Gera Shailesh V Shrikhande Duane T Smoot Hassan Ashktorab Sanjay Gupta 2020World Journal of Gastroenterology2020,26,6:3
5Synthesis of surface imprinted polymers based on wrinkled flower-like magnetic graphene microspheres with favorable recognition ability for BSA显示文摘Nowadays,the employing of molecular imprinting technique in the analysis and separation of proteins from complex biological samples has been widely favored by researchers.To enrich the types of surface protein imprinted materials and expand the application fields of graphene materials,novel surface molecular imprinted polymers(MIPs)based on magnetic graphene microspheres Fe_(3)O_(4)@r GO@MIPs are first synthesized in this paper.Fe_(3)O_(4)@r GO@MIPs are prepared by oxidative self-polymerization of dopamine on the surface of magnetic graphene(Fe_(3)O_(4)@r GO)composite microspheres.Bovine serum albumin(BSA)is selected as protein template.Fe_(3)O_(4)@r GO microspheres with wrinkled flower-like structure are obtained by compounding Fe_(3)O_(4)and graphene oxide in an appropriate ratio via the method of high-temperature reduction self-assembly.The microspheres exhibit promising dispersibility,high external surface area,rich pore structure,and sufficient magnetic properties.These advantages not only prevent the agglomeration of imprinted microspheres in the aqueous phase,which is conducive to contact and static adsorption,but also increase the amount of protein imprinting.Additionally,sufficient magnetic properties ensure fast and effective separation of the adsorbents.While the adsorption capacity is increased,the separation procedure becomes simple.The binding capacity of Fe_(3)O_(4)@r GO@MIPs for BSA can reach 317.58 mg/g within 60 min,and the imprinting factor(IF)is 4.24.More importantly,Fe_(3)O_(4)@r GO@MIPs can specifically recognize the target BSA from the mixed proteins and the actual sample.There is no significant decrease in the adsorption amount,IF,and magnetic properties after eight runs.It is promising to be used in the separation of proteins from the actual biological samples.Zuoting Yang Ke Yang Yuhong Cui Tariq Shah Mudasir Ahmad Qiuyu Zhang Baoliang Zhang 2021Journal of Materials Science & Technology2021,,15:2

8Invasion by Conyza sumatrensis alters soil microbial community structure in urban ecosystems显示文摘Introduction:Whether invasive plants stimulate or inhibit the soil microbial diversity is still an open question.Despite large-scale invasion by Conyza sumatrensis(Retz.)E.Walker in the urban ecosystems of the Srinagar city of the Kashmir Himalayan region,limited information exists on its impact,particularly,on the belowground microbial diversity.The present study was thus conducted to compare the soil microbial(bacterial and ascomycetous fungal)diversity between the sites invaded by C.sumatrensis and un-invaded(control)sites.Methods:Soil metagenome was extracted from C.sumatrensis invaded and un-invaded plots at the three study sites.A total of six plots(5×5 m each in size),including three invaded by C.sumatrensis and three un-invaded plots were nested within each study site.DNA after amplification was subject to denaturing gradient gel electrophoresis(DGGE);the bands were extracted from the DGGE gel,re-amplified,and sequenced for identification of the species.Results:The number of bacterial species was reduced in the invaded plots at two out of the three sites while as it was relatively higher in the un-invaded plots with many species exclusively found in these plots.Fungal species richness was higher in the invaded plots compared to the un-invaded plots at all the three sites.Also,more fungal species were found to occur exclusively in the invaded plots without being represented in the un-invaded plots.Conclusions:Invasion by C.sumatrensis alters soil microbial community structure in the urban ecosystems in the Kashmir Himalaya.How this species does so and what benefits does it draw from such alteration promise to be an interesting future discourse.Nazima Rasool Zafar A.Reshi Damase P.Khasa Mudasir Roshan Manzoor A.Shah 2016Ecological Processes2016,5,1:1

METHODS: The isolation of compounds was achieved by chromatographic techniques and structure of the isolates was established by UV, IR, HRESI-MS and NMR including 1D and 2D experiments. RESULTS: Bioassay-guided fractionation of an ethanolic extract of Polygonum amplexicaule led to the isolation of a hitherto unidentified compound, 5, 6-dihydropyranobenzopyrone (1) along with nine previously known com-pounds (2-10). Compounds 2-10 were identified as amplexicine (2), catechin (3), rutin (4), quercetin-3-O-β-D-galactopyranoside (5), chlorogenic acid (6), galloyl glucose (7), caffeic acid (8), gallic acid (9) and scopletin (10). CONCLUSION: Compound 1 is new. Compounds 1-10 exhibited considerable antioxidant activity in a 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging assay.Mudasir A. Tantry Mohamed M. 17Statistical optimization of lignocellulosic waste containing culture medium for enhanced production of cellulase by Bacillus tequilensis G9显示文摘The ever increasing energy demands of modern civilization and rapidly dwindling fossil fuels point towards a renewable substitute like biofuels.However,higher costs associated with biofuel productions is the major bottleneck for its commer-cialization.The present study demonstrates the use of a statistical approach called response surface methodology(RSM)to investigate the optimum parameters for maximum production of cellulase by Bacillus tequilensis G9.The Plackett-Bur-man design(PB)of the RSM analysis indicated grass straw(GS)concentration,pH,FeSO4,inoculum,MgSO4,incubation period and NH4Cl as significant variables that influence the cellulase production.Further,to propose the best medium for the maximum production of cellulase by B.tequilensis G9,the most influential parameters,namely concentrations of GS as substrate,FeSO4,pH,inoculum size,etc.were fine-tuned by central composite design(CCD)involving four factors and five levels.The CCD analysis demonstrated 8%substrate concentration,1.5%of inoculum along with 10 ppm FeSO4 and a pH of 5.5 in media as optimum conditions for highest enzyme production.The field emission scanning electron microscopic analysis of the treated GS showed structural alterations depicting significant deconstruction caused by B.tequilensis G9.The yield of the partially purified cellulase proteins were found to be 21%revealing molecular mass between 30 and 97 kDa.The enhanced cellulase production by B.tequilensis G9 demonstrated in our study brands its applications in many industrial processes like biorefinery,biofuels,etc.Mudasir A.Dar Kiran D.Pawar Jyoti M.Chintalchere Radhakrishna S.Pandit 2019Waste Disposal and Sustainable Energy2019,1,3:0
18Exploring the region-wise diversity and functions of symbiotic bacteria in the gut system of wood-feeding termite,Coptotermes formosanus,toward the degradation of cellulose,hemicellulose,and organic dyes显示文摘The wood-feeding termite Coptotermes formosanus represents a unique and impressive system for lignocellulose degradation.The highly efficient digestion of lignocellulose is achieved through symbiosis with gut symbionts like bacteria.Despite extensive research during the last three decades,diversity of bacterial symbionts residing in individual gut regions of the termite and their associated functions is still lacking.To this end,cellulose,xylan,and dye-decolorization bacteria residing in foregut,midgut,and hindgut regions of C.formosanus were enlisted by using enrichment and culture-dependent molecular methods.A total of 87 bacterial strains were successfully isolated from different gut regions of C.formosanus which belonged to 27 different species of 10 genera,majorly affiliated with Proteobacteria(80%)and Firmicutes(18.3%).Among the gut regions,37.9%of the total bacterial isolates were observed in the hindgut that demonstrated predominance of cellulolytic bacteria(47.6%).The majority of the xylanolytic and dye-decolorization bacteria(50%)were obtained from the foregut and midgut,respectively.Actinobacteria represented by Dietza sp.was observed in the hindgut only.Based on species richness,the highest diversity was observed in midgut and hindgut regions each of which harbored seven unique bacterial species.The members of Enterobacter,Klebsiella,and Pseudomonas were common among the gut regions.The lignocellulolytic activities of the selected potential bacteria signpost their assistance to the host for lignocellulose digestion.The overall results indicate that C.formosanus harbors diverse communities of lignocellulolytic bacteria in different regions of the gut system.These observations will significantly advance our understanding of the termite–bacteria symbiosis and their microbial ecology uniquely existed in different gut regions of C.formosanus,which may further shed a light on its potential values at termite-modeled biotechnology.Mudasir A.Dar Rongrong Xie Radhakrishna S.Pandit Blessing Danso Chenchen Dong Jianzhong Sun 2022Insect Science2022,29,5:0
