1Profiling the ionome of rice and its use in discriminating geographical origins at the regional scale, China显示文摘Element profile was investigated for their use to trace the geographical origin of rice (Oryza sativa L.) samples. The concentrations of 13 elements (calcium (Ca), potassium (K), magnesium (Mg), phosphorus (P), boron (B), manganese (Mn), iron (Fe), nickel (Ni), copper (Cu), arsenic (As), selenium (Se), molybdenum (Mo), and cadmium (Cd)) were determined in the rice samples by inductively coupled plasma optical emission and mass spectrometry. Most of the essential elements for human health in rice were within normal ranges except for Mo and Se. Mo concentrations were twice as high as those in rice from Vietnam and Spain. Meanwhile, Se concentrations were three times lower in the whole province compared to the Chinese average level of 0.088 mg/kg. About 12% of the rice samples failed the Chinese national food safety standard of 0.2 mg/kg for Cd. Combined with the multi-elemental profile in rice, the principal component analysis (PCA), discriminant function analysis (DFA) and Fibonacci index analysis (FIA) were applied to discriminate geographical origins of the samples. Results indicated that the FIA method could achieve a more effective geographical origin classification compared with PCA and DFA, due to its efficiency in making the grouping even when the elemental variability was so high that PCA and DFA showed little discriminatory power. Furthermore, some elements were identified as the most powerful indicators of geographical origin: Ca, Ni, Fe and Cd. This suggests that the newly established methodology of FIA based on the ionome profile can be applied to determine the geographical origin of rice.Gang Li Luis Nunes Yijie Wang Paul N. 3Bridging the injured spinal cord with neural stem cells显示文摘Spinal cord injury(SCI)damages not only the gray matter neurons,but also the white matter axonal tracts that carry signals to and from the brain,resulting in permanent loss of function below injury.Neural stem cells(NSCs)have high therapeutic potential for reconstruction of the injured spinal cord since they can potentially form neuronal relays to bridge functional connectivity between separated spinal cord segments.This requires host axonal regeneration into and connectivity with donor neurons,and axonal growth and connectivity of donor neurons to host central nervous system(CNS)circuitry.In this mini-review,we will discuss key studies that explore novel neuronal relay formation by grafting NSCs in models of SCI,with emphasis on long-distance axonal growth and connectivity of NSCs grafted into injured spinal cord.Jennifer N. 4A short-chain α-neurotoxin from Naja naja atra produces potent cholinergic-dependent analgesia显示文摘Objective To investigate the analgesia induced by cobrotoxin (CT) from venom of Naja naja atra, and the effects of atropine and naloxone on the antinociceptive activity of CT in rodent pain models. Methods CT was administered intraperitoneally (33.3,50,7μg/kg) , intra-cerebral venticularly (2.4μg /kg) or microinjected into periaque-ductal gray (PAG,1.2μg /kg). The antinociceptive action was tested using the hot-plate test and the acetic acid writhing test in mice and rats. The involvement of cholinergic system and the opioid system in CT-induced analgesia was examined by pretreatment of animals with atropine (0.5mg/kg, im or 10mg/kg, ip) or naloxone (3mg/kg,ip) . The effect of CT on motor activity was tested using the Animex test. Results CT (33.3,50 and 75μg/kg,ip) exhibited a dose-dependent analgesic action in mice as determined with hot-plate test and acetic acid writhing test. In the mouse acetic acid writhing test, the intra-cerebral ventricle administration of CT 2.4μg/kg (1/23th of a systemic dose) produced marked analgesic effects. Microinjection of CT 1.2μg/kg (1/46th of systemic dose) into the PAG also elicited a robust analgesic action in the hot-plate test in rats. Atropine at 0.5 mg/kg (im) or naloxone at 3 mg/kg (ip) failed to block the analgesic effects of CT, but atropine at 10 mg/kg (ip) did antagonize the analgesia mediated by CT in the mouse acetic acid writhing test. At the highest effective dose of antinociception (75μg/kg) , CT did not change the spontaneous mobility of mice. Conclusion These results suggest that CT from Naja naja atra venom has analgesic effects. Central nervous system may be involved in CT' analgesic effects and the PAG may be the primary central site where CT exerts its effects. 6动脉瘤性蛛网膜下腔出血患者的重症监护处理:神经危重症监护学会多学科共识会议的推荐意见显示文摘蛛网膜下腔出血(subarachnoidhemorrhage,SAH)是一种可对中枢神经系统造成毁灭性打击并对其他多个器官产生显著影响的急性脑血管病。SAH患者被常规收入重症监护病房并由多学科团队进行治疗。由于高质量研究证据的缺乏,使得治疗方法各异,可供选择的指导意见不多。现有的指南主要强调危险因素、预防、自然史以及再出血的预防,很少涉及SAH患者的重症监护问题。美国神经危重症监护学会组织了一次国际性多学科共识会议来探讨SAH的重症监护处理。根据发表的文章和研究领域,此次会议召集了来自欧洲和北美地区的神经重症监护科、神经外科、神经内科、介入神经放射科和神经麻醉科方面的专家。根据临床经验和制定实践指南方面的经验,选择4名经验丰富的神经重症监护专家组成评判委员会。应用推荐分级的评估、制定与评价(GradesofRecommendationsAssessment,DevelopmentandEvaluation,GRADE)系统进行文献回顾,结合参会者的经验、评判委员会的审评以及文献讨论产生推荐意见。应用GRADE系统制定推荐意见,其制定原则不仅强调研究资料的质量,而且还重视利弊权衡与实践转化。对SAH患者日常处理过程中面临的所有问题均提供指导和推荐意见,即使缺乏高质量的证据。Michael N. 10Establishment of Withania somnifera Hairy Root Cultures for the Production of Withanolide A显示文摘Withania sominifera (Indian ginseng) was transformed by Agrobacterium rhizogenes. Explants from seedling roots, stems, hypocotyls, cotyledonary nodal segments, cotyledons and young leaves were inoculated with A. rhizogenes strain R1601. Hairy (transformed) roots were induced from cotyledons and leaf explants. The transgenic status of hairy roots was confirmed by polymerase chain reaction using nptII and rolB specific primers and, subsequently, by Southern analysis for the presence of nptII and rolB genes in the genomes of transformed roots. Four clones of hairy roots were established; these differed in their morphology. The doubling time of faster growing cultures was 8-14 d with a fivefold increase in biomass after 28 d compared with cultured, non-transformed seedling roots. MS-based liquid medium was superior for the growth of transformed roots compared with other culture media evaluated (SH, LS and N6), with MS-based medium supplemented with 40 g/L sucrose being optimal for biomass production. Cultured hairy roots synthesized withanolide A, a steroidal lactone of medicinal and therapeutic value. The concentration of withanolide A in transformed roots (157.4 μg/g dry weight) was 2.7-fold more than in non-transformed cultured roots (57.9 μg/g dry weight).Hosakatte N.
