| 1 | Preparation and characterization of nanoparticles from quaternized cyclodextrin-grafted chitosan associated with hyaluronic acid for cosmetics显示文摘Hyaluronic acid(HA, 20–50 kDa) is a hydrophilic macromolecule with anti-wrinkle effects and moisturizing properties. However, its high molecular weight prevents it from penetrating into the deeper layers of the skin and, thus, limits its benefits to topical effects. Thus, the objective of this study is to prepare nanoparticles of quaternized cyclodextrin-grafted chitosan(QCD-g-CS) associated with HA in different molar ratios of QCD-g-CS and HA. The conjugation of the carboxylic moieties of HA and the amides of QCD-g-CS was confirmed by Fourier-transform infrared spectroscopy. Thus, the system was optimized to create nanoparticles with a small size(235.63 ± 21.89 nm), narrow polydispersity index(0.13 ± 0.02), and zeta potential of 16.07 ± 0.65 m V. The association efficiency and loading efficiency were determined by ultra-performance liquid chromatography as 86.77 ± 0.69% and 10.85 ± 0.09%, respectively. The spherical morphology of the obtained nanoparticles was confirmed by transmission electron microscopy. Moreover, the in-vitro hydrating ability was significantly higher( P < 0.001) than that of bulk HA(3.29 ± 0.41 and 1.71 ± 0.05 g water/g sample, respectively). The safety of these nanoparticles at concentrations in the range of 0.01–0.10 mg/ml was confirmed via tests on human skin fibroblasts. | 6 | 一种新型莱克多巴胺-蛋白质载体结合技术在侧向流试纸条检测动物饲料中莱克多巴胺含量中的应用(英文)显示文摘目的:开发一种新型简单快速的克莱多巴胺-蛋白质载体结合方法,用于侧向流试纸条检测动物饲料。创新点:基于曼尼希反应的克莱多巴胺-牛血清蛋白(RAC–BSA)结合方法,具有一步性、简单、快速,且所需试剂少的优点。方法:通过曼尼希反应产生RAC–BSA结合物,制备竞争型侧向流试纸条。将RAC标记于猪饲料中,以体积比10:90的甲醇:电泳缓冲液作为提取缓冲液,在25min内完成样品制备。通过裸眼观察定性评估RAC–BSA结合物的免疫应答以及检测灵敏性,通过智能手机和电脑软件(ImageJ)进行半定量分析。结论:在最佳条件下,侧向流试纸条检测的检出限(LOD)在5 min裸眼评估下为1 ng/g;半定量分析中,其线性范围为0.075~0.750ng/g,计算的LOD为0.10 ng/g,定量限为0.33 ng/g,且相关性较好(R2=0.992)。回收率为96.4%~103.7%,同实验内及不同实验间的相对标准偏差为2.5%~3.6%。通过与酶联免疫吸附法对比,该侧向流试纸条检测结果具有较高的准确性和特异性。因此,通过曼尼希反应制备RAC–BSA结合物是一步的简单的结合法,可用于侧向流试纸条检测法。 | Pattarachaya PREECHAKASEDKIT Nattaya NGAMROJANAVANICH Nanthika KHONGCHAREONPORN Orawon CHAILAPAKUL | 2019 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2019，20，2： | 0 |
| 7 | Modified albumin-bilirubin predicted survival of unresectable hepatocellular carcinoma patients treated with immunotherapy显示文摘BACKGROUND Modified albumin-bilirubin(mALBI)grade has been established as a survival determinant in hepatocellular carcinoma(HCC)patients who receive locoregional and targeted therapies.AIM To investigate whether mALBI could predict survival in unresectable HCC(uHCC)patients who were treated with atezolizumab plus bevacizumab(AB).METHODS A single-center,retrospective cohort study enrolled uHCC patients who received AB treatment between September 2020 and April 2023 and were followed up until June 2023.An association between mALBI and patient survival was determined using Cox proportional hazards analysis.RESULTS Of the 83 patients,67 patients(80.7%)were male with the mean age of 60.6 years.Among them,22 patients(26.5%)were classified as Barcelona Clinic Liver Cancer B,and 61 patients(73.5%)were classified as Barcelona Clinic Liver Cancer C.Cirrhosis was present in 76 patients(91.6%),with 58 patients classified as Child-Turcotte-Pugh(CTP)A and 18 as CTP B.The median overall survival(OS)and progression-free survival were 13.0 mo[95%confidence interval(CI):5.2-20.8]and 9.0 mo(95%CI:5.0-13.0),respectively.The patients were divided into two groups based on mALBI grades:42 patients(50.6%)in the mALBI 1+2a group;and 41 patients(49.4%)in the mALBI 2b+3 group.During the median follow-up period of 7.0 mo,the mALBI 1+2a group exhibited significantly better survival compared to the mALBI 2b+3 group,with a median OS that was not reached vs 3.0 mo(95%CI:0.1-6.0,P<0.001).In a subgroup of patients with CTP A,the mALBI 1+2a group also showed significantly longer survival compared to the mALBI 2b+3 group,with a median OS that was not reached vs 6.0 mo(95%CI:3.4-8.6,P<0.001).In the multivariate analysis,both CTP class and mALBI grade were independently associated with survival,with adjusted hazard ratios(95%CI)of 2.63(1.19-5.78,P=0.020)and 3.90(1.71-8.90,P=0.001),respectively.CONCLUSION mALBI grades can determine survival of uHCC patients receiving AB treatment,particularly those who have mildly impaired liver function.This highlights the importance of assessing mALBI before initiating AB treatment to optimize therapeutic efficacy in clinical practice. | Huttakan Navadurong Thaninee Prasoppokakorn Nanicha Siriwong Chonlada Phathong Nattaya Teeyapun Suebpong Tanasanvimon Kessarin Thanapirom Piyawat Komolmit Pisit Tangkijvanich Sombat Treeprasertsuk Roongruedee Chaiteerakij | 2023 | World Journal of Gastrointestinal Oncology2023，15，10： | 0 |
