1A urinary proteomic landscape of COVID-19 progression identifies signaling pathways and therapeutic options显示文摘Signaling pathway alterations in COVID-19 of living humans as well as therapeutic targets of the host proteins are not clear.We analyzed 317 urine proteomes,including 86 COVID-19,55 pneumonia and 176 healthy controls,and identified specific RNA virus detector protein DDX58/RIG-I only in COVID-19 samples.Comparison of the COVID-19 urinary proteomes with controls revealed major pathway alterations in immunity,metabolism and protein localization.Biomarkers that may stratify severe symptoms from moderate ones suggested that macrophage induced inflammation and thrombolysis may play a critical role in worsening the disease.Hyper activation of the TCA cycle is evident and a macrophage enriched enzyme CLYBL is up regulated in COVID-19 patients.As CLYBL converts the immune modulatory TCA cycle metabolite itaconate through the citramalyl-CoA intermediate to acetyl-CoA,an increase in CLYBL may lead to the depletion of itaconate,limiting its anti-inflammatory function.These observations suggest that supplementation of itaconate and inhibition of CLYBL are possible therapeutic options for treating COVID-19,opening an avenue of modulating host defense as a means of combating SARS-CoV-2 viruses.Yuntao Liu Lan Song Nairen Zheng Jinwen Shi Hongxing Wu Xing Yang Nianci Xue Xing Chen Yimin Li Changqing Sun Cha Chen Lijuan Tang Xiaotian Ni Yi Wang Yaling Shi Jianwen Guo Guangshun Wang Zhongde Zhang Jun Qin 2022Science China(Life Sciences)2022,65,9:1

16Sino-Turkish Tourism Cooperation under the Framework of the Belt and Road Initiative显示文摘In recent years,China has vigorously promoted the implementation of the Belt and Road Initiative(BRI).As a key area of communication among peoples,tourism has become an important aspect for cooperation between China and the countries along the‘Belt and Road’.China established the Ministry of Culture and Tourism in 2018 to promote the cultural and tourist industries,providing important support for their development.Meanwhile,Turkey has abundant religious and cultural tourism resources,and it is actively tapping the Chinese market,hoping to attract more Chinese tourists.This has laid the foundation for tourism cooperation between the two countries.In 2018 the Year of Turkish Tourism was celebrated in China.Within the year a series of cultural and tourist activities by Turkish organisations were carried out in various Chinese cities for the purpose of enhancing the appeal of Turkish tourism.Despite the outstanding advantages of Turkey’s tourist resources,its unstable security situation and political disputes between the two countries have negative impacts on tourism cooperation.Moreover,Turkey’s domestic tourist industry is facing a shortage of measures to effectively meet the language needs of Chinese tourists,which also inhibits the willingness of Chinese tourists to travel to Turkey.Song Niu Nianci Li 2019Asian Journal of Middle Eastern and Islamic Studies2019,13,3:0

18Rassf2 overexpression mediated by AAV promotes the supporting cell-to-hair cell transformation in the cochlea显示文摘Sensory hair cells are responsible for detecting and transmitting sound in the inner ear,and damage to HCs leads to hearing loss.HCs do not regenerate spontaneously in adult mammals,which makes the hearing loss permanent.However,hair cells and supporting cells have the same precursors in the inner ear,and in newborn mice,the adjacent SCs can be activated by gene manipulation to differentiate into newly regenerated hair cells.Here,we demonstrate the role of the Ras association domain family member 2(Rassf2)in supporting cell to hair cell trans-differentiation in the inner ear.Using the AAV vector(AAV-ie)to upregulate Rassf2 expression promoted supporting cell division and hair cell production in cultured cochlear organoids.Also,AAV-Rassf2 enhanced the regenerative ability of Lgr5+SCs in the postnatal cochlea without impairing hearing,and this might due to the modulation of the Wnt,Hedgehog and Notch signaling pathways.Furthermore,AAV-Rassf2 enhances cochlear supporting cell division and hair cell production in the neomycin injury model.In summary,our results suggest that Rassf2 is a key component in HC regenerative repair,and gene modulation mediated by adeno-associated virus may be a promising gene therapy for hearing repair.Liyan Zhang Jieyu Qi Yuan Fang Fangzhi Tan Yinyi Zhou Ziyu Zhang Qiuhan Sun Nianci Li Yideng Huang Jingwu Sun Renjie Chai 2023Engineered Regeneration2023,4,3:0
19Loop CD20/CD19 CAR-T cells eradicate B-cell malignancies efficiently显示文摘CD19 chimeric antigen receptor(CAR)T cells have shown robust efficacy in relapsed and refractory acute lymphoblastic leukemia(R/R ALL),but compromising result in chronic lymphoblastic leukemia(CLL)and non-Hodgkin’s lymphoma(NHL).CD19-relapse and the lack of CAR-T cell persistence which result in treatment failure are considerable obstacles to overcome.CAR-T targeting CD20 is an option for salvaging CD19 CAR-T failure.Previous studies have established variant structures of bispecific CAR-T which could avoid antigen-loss and immune escape.Here,we constructed tandem and loop CAR structures targeting both CD19 and CD20 antigen.Bispecific CAR-T cells could eliminate either CD19 or CD20 negative lymphoma cells,suggesting they exhibited dual antigen targeting of CD19 and CD20.By comparing the efficiency of four bispecific CAR modified T cells,it was found that loop2019 CAR was the best structure among them to eradicate lymphoma cell lines and patients’primary lymphoma or CLL cells in a very low dose in vitro and prolong the survival time dramatically in lymphoma xenograft mice model.These data highlighted the potential of loop2019 CAR-T in clinical treatment.Zhaoqi Chen Yan Liu Nianci Chen Haiyan Xing Zheng Tian Kejing Tang Qing Rao Yingxi Xu Ying Wang Min Wang Jianxiang Wang 2023Science China(Life Sciences)2023,66,4:0
