1In vitro α-amylase inhibitory activity and in vivo hypoglycemic effect of methanol extract of Citrus macroptera Montr.fruit显示文摘Objective:To investigate the therapeutic effects of methanol extract of Citrus macroptera Montr,fruit inα-amylase inhibitory activity(in vitro)and hypoglycemic activity in normal and glucose induced hyperglycemic rats(in vivo).Methods:Fruits of Citrus macroptera without rind was extracted with pure methanol following cold extraction and tested for presence of phytochemical constituents,α-amylase inhibitory activity,and hypoglycemic effect in normal rats and glucose induced hyperglycemic rats.Results:Presence of saponin,steroid and terpenoid were identified in the extract.The results showed that fruit extract had moderateα-amylase inhibitory activity[IC_(50)value=(3.638±0.190)mg/mL]as compared to acarbose.Moreover at 500 mg/kg and 1000 mg/kg doses fruit extract significantly(P<0.05 and P<0.01 respectively)reduced fasting blood glucose level in normal rats as compared to glibenclamide(5 mg/kg).In oral glucose tolerance test,500 mg/kg dose significantly reduced blood glucose level(P<0.05)at 2 h but 1000 mg/kg dose significantly reduced blood glucose level at 2 h and 3 h(P<0.05 and P<0.01 respectively)whereas glibenclamide(5 mg/kg)significantly reduced glucose level at every hour after administration.Overall time effect is also considered extremely significant with F value=23.83 and P value=0.0001 in oral glucose tolerance test.Conclusion:These findings suggest that the plant may be a potential source for the development of new oral hypoglycemic agent.Nizam Uddin Rakib Hasan Monir Hossain Arjyabrata Sarker A.H.M.Nazmul Hasan A.F.M.Mahmudul Islam Mohd.Motaher H.Chowdhury Sohel Rana 2014Asian Pacific Journal of Tropical Biomedicine2014,4,6:4
2Germline mutation analysis of MLH1 and MSH2 in Malaysian Lynch syndrome patients显示文摘AIM: To investigate the protein expression profi le of mismatch repair (MMR) genes in suspected cases of Lynch syndrome and to characterize the associated germline mutations. METHODS: Immunohistochemical analysis of tumor samples was performed to determine the protein expression profile of MMR protein. Germline mutation screening was carried out on peripheral blood samples. The entire exon regions of MLH1 and MSH2 geneswere amplifi ed by polymerase chain reaction, screened by denaturing high performance liquid chromatography (dHPLC) and analyzed by DNA sequencing to characterize the germline mutations. RESULTS: Three out of 34 tissue samples (8.8%) and four out of 34 tissue samples (11.8%) showed loss of nuclear staining by immunohistochemistry, indicating the absence of MLH1 and MSH2 protein expression in carcinoma cells, respectively. dHPLC analysis followed by DNA sequencing showed these samples to have germline mutations of MSH2 gene. However, no deleterious mutations were identifi ed in any of the 19 exons or coding regions of MLH1 gene, but we were able to identify MLH1 promoter polymorphism, -93G > A (rs1800734), in 21 out of 34 patients (61.8%). We identified one novel mutation, transversion mutation c.2005G > C, which resulted in a missense mutation (Gly669Arg), a transversion mutation in exon 1, c.142G > T, which resulted in a nonsense mutation (Glu48Stop) and splice-site mutation, c.2006-6T > C, which was adjacent to exon 13 of MSH2 gene. CONCLUSION: Germline mutations were identified in four Malaysian Lynch syndrome patients. 3Experimental Study on Cross-Flow Induced Vibrations in Heat Exchanger Tube Bundle显示文摘Vibration in heat exchangers is one of the main problems that the industry has faced over last few decades. Vibration phenomenon in heat exchangers is of major concern for designers and process engineers since it can lead to the tube damage, tube leakage, baffle damage, tube collision damage, fatigue, creep etc. In the present study, vibration response is analyzed on single tube located in the centre of the tube bundle having parallel triangular arrangement(60°) with P/D ratio of 1.44. The experiment is performed for two different flow conditions. This kind of experiment has not been reported in the literature. Under the first condition, the tube vibration response is analyzed when there is no internal flow in the tube and under the second condition, the response is analyzed when the internal tube flow is maintained at a constant value of 0.1 m/s. The free stream shell side velocity ranges from 0.8 m/s to 1.3 m/s, the reduced gap velocity varies from 1.80 to 2.66 and the Reynolds number varies from 44500 to 66000. It is observed that the internal tube flow results in larger vibration amplitudes for the tube than that without internal tube flow. It is also established that over the current range of shell side flow velocity, the turbulence is the dominant excitation mechanism for producing vibration in the tube since the amplitude varies directly with the increase in the shell side velocity. 4Nanoindentation of Teeth-A Review显示文摘This paper reviews some of the nanoindentation research that has been done on teeth and discusses the usefulness of this technique in studying the structure-property-function relationship at the micro and nanometer scale. In particular, examples on the use of nanoindentation technique in investigating the effects of cleansing and bleaching agents as well as drinks on the micro- and nanostructures and mechanical properties of teeth are highlighted. Although nanoindentation on teeth is a relatively new area of research, it provides an excellent way of probing and relating the structures and mechanical properties of teeth at the submicron and nanometer scales that were previously not possible but which can now greatly benefit dental research. For example, a nanoindenter with a high resolution imaging capability can help to elucidate the mechanisms with which certain diseases can damage and compromise the structural integrity of teeth at the micro- and nanometer scale. 5Methanolic extract of Abrus precatorius promotes breast cancer MDA-MB-231 cell death by inducing cell cycle arrest at G0/G1 and upregulating Bax显示文摘Objective:To determine the anti-proliferative activity of Abrus precatorius(A.precatorius)leaf extracts and their effect on cell death.Methods:A.precatorius leaves were extracted successively with hexane,ethyl acetate and methanol by Soxhlet extraction.Aqueous extract was prepared by decoction at 50 ℃.Extracts of A.precatorius leaves were used to treat selected cancer and normal cell lines for72 h.Furthermore,3-(4,5-dimethyl thiazol-2-yl)2,5-diphenyl tetrazolium bromide assay was performed to determine cell viability.Analysis of cell cycle arrest,apoptosis assay and apoptosis protein expressions were determined by flow cytometry.Results:Methanolic extract of A.precatorius leaves showed the lowest IC50 on MDA-MB-231 cells at(26.40±5.40)μg/mL.Flow cytometry analysis revealed that cell arrest occurred at G0/G1 phase and the apoptosis assay showed the occurrence of early apoptosis at 48 h in MDAMB-231 cells treated with methanolic extract of A.precatorius leaves.Methanolic extract of A.precatorius leaves induced apoptosis by upregulation of Bax,p53 and caspase-3 and downregulation of Bcl-2.Conclusions:Methanolic extract of A precatorius leaves promotes MDA-MB-231 cell death by inducing cell cycle arrest and apoptosis possibly via the mitochondrial-related pathway.Wan Suriyani Wan-Ibrahim Norzila Ismail Siti Farhanah Mohd-Salleh Aidy Irman Yajid Michael Pak-KaiWong Mohd Nizam Md Hashim 2019Asian Pacific Journal of Tropical Biomedicine2019,9,6:2
6In situ biosynthesized silver nanoparticle-incorporated synthesized zeolite A using Orthosiphon aristatus extract for in vitro antibacterial wound healing显示文摘The capability of synthesized zeolite A(SZ)to immobilize Ag ions(Ag-SZ)and Ag nanoparticles(AgNp-SZ)were comparatively studied.A novel approach of in situ biosynthesized AgNP-incorporated synthesized zeolite A(AgNp-SZ)was synthesized at an optimum volume of 0.4mL of the Orthosiphon aristatus(O.aristatus)leaves plant extract(5%)using an in situ approach.In comparison,Ag-Sz was produced by loading the synthesized zeolite with Ag ions.All synthesized materials were characterized for their morphologies and physicochemicalproperties.The characterization analysesvalidate thathe biosynthesized AgNP(<100 nm)using O.aristatus leaves extract was incorporated into the zeolite A.The antibacterial testing confirmed that these materials have antibacterial activity against Escherichia coliATCC11229 and Staphylococcus aureus ATCC 6538.MIC/MBC analysis demonstrated that in 0.9%saline solution,AgNP-SZ had higher antibacterial activity than Ag-Sz.The in vitro cell viability and migration assays were further examined towards human skin fibroblast cells HSF 1184.Results show that the materials are not cytotoxicto HSF1184,and the biosynthesized AgNP-SZ promotes cell migration and proliferation higher than Ag-SZ.This research proved that the biocompatible antibacterial wound healing agent of AgNP-SZ can be synthesized using an in situ approach where the reduction process of Ag ions in the zeolite Acan be performed using plant extract.Muhammad Hariz Asraf Nor Suriani Sani Craig Denver Williams Khairunadwa Jemon Nik Ahmad Nizam Nik Maliek 2022Particuology2022,20,8:2

15Pre-emptive live donor kidney transplantation-moving barriers to opportunities:An ethical,legal and psychological aspects of organ transplantation view显示文摘Live donor kidney transplantation(LDKT)is the optimal treatment modality for end stage renal disease(ESRD),enhancing patient and graft survival.Pre-emptive LDKT,prior to requirement for renal replacement therapy(RRT),provides further advantages,due to uraemia and dialysis avoidance.There are a number of potential barriers and opportunities to promoting pre-emptive LDKT.Significant infrastructure is needed to deliver robust programmes,which varies based on socio-economic standards.National frameworks can impact on national prioritisation of pre-emptive LDKT and supporting education programmes.Focus on other programme’s components,including deceased kidney transplantation and RRT,can also hamper uptake.LDKT programmes are designed to provide maximal benefit to the recipient,which is specifically true for pre-emptive transplantation.Health care providers need to be educated to maximize early LDKT referral.Equitable access for varying population groups,without socioeconomic bias,also requires prioritisation.Cultural barriers,including religious influence,also need consideration in developing successful outcomes.In addition,the benefit of pre-emptive LDKT needs to be emphasised,and opportunities provided to potential donors,to ensure timely and safe work-up processes.Recipient education and preparation for pre-emptive LDKT needs to ensure increased uptake.Awareness of the benefits of pre-emptive transplantation require prioritisation for this population group.We recommend an approach where patients approaching ESRD are referred early to pre-transplant clinics facilitating early discussion regarding pre-emptive LDKT and potential donors for LDKT are prioritized for work-up to ensure success.Education regarding preemptive LDKT should be the norm for patients approaching ESRD,appropriate for the patient’s cultural needs and physical status.Pre-emptive transplantation maximize benefit to potential recipients,with the potential to occur within successful service delivery.To fully embrace preemptive transplantation as the norm,investment in infrastructure,increased awareness,and donor and recipient support is required.David van Dellen Lisa Burnapp Franco Citterio Nizam Mamode Greg Moorlock Kristof van Assche Willij CZuidema Annette Lennerling Frank JMF Dor 2021World Journal of Transplantation2021,11,4:1
