1草菇废棉培养料的理化性质及木质纤维素变化分析显示文摘为探究草菇生产中产量不稳定的内在原因,本研究通过测定废棉培养料不同时期的理化性状、木质纤维素含量及降解酶活性,对草菇(Volvariella volvacea)低产和高产批次基质的降解情况进行了对比分析。结果表明:在培养料的整个发酵阶段,两批次的含水量和碳氮比均逐渐降低,灰分含量、含氮量则逐渐升高,同时高产批次的羧甲基纤维素酶(CMCase)和木聚糖酶活性一直显著高于低产批次。发酵结束时,高产批次的碳氮比较低产批次高4.60,p H值和含水量无明显差异。草菇生长发育阶段,高、低产两批次碳氮比的下降幅度分别为25.71%和12.77%,且前者纤维素相对含量的下降幅度显著大于后者。原基形成时低产批次的漆酶(Laccase)活性是高产批次的2.07倍,采收时低产批次子实体的平均单重较高产批次高6.66 g。本研究对草菇栽培基质的科学优化及工厂化生产进程的推进具有重要参考意义。刘顺杰 张亚茹 陈明杰 马丹丹 Kakumyan Pattana 郭倩 赵妍 2021分子植物育种2021,19,7:4
2草菇锰超氧化物歧化酶基因异源表达条件优化及其耐胁迫功能分析显示文摘根据草菇(Volvariella volvacea)的锰超氧化物歧化酶(Mn-SOD)基因序列,构建重组质粒pBAR GPE1/Mn-SOD,转化大肠杆菌(Escherichia coli)Stbl3菌株,采用单因素实验对诱导剂IPTG浓度(0、0.2、0.5、1.0、1.5、2.0 mmol·L^-1)、诱导培养时间(0、2、4、6、8、10 h)及MnCl2浓度(0、0.2、0.5、1.0、1.5、2.0 mmol·L^-1)进行优化,以获得最优表达条件。在最优条件下分别进行耐热性、耐冷性及耐盐性分析,测定重组菌pBAR GPE1/Mn-SOD和对照菌pBAR GPE1的SOD酶活力及生长情况。结果表明:最优表达条件为IPTG浓度1 mmol·L^-1,诱导培养时间6 h,不需要添加MnCl2;在最优条件下,与对照菌相比,发现重组菌的SOD酶活力显著提高,生长情况较好,提高了转化菌株的耐热性、耐冷性和耐盐性。杨焕玲 游华芳 赵妍 KAKUMYAN Pattana 常婷婷 余昌霞 陈明杰 2020食用菌学报2020,27,4:3

4Novel ABCB11 mutations in a Thai infant with progressive familial intrahepatic cholestasis显示文摘Progressive familial intrahepatic cholestasis(PFIC) type 2 is caused by mutations in ABCB11,which encodes bile salt export pump(BSEP).We report a Thai female infant who presented with progressive cholestatic jaundice since 1 mo of age,with normal serumγ-glutamyltransferase.Immunohistochemical staining of the liver did not demonstrate BSEP along the canaliculi,while multidrug resistance protein 3 was expressed adequately.Novel mutations in ABCB11,a four-nucleotide deletion in exon 3,c.90_93delGAAA,and a single-nucleotide insertion in exon 5,c.249_250insT, were identified,with confirmation in her parents. 17A novel aeration strategy in repeated-batch fermentation for efficient ethanol production from sweet sorghum juice显示文摘To improve the efficiency of ethanol production in a batch fermentation from sweet sorghum juice under a very high gravity(VHG)condition(~290 g/L of total sugar)by Saccharomyces cerevisiae NP01,repeatedbatch fermentation under an aerated condition(2.5 vvm for the first 4 h during every cycle)was done in a5-L fermenter.The average ethanol concentration(P),productivity(Qp)and yield(Yp/s)for five successive cycles were 112.31 g/L,1.55 g/L·h^-1 and 0.44,respectively with 80.97%sugar consumption.To complete sugar consumption,the total sugar of the juice was reduced to a high gravity(HG)level(~240 g/L).The results showed that yeast extract was not necessary for ethanol production,and aeration during every other cycle i.e.,alternating cycles,was sufficient to promote both yeast growth and ethanol production.The average P,Qpand Yp/svalues for eight successive cycles with aeration during alternating cycles were97.58 g/L,1.98 g/Láh and 0.41,respectively with 91.21%sugar consumption.The total fatty acids in the yeast cells under the aerated condition were^50%higher than without aeration,irrespective the initial sugar concentration,whereas the ergosterol contents under aeration condition were^29%to 49%higher than those without aeration.Niphaphat Phukoetphim Naulchan Khongsay Pattana Laopaiboon Lakkana Laopaiboon 2019Chinese Journal of Chemical Engineering2019,27,7:1
