| 1 | 他汀、高密度脂蛋白胆固醇与冠状动脉粥样硬化消退显示文摘背景:他汀类药物可以降低低密度脂蛋白胆固醇(low—density lipoprotein cholesterol,LDL—C)水平并减缓冠状动脉粥样硬化的进展。然而,对于他汀类药物所致高密度脂蛋白胆固醇(high—density lipoprotein cholesterol,HDL—C)变化与疾病进展的关系,目前仍无相关数据描述。目的:了解LDL—C和HDL—C水平变化与动脉粥样硬化的相互关系。设计、地点及患者:对4项前瞻性随机试验(1999~2005年在美国、北美、欧洲和澳大利亚进行)的原始数据进行事后分析。其中1455例经血管造影证实的冠心病患者在接受他汀治疗(18个月或24个月)时进行了系列血管内超声扫描。所有超声分析均在相同的核心实验室中进行。主要观测指标:脂蛋白水平变化与冠状动脉粥样斑块体积的相互关系。结果:他汀治疗期间,LDL—C平均(SD)水平从124.0(38.3)mg,/dL(3.2[0.99]mmol/L)降至87.5(28.8)mg,/dL(2.3[0.75]mmol/L)(下降23.5%;P〈0.001),HDL—C水平从42.5(11.0)mg/dL(1.1[0.28]mmol/L)升至45.1(11.4)mg/dL(1.2[0.29]mmol/L)(上升7.5%;P〈0.001)。LDL—C与HDL—C比值从平均(SD)3.0(1.1)降至2.1(0.9)(下降26.7%;P〈0.001)。这些变化同时伴随平均(SD)百分粥样斑块体积的增加(从39.7%[9.8%]增至40.1%[9.7%])(增加0.5%[3.9%];P=0.001)以及平均(SD)总粥样斑块体积的减小(2.4[23.6]mm^3;P〈0.001)。在单变量分析中,LDL—C、总胆固醇、非HDL胆固醇、载脂蛋白B以及载脂蛋白B与载脂蛋白A-1比值平均水平和治疗引起的变化与动脉粥样硬化进展速率显著相关,而治疗所致HDL—C变化则与动脉粥样斑块体积负相关。在多变量分析中,平均LDL—C水平(β系数,0.11[95%可信区间,0.07~0.15])以及HDL—C升高(β系数,-0.26[95%可信区间,-0.41--0.10])依然为动脉粥样硬化消退的独立预测因子。动脉硬化显著消退(动脉粥样斑块体积减少≥5%)见于治疗期间LDL—C水平低于均值(87.5mg/dL)和HDL—C升高百分比大于均值(7.5%;P〈0.001)的患者。临床事件的发生未见显著差异。结论:在LDL—C显著降低以及HDL上升〉7.5%时,他汀治疗与冠状动脉粥样硬化消退相关。这些结果提示他汀治疗的获益源自致动脉粥样硬化脂蛋白的降低以及HDL.C的升高。尽管血脂水平的这些变化与动脉粥样硬化消退相关,但是后者是否可以转化为临床事件的显著减少,并改善临床预后仍有待进一步研究确定。 | Stephen J. | 4 | Enhancement of Growth and Grain Yield of Rice in Nutrient Deficient Soils by Rice Probiotic Bacteria显示文摘Plant associated bacteria are promising alternatives to chemical fertilizers for plant growth and yield improvement in an eco-friendly manner.In this study,rice associated bacteria were isolated and assessed for mineral phosphate solubilization and indole-3-acetic acid(IAA) production activity in vitro.Six promising strains,which were tentatively identified as phylotaxon Pseudochrobactrum sp.(BRRh-1),Burkholderia sp.(BRRh-2),Burkholderia sp.(BRRh-3),Burkholderia sp.(BRRh-4),Pseudomonas aeruginosa(BRRh-5 and BRRh-6) based on their 16 S rRNA gene phylogeny,exhibited significant phosphate solubilizing activity in National Botanical Research Institute phosphate growth medium,and BRRh-4 displayed the highest phosphate solubilizing activity,followed by BRRh-5.The p H of the culture broth declined,resulting in increase of growth rate of bacteria at p H 7,which might be due to organic acid secretion by the strains.In presence of L-tryptophan,five isolates synthesized IAA and the maximum IAA was produced by BRRh-2,followed by BRRh-1.Application of two most efficient phosphate solubilizing isolates BRRh-4 and BRRh-5 by root dipping(colonization) of seedling and spraying at the flowering stage significantly enhanced the growth and grain yield of rice variety BRRI dhan-29.Interestingly,application of both strains with 50% of recommended nitrogen,phosphorus and potassium fertilizers produced equivalent or higher grain yield of rice compared to the control grown with full recommended fertilizer doses,which suggests that these strains may have the potential to be used as bioinoculants for sustainable rice production. | Md Mohibul Alam KHAN Effi HAQUE Narayan Chandra PAUL Md Abdul KHALEQUE Saleh M. | 5 | Rapid Detection of Wheat Blast Pathogen Magnaporthe oryzae Triticum Pathotype Using Genome-Specific Primers and Cas12a-mediated Technology显示文摘Wheat blast,caused by the fungus Magnaporthe oryzae Triticum(MoT)pathotype,is a devastating disease persistent in South America and Bangladesh.Since MoT generally fails to cause visual symptoms in wheat until the heading stage when the infection would have advanced,disease control by fungicide application solely based on the detection of visual symptoms is ineffective.To develop an accurate and sensitive method to detect MoT at the seedling and vegetative stages for disease control,we sequenced the genomes of two MoT isolates from Brazil and identified two DNA fragments,MoT-6098 and MoT-6099,that are present in the MoT genome but not in the genome of the rice-infecting Magnaporthe oryzae Oryzae(MoO)pathotype.Using polymerase chain reaction(PCR),we confirmed the specificity of the two markers in 53 MoT and MoO isolates from South America and Bangladesh.To test the efficiency of the two markers,we first established a loop-mediated isothermal amplification(LAMP)method to detect MoT at isothermal conditions,without the use of a PCR machine.Following this,we used the Cas12a protein and guide RNAs(gRNAs)to target the MoT-6098 and MoT-6099 sequences.The activated Cas12a showed indiscriminate single-stranded deoxyribonuclease(ssDNase)activity.We then combined targetdependent Cas12a ssDNase activation with recombinase polymerase amplification(RPA)and nucleic acid lateral flow immunoassay(NALFIA)to develop a method that accurately,sensitively,and cost-effectively detects MoT-specific DNA sequences in infected wheat plants.This novel technique can be easily adapted for the rapid detection of wheat blast and other important plant diseases in the field. | Houxiang Kang Ye Peng Kangyu Hua Yufei Deng Maria Bellizzi Dipali Rani Gupta Nur Uddin Mahmud Alfredo S.Urashima Sanjoy Kumar Paul Gary Peterson Yilin Zhou Xueping Zhou Md Tofazzal Islam Guo-Liang Wang | 2021 | Engineering2021，7，9： | 4 |

| 10 | Glutamate triggers the expression of functional ionotropic and metabotropic glutamate receptors in mast cells显示文摘Mast cells are emerging as players in the communication between peripheral nerve endings and cells of the immune system.However,it is not clear the mechanism by which mast cells communicate with peripheral nerves.We previously found that mast cells located within healing tendons can express glutamate receptors,raising the possibility that mast cells may be sensitive to glutamate signaling.To evaluate this hypothesis,we stimulated primary mast cells with glutamate and showed that glutamate induced the profound upregulation of a panel of glutamate receptors of both the ionotropic type(NMDAR1,NMDAR2A,and NMDAR2B)and the metabotropic type(mGluR2 and mGluR7)at both the mRNA and protein levels.The binding of glutamate to glutamate receptors on the mast cell surface was confirmed.Further,glutamate had extensive effects on gene expression in the mast cells,including the upregulation of proinflammatory components such as IL-6 and CCL2.Glutamate also induced the upregulation of transcription factors,including Egr2,Egr3 and,in particular,FosB.The extensive induction of FosB was confirmed by immunofluorescence assessment.Glutamate receptor antagonists abrogated the responses of the mast cells to glutamate,supporting the supposition of a functional glutamate–glutamate receptor axis in mast cells.Finally,we provide in vivo evidence supporting a functional glutamate–glutamate receptor axis in the mast cells of injured tendons.Together,these findings establish glutamate as an effector of mast cell function,thereby introducing a novel principle for how cells in the immune system can communicate with nerve cells. | Md Abdul Alim Mirjana Grujic Paul W.Ackerman Per Kristiansson Pernilla Eliasson Magnus Peterson Gunnar Pejler | 2021 | Cellular & Molecular Immunology2021，18，10： | 2 |
