| 2 | 滇池中溶藻细菌的分离鉴定及其溶藻效应显示文摘【背景】藻类水华或赤潮在世界范围内频发,带来各种危害,亟需找到有效途径控制水华或赤潮。溶藻细菌具有杀死藻类控制藻类生物量的能力,可以作为防治水华和赤潮的有效工具。【目的】分离并鉴定滇池中的铜绿微囊藻(Microcystisaeruginosa)及其溶藻细菌,对溶藻菌作用于铜绿微囊藻的溶藻效应进行研究,初步了解其溶藻特性与溶藻机制。【方法】采用LB平板稀释涂布,再经多次划线分离纯化细菌,测定16SrRNA基因序列以鉴定细菌种类;采用毛细管分离的方法分离铜绿微囊藻,并测定其cpcBA基因序列以鉴定蓝藻种类;采用热乙醇法提取叶绿素a,从而计算溶藻效率;基于过氧化氢酶(CAT)、还原型谷胱甘肽(GSH)和丙二醛(MDA)探究藻细胞在溶藻菌处理下的抗氧化系统响应。【结果】共分离获得11株微囊藻和17株针对铜绿微囊藻的高效溶藻菌。选取其中一株生长速度最快的铜绿微囊藻DCM4和一株溶藻效果最好的溶藻菌Sp37 (Bacillus siamensis)进行后续研究。Sp37对DCM4的4 d溶藻率达到92.4%±1.5%,且对微囊藻属的水华微囊藻(M. flos-aquae)和惠氏微囊藻(M.wesenbergii)均有溶藻效果,而对绿藻没有溶藻效果。Sp37的原菌液和无菌滤液对DCM4的4d溶藻率分别为86.8%±4.3%和81.1%±2.2%,两者没有显著差异(P>0.05)。Sp37菌体对DCM4的溶藻率为25.4%±7.3%。Sp37无菌滤液经不同温度和pH处理之后的溶藻率与未经处理的无菌滤液的溶藻率无明显差异。Sp37无菌滤液处理藻细胞会使藻细胞的CAT、GSH和MDA含量发生变化。【结论】菌株Sp37对铜绿微囊藻DCM4具有高效的溶藻作用,而且对微囊藻属具有一定的溶藻特异性。Sp37是通过分泌胞外物质间接溶藻,且溶藻物质具有热稳定性和酸碱稳定性。Sp37无菌滤液处理藻细胞会触发藻细胞抗氧化系统,并且会损伤藻细胞膜。Sp37无菌滤液很可能是通过对藻细胞造成氧化胁迫,最终导致藻细胞死亡的。 | 王琪 Paulina Simon 刘锦钰 迟玉新 代先祝 杜红霞 张晓辉 五十岚泰夫 杨彩云 罗锋 | 2018 | 微生物学通报2018,45,12: | 8 |
| 3 | Comprehensive Approach for Porous Materials Analysis Using a Dedicated Preprocessing Tool for Mass and Heat Transfer Modeling显示文摘The paper presents a comprehensive, newly developed software – poROSE(poROus materials examination SoftwarE) for the qualitative and quantitative assessment of porous materials and analysis methodologies developed by the authors as a solution for emerging challenges. A low porosity rock sample was analyzed and thanks to the developed and implemented methodologies in poROSE software, the main geometrical properties were calculated. A tool was also used in preprocessing part of the computational analysis to prepare a geometrical representation of the porous material. The basic functions as elimination of blind pores in the geometrical model were completed and the geometrical model was exported for CFD software. As a result, it was possible to carry out calculations of the basic properties of the analyzed porous material sample. The developed tool allows to carry out quantitative and qualitative analysis to determine the most important properties characterized porous materials. In presented tool the input data can be images from X-ray computed tomography(CT), scanning electron microscope(SEM) or focused ion beam with scanning electron microscope(FIB-SEM) in grey level. A geometric model developed in the proper format can be used as an input to modeling mass, momentum and heat transfer, as well as, in strength or thermo-strength analysis of any porous materials. In this example, thermal analysis was carried out on the skeleton of rock sample. H. pylori has very high rates of infection in human populations, and it is estimated that over 50% of the world population is infected. Recently, certain extra-gastric manifestations, linked to H. pylori infection, have been widely investigated. Noteworthy, a growing body of evidences supports an association between H. pylori infection with lung cancer. | 7 | H2O2诱导ARPE-19细胞损伤模型的建立及蓝莓花青素的保护作用显示文摘目的建立H_2O_2诱导视网膜色素上皮细胞ARPE^(-1)9(acute retinal pigment epithelial^(-1)9)细胞损伤模型,研究蓝莓花青素对细胞的保护作用。方法采用MTT法测定细胞存活率,免疫荧光法测定细胞活性氧(reactive oxygen species,ROS)水平。以浓度0~3 200μmol·L^(-1)的H_2O_2为诱导剂,刺激细胞2、6、24 h,用1、5、10 mg·L^(-1)花青素提取物(blueberry anthocyanin extract,BAE)预处理细胞6、24 h,比较对细胞的保护作用,确定H_2O_2的浓度及诱导时间。通过ARPE^(-1)9细胞损伤模型的建立,研究蓝莓花青素对细胞的保护作用。结果以800μmol·L^(-1)H_2O_2刺激2 h为条件,建立ARPE^(-1)9细胞损伤模型,细胞存活率为63.69%。5 mg·L^(-1)蓝莓花青素提取物、锦葵色素(malvidin,Mv)及其葡萄糖苷(malvidin-3-glucoside,Mv-3-glc)和半乳糖苷(malvidin-3-galactoside,Mv-3-gal)预处理6 h后,细胞存活率明显提高(P<0.01),分别达到86.57%、115.72%、98.15%、127.97%;细胞中ROS水平明显下降(P<0.05),依次减少52.38%、95.38%、77.55%、116.09%。结论蓝莓花青素可以有效抑制H_2O_2诱导产生的视网膜细胞氧化损伤,其中主成分锦葵色素及其糖苷发挥重要作用。 | 吴寒 HUTABARAT Ruth Paulina 黄午阳 | 2018 | 中国药理学通报2018,34,2: | 5 |

| 9 | Impact of Helicobacter pylori on the healing process of the gastric barrier显示文摘AIM To determine the impact of selected well defined Helicobacter pylori(H. pylori) antigens on gastric barrier cell turnover.METHODS In this study,using two cellular models of gastric epithelial cells and fibroblasts,we have focused on exploring the effects of well defined H. pylori soluble components such as glycine acid extract antigenic complex(GE),subunit A of urease(Ure A),cytotoxin associated gene A protein(Cag A) and lipopolysaccharide(LPS) on cell turnover by comparing the wound healing capacity of the cells in terms of their proliferative and metabolic activity as well as cell cycle distribution. Toxic effects of H. pylori components have been assessed in an association with damage to cell nuclei and inhibition of signal transducer and activator of transcription 3(STAT3) phosphorylation. RESULTS We showed that H. pylori GE,Cag A and Ure A promoted regeneration of epithelial cells and fibroblasts,which is necessary for effective tissue healing. However,in vivo increased proliferative activity of these cells may constitute an increased risk of gastric neoplasia. In contrast,H. pylori LPS showed a dose-dependent influence on the process of wound healing. At a low concentration(1 ng/m L) H. pylori LPS accelerated of healing epithelial cells,which was linked to significantly enhanced cell proliferation and MTT reduction as well as lack of alterations in cell cycle and downregulation of epidermal growth factor(EGF) production as well as cell nuclei destruction. | 10 | Plant Growth-Promoting Traits in Rhizobacteria of Heavy Metal-Resistant Plants and Their Effects on Brassica nigra Seed Germination显示文摘Phytoremediation is an emerging technology that uses plants and their associated microbes to clean up pollutants from the soil, water, and air. In order to select the plant growth-promoting rhizobacteria(PGPR) for phytoremediation of heavy metal contamination, 60 bacterial strains were isolated from the rhizosphere of two endemic plants, Prosopis laevigata and Spharealcea angustifolia, in a heavy metal-contaminated zone in Mexico. These rhizobacterial strains were characterized for the growth at different pH and salinity, extracellular enzyme production, solubilization of phosphate, heavy metal resistance, and plant growth-promoting(PGP) traits, including production of siderophores and indol-3-acetic acid(IAA). Overall, the obtained rhizobacteria presented multiple PGP traits. These rhizobacteria were also resistant to high levels of heavy metals(including As as a metalloid)(up to 480 mmol L(-1)As(V), 24 mmol L(-1)Pb(Ⅱ), 21 mmol L(-1)Cu(Ⅱ), and 4.5 mmol L(-1)Zn(Ⅱ)). Seven rhizobacterial strains with the best PGP traits were identified as members of Alcaligenes, Bacillus, Curtobacterium, and Microbacterium, and were selected for further bioassay.The inoculation of Brassica nigra seeds with Microbacterium sp. CE3R2, Microbacterium sp. NE1R5, Curtobacterium sp. NM1R1,and Microbacterium sp. NM3E9 facilitated the root development; they significantly improved the B. nigra seed germination and root growth in the presence of heavy metals such as 2.2 mmol L(-1)Zn(Ⅱ). | 13 | Outcomes of critically ill cancer patients with Acinetobacter baumannii infection显示文摘AIM: To describe the intensive care unit(ICU) outcomes of critically ill cancer patients with Acinetobacter baumannii(AB) infection.METHODS: This was an observational study that included 23 consecutive cancer patients who acquired AB infections during their stay at ICU of the National Cancer Institute of Mexico(INCan), located in Mexico City. Data collection took place between January 2011, and December 2012. Patients who had AB infections before ICU admission, and infections that occurred during the first 2 d of ICU stay were excluded. Data were obtained by reviewing the electronic health record of each patient. This investigation was approved by the Scientific and Ethics Committees at INCan. Because of its observational nature, informed consent of the patients was not required.RESULTS: Throughout the study period, a total of 494 critically ill patients with cancer were admitted to the ICU of the INCan, 23(4.6%) of whom developed AB infections. Sixteen(60.9%) of these patients had hematologic malignancies. Most frequent reasons for ICU admission were severe sepsis or septic shock(56.2%) and postoperative care(21.7%). The respiratory tract was the most frequent site of AB infection(91.3%). The most common organ dysfunction observed in our group of patients were the respiratory(100%), cardiovascular(100%), hepatic(73.9%) and renal dysfunction(65.2%). The ICU mortality of patients with 3 or less organ system dysfunctions was 11.7%(2/17) compared with 66.6%(4/6) for the group of patients with 4 or more organ system dysfunctions(P = 0.021). Multivariate analysis identified blood lactate levels(BLL) as the only variable independently associated with inICU death(OR = 2.59, 95%CI: 1.04-6.43, P = 0.040). ICU and hospital mortality rates were 26.1% and 43.5%, respectively.CONCLUSION: The mortality rate in critically ill patients with both HM, and AB infections who are admitted to the ICU is high. | 14 | Perovskite type lanthanum manganite:Morpho-structural analysis and electrical investigations显示文摘Perovskite-type oxides of LaMnO_3 were synthesized by means of the sol-gel method, in the presence of citric acid as gelling agent. The precursors used were Mn(NO_3)_2·H_2O, La_2O_3, and NaOH,mixed in the stoichiometric ratio to obtain perovskite materials. The obtained gel was heat-treated at 400, 600 and 800 ℃ respectively, for 6 h. X-ray diffraction and FT-IR spectroscopy were used to analyze the phase transformation as a function of temperature, and the Rietveld refinement was used in order to characterize the materials obtained structurally. The average crystallite size of the products was calculated from XRD data and the average particle size was measured from the TEM micrographs.
