| 2 | The E3 Ubiquitin Ligase gp78 Protects against ER Stress in Zebrafish Liver显示文摘Enhanced endoplasmic reticulum(ER)-associated protein degradation(ERAD) activity by the unfolded protein response(UPR) represents one of the mechanisms for restoring ER homeostasis.In vitro evidence indicates that the mammalian gp78 protein is an E3 ubiquitin ligase that facilitates ERAD by polyubiquitinating and targeting proteins for proteasomal degradation under both physiologic and stress conditions.However,the in vivo function of gp78 in maintaining ER protein homeostasis remains untested.Here we show that like its mammalian counterpart,the zebrafish gp78 is also an E3 ubiquitin ligase as revealed by in vitro ubiquitination assays.Expression analysis uncovered that gp78 is highly expressed in several organs,including liver and brain,of both larval and adult fish.Treatment of larvae or adult fish with tunicamycin induces ER stress and upregulates the expression of several key components of the gp78 ERAD complex in the liver.Moreover,liver-specific overexpression of the dominant-negative form of gp78(gp78-R2M) renders liver more sensitive to tunicamycin-induced ER stress and enhances the expression of sterol response element binding protein(Srebp)-target genes,which was largely suppressed in fish overexpressing wild-type gp78.Together,these data indicate that gp78 plays a critical role in protecting against ER stress in liver. | Zhiliang Chen Petek Ballar Yu Fu Jia Luo Shaojun Du Shengyun Fang | 2014 | Journal of Genetics and Genomics2014，41，7： | 3 |
| 3 | 海兰W-80父母代种鸡产蛋曲线数学模型分析研究显示文摘为探究海兰W-80父母代种鸡在我国饲养环境下的产蛋规律,本实验分别测定了3个种鸡场各10827、10147、21204只海兰W-80父母代种鸡全期产蛋性能,分别采用伍德模型(Woodmodel)、分室模型(McMillan model)和杨宁模型拟合其产蛋率曲线,使用Gompertz、Logistic和Von Bertalanffy模型拟合其累计产蛋数曲线,根据R^(2)、赤池信息准则(AIC)和贝叶斯信息准则(BIC)选择最优模型,并与饲养标准进行产蛋性能比较。结果显示,曲线拟合模型F检验均差异显著,可用于海兰W-80父母代种鸡产蛋率和累计产蛋数拟合;产蛋率拟合杨宁模型R^(2)最高(R^(2)=0.987~0.994),分室模型次之(R2=0.927~0.977),伍德模型拟合度最低(R2=0.509~0.652),且AIC与BIC指数依次为杨宁模型<分室模型<伍德模型。Gompertz、Logistic、Von Bertalanffy模型拟合3个种鸡场累计产蛋数曲线R^(2)分别为0.996~0.997、0.989~0.990、0.998~0.999,且VonBertalanffy模型AIC与BIC指数最低。本实验中海兰W-80父母代种鸡开产较饲养标准早4~11 d,70周龄累计产蛋数高于标准上限5~12个,但92%产蛋率高峰持续时间较短。综上,杨宁模型和Von Bertalanffy模型分别适用于海兰W-80父母代种鸡产蛋率和累计产蛋数的拟合,可评估和预测其产蛋规律。此外,应避免种鸡开产过早,以防产蛋高峰持续时间缩短,影响全期产蛋性能。 | 曾丹 闫文亮 Petek Settar Turkmut 李秀业 王斌 韩晓飞 石玉祥 高亚辉 | 2021 | 中国畜牧杂志2021，57，10： | 2 |
| 4 | 十五肽BPC157减弱慢性苯丙胺诱导的行为障碍(英文)显示文摘AIM: To investigate the effect of pentadecapeptide BPC 157 on chronic exposure to amphetamine in rats, particularly the changes commonly referred in chronic amphetamine studies as tolerance ( lesser grade of stereotyped behavior, without increased excitability) and reverse tolerance (ie, prominent stereotyped behavior and heightened startle response upon late amphetamine challenges). METHODS: After initial application (initial single dose-regimen), amphetamine (10 mg/kg, ip) was given once daily till d 5 ( continuous administration-regimen), and thereafter on d 8, 16, and 46 (intermittent administration regimen). For stereotyped behavior and heightened startle response the observation period was 120 min after amphetamine application, and each animal was observed for 10 s in 5 min intervals. Pentadecapeptide BPC 157 (10 μg/kg or 10 ng/kg, ip) or saline (5.0 mL/kg, ip) were given only at the beginning of the experiment, simultaneously with the initial dose of amphetamine.
