1Differentiation of embryonic stem cells into corneal epithelium显示文摘Our project was to determine whether embryonic stem (ES) cells could be induced to differentiate into corneal epithelia by superficial corneoscleral limbal stroma. To achieve this goal, ES-GFP cell line D3 was pre-induced by retinoic acid (RA). The pre-induced cells were seeded on deepithelialized superficial corneoscleral slices (SCSS) to form a monolayer, and di-vided into three groups. Group 1 was cultured and passaged in vitro for direct detection. Group 2 was exposed to air-liquid interfaces for 10 days and implanted into the subcutaneous layer of nude mice for 2 weeks for further induction in vivo. Group 3 was cultured in vitro without any in-ducing factors for control. There were no teratomas found in nude mice which were implanted with differentiated ES cells after two weeks. The differentiated cells showed an appearance of epithelia both in vitro and in vivo. Expression of CK3, P63 and PCNA was detected by immuno-histochemical staining in the differentiated cells in group 1 and 2. Microvillis and zonula oc-cludens were observed on the surface of the differentiated cells under an electron microscope. In the control group, ES cells differentiated freely without any inducing factors. Most cells were shed and formed a neuronal dendrite-like structure, and a minority of cells appeared polymorphic. These results demonstrate that ES cells can differentiate into corneal epithelia on the surface of SCSS under the controlled condition. 2Variation of activation energy determined by a modified Arrhenius approach:Roles of dynamic recrystallization on the hot deformation of Ni-based superalloy显示文摘The hot deformation behaviors of Ni18 Cr9 Co9 Fe5 Nb3 Mo superalloy were explored in the formation temperature range free ofγ’phase with various strain rates applied.The hot deformation behaviors are initially modeled with Arrhenius equation which gives an average activation energy of 581.1 kJ mol^(-1).A modified Arrhenius approach,including the updated Zener-Hollomon parameter is proposed to consider the change of activation ene rgy under different deformation conditions which turns out a relatively accurate computation for activation energy of hot deformation,i.e.,the standard variance for modified model calculated in the covered deformation condition is just 35.4%of that for Arrhenius equation.The modified model also proposes a map for activation ene rgy which ranges from 571.5-589.0 kJ mol^(-1)for various deformation conditions.Microstructural features of the representative superalloy specimens were characterized by electron backscattered diffraction(EBSD)techniques in order to clarify the influence of activation energy on the microstructural formation.It is found that the Ni-based superalloy samples with higher activation energy are promoted by the degree of dynamic recrystallization which suggests that the rise in activation energy gives either a better recrystallization rate or finer grains.Peiru Yang Chenxi Liu Qianying Guo Yongchang Liu 2021Journal of Materials Science & Technology2021,,13:8
3Controlled synthesis of high-quality W-Y2O3 composite powder precursor by ascertaining the synthesis mechanism behind the wet chemical method显示文摘As an emerging preparation technology,wet chemical method has been employed widely to produce lots of alloy materials such as W and Mo based alloys,owing to its unique technical advantages.Ascertaining the synthesis mechanism behind wet chemical method is indispensable for controlled synthesis of highquality W-Y2 O3 composite powder precursor.The co-deposition mechanism of yttrium and tungsten component behind the wet chemical method of preparing yttrium-doped tungsten composite nanopowder was investigated systematically in this work.A series of co-deposited composite powders fabricated under different acidity conditions were used as research targets for investigating the effect of surface composition and structure on co-deposition efficiency.It was found that white tungstic acid has more W-OH bonds and much higher co-deposition efficiency with Y^3+ions than yellow tungstic acid.It is illustrated that the coordination reaction between W-OH bonds on tungstic acid particles and Y^3+ions brings the co-deposition of yttrium and tungsten component into being.Through displacing H^+ions in W-OH bonds,Y^3+ions can be adsorbed on the surface of or incorporated into tungstic acid particles in form of ligand.Consequently,to control and regulate Y2 O3 content in powder precursor accurately,H^+ion concentration in wet chemical reaction should be in range of 0.55-2.82 mol L^-1 to obtain white tungstic acid.Besides,H^+ion concentration also has prominent effect on the grain size and morphology of reduced powder precursor.The optimal value should be around 1.58 mol L^-1,which can lead to minimum W grain size(about 17 nm) without bimodal structure.The chemical mechanism proposed in this work could produce great sense to preparation of high-quality precursor for sintering high-performance Y2 O3 dispersion strengthened W based alloys.Our work may also shed light on the approach to exploit analogous synthesis mechanism in other alloy systems.Zhi Dong Nan Liu Weiqiang Hu Zongqing Ma Chong Li Chenxi Liu Qianying Guo Yongchang Liu 2020Journal of Materials Science & Technology2020,36,1:7
4Formal analysis of TPM2.0 key management APIs显示文摘The trusted platform module(TPM),a system component implemented on physical resources,is designed to enable computers to achieve a higher level of security than the security level that it is possible to achieve by software alone.For this reason,the TPM provides a way to store cryptographic keys and other sensitive data in its memory,which is shielded from access by any entity other than the TPM.Users who want to use those keys and data to achieve some security goals are restricted to interact with the TPM through its APIs defined in the TPM specification.Therefore,whether the TPM can provide Protected Capabilities it claimed depends to a large extent on the security of its APIs.In this paper,we devise a formal model,which is accessible to a fully mechanized analysis,for the key management APIs in the TPM2.0 specification.We identify and formalize security properties of these APIs in our model and then successfully use the automated prover Tamarin to obtain the first mechanized analysis of them.The analysis shows that the key management subset of TPM APIs preserves the secrecy of non-duplicable keys for unbounded numbers of fresh keys and handles.The analysis also reports that the key duplication mechanism,used to duplicate a key between two hierarchies,is vulnerable to impersonation attacks,which enable an adversary to recover the duplicated key of the originating hierarchy or import his own key into the destination hierarchy.Aiming at avoiding these vulnerabilities,we proposean approach,which restricts the originating and destination TPMs to authenticate each other’s identity during duplication.Then we formally demonstrate that our approach maintains the secrecy of duplicable keys when they are duplicated.Qianying Zhang Shijun Zhao Yu Qin Dengguo Feng 2014Chinese Science Bulletin2014,59,32:5
5Aquaporin 5 is degraded by autophagy in diabetic submandibular gland显示文摘Autophagy is a catabolic process which is involved in the development of many diseases including diabetes mellitus and its complications. Hyposalivation is a common complication of diabetes mellitus, whereas its mechanism remains unclear. Here, we observed that the stimulated salivary flow rate of SMG was significantly decreased in db/db mice, a diabetic mice model. The expressions of aquaporin 5(AQP5), a water channel protein, were decreased, whereas the m RNA level of AQP5 was increased in SMGs of both diabetic patients and mice. Under transmission electron microcope, more autophagosomes were detected in diabetic SMGs. Expressions of autophagy related proteins LC3 II, Beclin-1 and ATG5 were increased, meanwhile autophagy substrate p62 was decreased in SMGs of diabetic patients and mice, indicating that autophagy was activated in diabetic SMG.Double immunofluorescence staining showed that the colocalization of AQP5 and LC3 was increased in SMGs of diabetic mice.In cultured SMG-C6 cells, high glucose(HG), but not high osmotic pressure, reduced AQP5 protein expression and induced autophagy. Moreover, inhibition of autophagy by 3-methyladenin, an autophagy inhibitor, or by autophagy-related gene 5 siRNA, decreased HG-induced AQP5 reduction in SMG-C6 cells. Additionally, the expression of p-p85, p-Akt and p-mTOR were decreased in HG-treated SMG-C6 cells. Pretreatment with 740 Y-P, a PI3 K agonist, significantly suppressed HG-induced autophagy and AQP5 degradation. Taken together, these results indicate that autophagy plays a crucial role in AQP5 degradation in diabetic SMG via PI3 K/Akt/mTOR signaling pathway, which contributes to the dysfunction of diabetic SMG. 6High-throughput screening identifies established drugs as SARS-CoV-2 PLpro inhibitors显示文摘A new coronavirus(SARS-CoV-2)has been identified as the etiologic agent for the COVID-19 outbreak.Currently,effective treatment options remain very limited for this disease;therefore,there is an urgent need to identify new anti-COVID-19 agents.In this study,we screened over 6,000 compounds that included approved drugs,drug candidates in clinical trials,and pharmacologically active compounds to identify leads that target the SARS-CoV-2 papain-like protease(PLpro).Together with main protease(Mpro),PLpro is responsible for processing the viral replicase polyprotein into functional units.There-fore,it is an attractive target for antiviral drug develop-ment.Here we discovered four compounds,YM155,cryptotanshinone,tanshinone I and GRL0617 that inhibit SARS-CoV-2 PLpro with IC50 values ranging from 1.39 to 5.63 pmol/L.These compounds also exhibit strong antiviral activities in cell-based assays.YM155,an anti-cancer drug candidate in clinical trials,has the most potent antiviral activity with an EC50 value of 170 nmol/L.In addition,we have determined the crystal structures of this enzyme and its complex with YM155,revealing a unique binding mode.YM155 simultaneously targets three'hot'spots on PLpro,including the substrate-binding pocket,the interferon stimulating gene product 15(ISG15)binding site and zinc finger motif.Our results demonstrate the efficacy of this screening and repur-posing strategy,which has led to the discovery of new drug leads with clinical potential for COVID-19 treatments.Yao Zhao Xiaoyu Du Yinkai Duan Xiaoyan Pan Yifang Sun Tian You Lin Han Zhenming Jin Weijuan Shang Jing Yu Hangtian Guo Qianying Liu Yan Wu Chao Peng Jun Wang Chenghao Zhu Xiuna Yang Kailin Yang Ying Lei Luke W.Guddat Wenqing Xu Gengfu Xiao Lei Sun Leike Zhang Zihe Rao Haitao Yang 2021Protein & Cell2021,12,11:4
7Formal Analysis of Trusted Platform Module Commands for Compromising User Key显示文摘The Trusted Platform Module (TPM) is a dedicated hardware chip designed to provide a higher level of security for computing platform. All TPM functionalities are implemented in TPM commands to achieve specific security goals. We attempt to analyze the security properties of these commands, especially the key management API. Our study utilizes applied pi calculus to formalize the commands and determine how their security properties affect TPM key management. The attacker is assumed to call TPM commands without bounds and without knowing the TPM root key, expecting to obtain or replace the user key. The analysis goal in our study is to guarantee the corresponding property of API execution and the integrity of API data. We analyze the security properties of TPM commands with a process reduction method, identify the key-handle hijack attack on a TPM newly created key, and propose reasonable solutions to solve the problem. Then, we conduct an experiment involving a key-handle attack, which successfully replaces a user key with an attacker's key using malicious TPM software. This paper discloses the weakness of the relationship between the key handle and the key object. 8Defect detection in atomic-resolution images via unsupervised learning with translational invariance显示文摘Crystallographic defects can now be routinely imaged at atomic resolution with aberration-corrected scanning transmission electron microscopy(STEM)at high speed,with the potential for vast volumes of data to be acquired in relatively short times or through autonomous experiments that can continue over very long periods.Automatic detection and classification of defects in the STEM images are needed in order to handle the data in an efficient way.However,like many other tasks related to object detection and identification in artificial intelligence,it is challenging to detect and identify defects from STEM images.Furthermore,it is difficult to deal with crystal structures that have many atoms and low symmetries.Previous methods used for defect detection and classification were based on supervised learning,which requires human-labeled data.In this work,we develop an approach for defect detection with unsupervised machine learning based on a one-class support vector machine(OCSVM).We introduce two schemes of image segmentation and data preprocessing,both of which involve taking the Patterson function of each segment as inputs.We demonstrate that this method can be applied to various defects,such as point and line defects in 2D materials and twin boundaries in 3D nanocrystals.Yueming Guo Sergei V.Kalinin Hui Cai Kai Xiao Sergiy Krylyuk Albert V.Davydov Qianying Guo Andrew R.Lupini 2021npj Computational Materials2021,,1:2
9Comparative study on the weight loss and lipid metabolism by tea polyphenols in diet induced obese C57BL/6J pseudo germ free and conventionalized mice显示文摘The role of gut microbiota in terms of host health is becoming increasingly important.In this study,the comparative effects of tea polyphenols(TPs)on weight loss and lipid metabolism on conventionalized mice(CVZ)and pseudo germ-free(PGF)mice(treated with antibiotics)were investigated.Our findings revealed that high fat(HF)diet considerably increased the body weight,total fat and upsurge lipid indices in CVZ mice but PGF mice were not sensitive to the effect of HF diet as CVZ mice.After the dietary administration of TP,body weight,perirenal fat and epididymal fat,liver weight,glucose(GLU)level,total chloestrol(TC level),high density lipoprotein-cholesterol(HDL-C)level significantly lowered in PGF mice as compared to CVZ mice group.However,the area of fat cells and triglyceride(TG)level were significantly increased in PGF mice.In CVZ mice,TP intervention resulted in a considerable drop in the Firmicutes/Bacteroides ratio as compared to PGF mice.The intestinal flora of PGF mice was severely reduced after antibiotic treatment,while TP administration restored intestinal diversity;the abundance of Akkermansia and Lactobacillus increased,whereas the abundance of Enterobacteriaceae and Prevotella reduced.Overall,we can assume that PGF obese mice administered with TP have less anti-obesity effects compared to obese CVZ mice.Kashif Hussain Yingying Yang Jie Wang Hengjuan Bian Xi Lei Junjie Chen Qianying Li Li Wang Qingping Zhong Xiang Fang Yutao Wang Hong Wei Yigang Tong Zhenlin Liao 2022Food Science and Human Wellness2022,11,3:2

16A new type-γ'/γ''coprecipitation behavior and its evolution mechanism in wrought Ni-based ATI 718Plus superalloy显示文摘In the precipitation-hardened Ni-based superalloy,typified by ATI 718 Plus,the nano-scaleγandγphase in duplet or triple coprecipitate morphology can provide superior high-temperature strength.Thus,it is of great sense to study the evolution ofγ’/γ’’coprecipitate during long term service at elevated temperature.In this study,the new-typeγ’/γ’’coprecipitates with a sandwich or compact configuration were found firstly in wrought ATI 718 Plus superalloy during long term thermal exposure at 705℃.These co-structure of theγ’/γ’’precipitates evidently inhibit the coarsening ofγ’phase.The increase of thermal exposure time evidently leads to the increase of the volume fraction ofγ’/γ’’coprecipitate and transformation of sandwich-typeγ’/γ’’coprecipitate to compact-typeγ’/γ’’coprecipitate,which is characterized asγphase precipitate at several faces of theγphase.The main evolution mechanism ofγ’/γ’’coprecipitates is element segregation,especially the composition variations of Al+Ti and Nb and their ratio of Al+Ti/Nb.In addition,the interfacial energy betweenγ’’phase andγmatrix also plays a key role on theγ’/γ’’coprecipitates evolution.The calculated results show that the longer thermal exposure time leads to the higher interfacial energy,which is beneficial for nucleation and precipitation ofγ’’phase on the faces ofγ’phase.Qianying Guo Zongqing Ma Zhixia Qiao Chong Li Teng Zhang Jun Li Chenxi Liu Yongchang Liu 2022Journal of Materials Science & Technology2022,,24:1
