1ABNORMAL FLOWER AND GRAIN 1 encodes OsMADS6 and determines palea identity and affects rice grain yield and quality显示文摘The palea and lemma are floral organ structures unique to grasses;these structures form the hull and directly affect grain size and quality. However, the molecular mechanisms controlling the development of the hull are not well understood. In this study, we characterized the rice(Oryza sativa) abnormal flower and grain1(afg1) mutant, a new allele of OsMADS6. Similar to previously characterized osmads6 alleles, in the afg1 floret, the palea lost its marginal region and acquired the lemma identity. However, in contrast to other osmads6 alleles, the afg1 mutant showed altered grain size and grain quality, with decreased total starch and amylose contents, and increased protein and soluble sugar contents. The analysis of transcriptional activity suggested that AFG1 is a transcriptional activator and may affect grain size by regulating the expression levels of several genes related to cell expansion and proliferation in the afg1 mutant. 2Mussel-inspired agarose hydrogel scaffolds for skin tissue engineering显示文摘Polysaccharide hydrogels are widely used in tissue engineering because of their superior biocompatibility and low immunogenicity.However,many of these hydrogels are unrealistic for practical applications as the cost of raw materials is high,and the fabrication steps are tedious.This study focuses on the facile fabrication and optimization of agarose-polydopamine hydrogel(APG)scaffolds for skin wound healing.The first study objective was to evaluate the effects of polydopamine(PDA)on the mechanical properties,water holding capacity and cell adhesiveness of APG.We observed that APG showed decreased rigidity and increased water content with the addition of PDA.Most importantly,decreased rigidity translated into significant increase in cell adhesiveness.Next,the slow biodegradability and high biocompatibility of APG with the highest PDA content(APG3)was confirmed.In addition,APG3 promoted full-thickness skin defect healing by accelerating collagen deposition and promoting angiogenesis.Altogether,we have developed a straightforward and efficient strategy to construct functional APG scaffold for skin tissue engineering,which has translation potentials in clinical practice.Ting Su Mengying Zhang Qiankun Zeng Wenhao Pan Yijing Huang Yuna Qian Wei Dong Xiaoliang Qi Jianliang Shen 2021Bioactive Materials2021,6,3:6
3The pleiotropic ABNORMAL FLOWER AND DWARF1 affects plant height, floral development and grain yield in rice显示文摘Moderate plant height and successful establishment of reproductive organs play pivotal roles in rice grain production. The molecular mechanism that controls the two aspects remains unclear in rice. In the present study,we characterized a rice gene, ABNORMAL FLOWER AND DWARF1(AFD1) that determined plant height, floral development and grain yield. The afd1 mutant showed variable defects including the dwarfism, long panicle, low seed setting and reduced grain yield. In addition, abnormal floral organs were also observed in the afd1 mutant including slender and thick hulls, and hull-like lodicules.AFD1 encoded a DUF640 domain protein and was expressed in all tested tissues and organs. Subcellular localization showed AFD1-green fluorescent fusion protein(GFP) was localized in the nucleus. 5Nitrogen doped tin oxide nanostructured catalysts for selective electrochemical reduction of carbon dioxide to formate显示文摘Tin/tin oxide materials are key electrocatalysts for selective conversion of CO_2 to formate/formic acid.Herein we report a tin oxide material with nitrogen doping by using ammonia treatment at elevated temperature. The N doped material demonstrated enhanced electrocatalytic CO_2 reduction activity, showing high Faradaic efficiency(90%) for formate at -0.65 V vs. RHE with partial current density of 4 mA/cm^2.The catalysis was contributed to increased electron negativity of N atom compared to O atom. Additionally, the N-doped catalyst demonstrates sulfur tolerance with retained formate selectivity. 6A fiber Bragg grating based earth and water pressures transducer with three-dimensional fused deposition modeling for soil mass显示文摘A novel fiber Bragg grating(FBG)sensor with three-dimensional(3D)fused deposition modeling(FDM)approach is proposed for effective stress measurement in soil mass.The three-diaphragm structure design is developed to measure earth and water pressures simultaneously.The proposed transducer has advantages of small size,high sensitivity,low cost,immunity to electromagnetic interference and rapid prototyping.The working principle,design parameters,and manufacturing details are discussed.The proposed transducer was calibrated for earth and water pressures measurement by using weights and a specially designed pressure chamber,respectively.The calibration results showed that the wavelength of the transducer was proportional to the applied pressure.The sensitivity coefficients of the earth and water pressures were 12.633 nm/MPa and 6.282 nm/MPa,respectively.Repeated tests and error analysis demonstrated the excellent stability and accuracy of the earth and water pressure measurements.The performance of the proposed transducer was further verified by a model experimental test and numerical analysis,which indicated that the proposed transducer has great potential for practical applications.Yue Qin Qiankun Wang Dongsheng Xu Jiaming Yan Shanshan Zhang 2022Journal of Rock Mechanics and Geotechnical Engineering2022,14,2:3
7Fecal microbiota transplantation from young donor mice improves ovarian function in aged mice显示文摘Advanced maternal age is characterized by declines in the quantity and quality of oocytes in the ovaries,and the aging process is accompanied by changes in gut microbiota composition.However,little is known about the relationship between gut microbiota and ovarian aging.By using fecal microbiota transplantation(FMT)to transplant material from young(5-week-old)into aged(42-week-old)mice,we find that the composition of gut microbiota in FMT-treated mice presents a“younger-like phenotype”and an increase of commensal bacteria,such as Bifidobacterium and Ruminococcaceae.Moreover,the FMT-treated mice show increased anti-inflammatory cytokine IL-4 and decreased pro-inflammatory cytokine IFN-γ.Fertility tests for assessing ovarian function reveal that the first litter size of female FMT-treated mice is significantly higher than that of the non-FMT group.Morphology analysis demonstrates a dramatic decrease in follicle atresia and apoptosis as well as an increase in cellular proliferation in the ovaries of the FMT-treated mice.Our results also show that FMT improves the immune microenvironment in aged ovaries,with decreased macrophages and macrophage-derived multinucleated giant cells(MNGCs).These results suggest that FMT from young donors could be a good choice for delaying ovarian aging.Li Xu Qiankun Zhang Xiaowei Dou Yipeng Wang Jianwei Wang Yong Zhou Xingyin Liu Jing Li 2022Journal of Genetics and Genomics2022,49,11:2

10Ammonia-induced oxidative stress triggered proinflammatory response and apoptosis in pig lungs显示文摘Ammonia,a common toxic gas,is not only one of the main causes of haze,but also can enter respiratory tract and directly affect the health of humans and animals.Pig was used as an animal model for exploring the molecular mechanism and dose effect of ammonia toxicity to lung.In this study,the apoptosis of type II alveolar epithelial cells was observed in high ammonia exposure group using transmission electron microscopy.Gene and protein expression analysis using transcriptome sequencing and western blot showed that low ammonia exposure induced T-cell-involved proinflammatory response,but high ammonia exposure repressed the expression of DNA repair-related genes and affected ion transport.Moreover,high ammonia exposure significantly increased 8-hydroxy-2-deoxyguanosine (8-OHdG) level,meaning DNA oxidative damage occurred.In addition,both low and high ammonia exposure caused oxidative stress in pig lungs.Integrated analysis of transcriptome and metabolome revealed that the up-regulation of LDHB and ND2 took part in high ammonia exposure-affected pyruvate metabolism and oxidative phosphorylation progress,respectively.Inclusion,oxidative stress mediated ammonia-induced proinflammatory response and apoptosis of porcine lungs.These findings may provide new insights for understanding the ammonia toxicity to workers in livestock farms and chemical fertilizer plants.Daojie Li Long Shen Di Zhang Xiaotong Wang Qiankun Wang Wenhao Qin Yun Gao Xiaoping Li 2023Journal of Environmental Sciences2023,,4:2
11MiR146a-loaded engineered exosomes released from silk fibroin patch promote diabetic wound healing by targeting IRAK1显示文摘Unhealable diabetic wounds need to be addressed with the help of newer,more efficacious strategies.Exosomes combined with biomaterials for sustained delivery of therapeutic agents are expected to bring new hope for chronic wound treatment.Here,the engineered exosomes modified for efficiently loading miR146a and attaching to silk fibroin patch(SFP)were demonstrated to promote diabetic wound healing.Silk fibroin binding peptide(SFBP)was screened through phage display,and SFBP-Gluc-MS2(SGM)and pac-miR146a-pac fusion protein were constructed.The designed exosomes(SGM-Exos,miR146a-Exos,and SGM-miR146a-Exos)were isolated from the engineered placental mesenchymal stem cells(PMSCs)transduced with SGM or/and pac-miR146a-pac protein.Gluc signals indicated SGM-Exo@SFP markedly increased the binding rate and the stability of SGM-Exo.Moreover,the loading efficiency of miR146a in SGM-miR146a-Exos was ten-fold higher than that in miR146a-Exos.Superior to untreated,SGM-miR146a-Exo-only treated,and SFP-only treated groups,SGM-miR146a-Exo@SFP drived wound healing associated with less inflammation,collagen deposition,and neovascularization.The transcriptomics analysis suggested anti-inflammatory and regenerative effects with SGM-miR146a-Exo@SFP treatment.Here,we show efficient exosome@biomaterial-based miRNA delivery systems for regenerative medicine and tissue engineering.Qiankun Li Wenzhi Hu Qilin Huang Jie Yang Bingmin Li Kui Ma Qian Wei Yaxi Wang Jianlong Su Mengli Sun Shengnan Cui Rungong Yang Haihong Li Xiaobing Fu Cuiping Zhang 2023Signal Transduction and Targeted Therapy2023,8,3:2

18Cordierite Ceramics Prepared from Poor Quality Kaolin for Electric Heater Supports: Sintering Process, Phase Transformation, Microstructure Evolution and Properties显示文摘To efficiently utilize the kaolin, an economical way of preparing cordierite ceramic with high performance for electric heater supports was put forward. In this study, sintering process, phase transformation, microstructure evolutions were systematically studied by heating microscope, X-ray diffraction, scanning electronic microscope and thermal analysis. Properties(physical properties, electrical properties and coefficient of thermal expansion(CTE)) were tested for comprehensive performance evaluation. The results showed that the utilization of poor quality kaolin broadened the firing range of cordierite ceramic which was from 1 200 to 1 380 ℃. Microstructure becomes loose with increasing of the pore size, which had significant influence on bending strength and electrical properties. High content of K2 O in poor quality kaolin was the reason for liquid phase generation in sintering process, which further leads to microstructural changes. The cordierite ceramic sintered at 1 320 ℃ had the properties as follows: CTE of 1.98×10^(-6) ℃^(-1)(500 ℃), bending strength of 90 MPa, apparent porosity of 15.1%, dielectric constant of 7.5(100 Hz), and volume resistivity of 1.05×109 Ω·cm(100 Hz). 20Scutellarin prevents acute alcohol-induced liver injury via inhibiting oxidative stress by regulating the Nrf2/HO-1 pathway and inhibiting inflammation by regulating the AKT,p38 MAPK/NF-κB pathways显示文摘Alcoholic liver disease(ALD)is the most frequent liver disease worldwide,resulting in severe harm to personal health and posing a serious burden to public health.Based on the reported antioxidant and anti-inflammatory capacities of scutellarin(SCU),this study investigated its protective role in male BALB/c mice with acute alcoholic liver injury after oral administration(10,25,and 50 mg/kg).The results indicated that SCU could lessen serum alanine aminotransferase(ALT)and aspartate aminotransferase(AST)levels and improve the histopathological changes in acute alcoholic liver;it reduced alcohol-induced malondialdehyde(MDA)content and increased glutathione peroxidase(GSH-Px),catalase(CAT),and superoxide dismutase(SOD)activity.Furthermore,SCU decreased tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),and IL-1βmessenger RNA(mRNA)expression levels,weakened inducible nitric oxide synthase(iNOS)activity,and inhibited nucleotide-binding oligomerization domain(NOD)-like receptor protein 3(NLRP3)inflammasome activation.Mechanistically,SCU suppressed cytochrome P450 family 2 subfamily E member 1(CYP2E1)upregulation triggered by alcohol,increased the expression of oxidative stress-related nuclear factor erythroid 2-related factor 2(Nrf2)and heme oxygenase-1(HO-1)pathways,and suppressed the inflammation-related degradation of inhibitor of nuclear factor-κB(NF-κB)-α(IκBα)as well as activation of NF-κB by mediating the protein kinase B(AKT)and p38 mitogen-activated protein kinase(MAPK)pathways.These findings demonstrate that SCU protects against acute alcoholic liver injury via inhibiting oxidative stress by regulating the Nrf2/HO-1 pathway and suppressing inflammation by regulating the AKT,p38 MAPK/NF-κB pathways.Xiao ZHANG Zhicheng DONG Hui FAN Qiankun YANG Guili YU Enzhuang PAN Nana HE Xueqing LI Panpan ZHAO Mian FU Jingquan DONG 2023Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2023,24,7:0
