1The chemosensory protein of Chinese honeybee, Apis cerana cerana: Molecular cloning of cDNA, immunocytochemical localization and expression显示文摘Chemosensory proteins (CSPs) are ubiquitous soluble small proteins isolated from sensory organs thought to be involved in chemical communication. Here we report the first cDNA of CSPs, called Ac-ASP3, cloned and characterized from antennas of adult worker bees in Chinese honeybee, Apis cerana cerana. The Ac-ASP3 cDNA comprises 2 exons, with an ORF of 393-bp encoding 130 aa. Protein signature analyses show that the protein consists of four conserved cysteines and a signal peptide with 19 aa in the N-terminal sequence. The deduced protein sequence shares high homology with Am-ASP3 of Apis mellifera and low similarity with other species of insects. Immunocytochemical localization shows that Ac-ASP3 is only specifically expressed on the antenna contact chemosensilla such as sensilla trichodea B and sensilla basiconica, whereas Ac-ASP3 is scarcely expressed on olfactory chemosensilla such as sensilla placodea. Real-time PCR of Ac-ASP3 transcripts shows that Ac-ASP3 is highly expressed on wings and legs, but expression is lower on antenna. Temporal expression patterns suggest that Ac-ASP3 is expressed during the period of pupa and adults from 1-d to 6-d stages when bees act as house bees, cleaning the comb and taking care of the queen and larvae in comb. The above evidence suggests that Ac-ASP3 is unique in species and is generally not involved in olfaction during searching for honey and pollen. 2Piriformospora indica confers cadmium tolerance in Nicotiana tabacum显示文摘Piriformospora indica, a root-colonizing endophytic fungus of Sebacinales, promotes plant growth and confers resistance against biotic and abiotic stresses. In order to confirm the influence of P. indica on growth, proline, malondialdehyde(MDA), chlorophyll, and cadmium(Cd) amounts in Nicotiana tabacum under Cd stress, hydroponics, pot and field trials were conducted. The results showed that P. indica can store Cd in plant roots and reduce leaf Cd content, reduce the concentration of MDA, and increase the proline and chlorophyll content and the activities of catalase, peroxidase, and superoxide dismutase under hydroponic Cd stress. RT-PCR analysis showed that the relative expression level of genes Gsh2, Ta PCS1, oas1, GPX, and Hsp70 in colonized plants was 4.3, 1.4, 2.9, 1.7, and 6.9fold higher than in un-colonized plants respectively. Cd exposure significantly reduced un-colonized plants' agronomic traits compared to P. indica-colonized ones. Our results suggested that P. indica can sequester Cd in roots, so that much less cadmium was transported to leaves, and the increased concentrations of antioxidant enzymes, pigments and proline contents, as well as the higher expression of stress-related phytochelatin biosynthesis genes in P. indica-inoculated plants, may also serve to protect N. tabacum plants against oxidative damage, enhancing Cd tolerance.Feiqiong Hui Jian Liu Qikang Gao Binggan Lou 2015Journal of Environmental Sciences2015,27,11:6
3Innovation on Distant Hybridization of Saline-tolerant Mud Flat Spartina and Rice Germplasm显示文摘To explore saline-tolerant high yield Spartina and rice germplasm resources,innovate upon distant hybridization technology,and develop new saline-tolerant mud flat Spartina germplasm,autonomous patent for invention was used to study distant hybridization of Spartina and rice in 2005- 2016. The study adopted tidal flat planting screening,phenotypic screening,molecular marker assisted selection,to find biological evidences of new germplasm,and also applied crossbreeding,conventional breeding,and molecular breeding,to select new saline-tolerant Spartina and rice varieties. With 12 years of efforts,the team developed 1498 crossbreeding combinations and established pedigree file and seed bank,tested 307 crossbreeding materials(57 hybrid seeds showed parental genetic components of Spartina and Rice); obtained Spartina/ Rice distant hybridization and seed setting,transplanting,hybridization material economic traits,and molecular biological experimental evidences; Rice♀× Spartina ♂ hybridization success rate of 1. 04- 1. 39%; obtained authorization of patent for invention of Distant Hybridization Methods for Spartina and Rice; formulated procedures for high yield cultivation of Spartina-Rice,planted 33. 33 hm2Spartina-Rice in coastal saline and alkaline land,cultivated 10 new saline-tolerant Spartina-Rice varieties,and the rice yield up to 5. 925- 8. 28 t / hm2. Results indicate that Spartina-Rice is saline-tolerant high yield rice germplasm,developed through sexual hybridization,conventional breeding,and molecular breeding technologies,and is optimal crop for saline and alkaline land. Success of Spartina-Rice solves the problem of saline-tolerant crop badly for transformation of saline and alkaline land,the problem of feeds for herbivores,as well as problem of environmental pollution resulted from straw burning. 12Tropical Cyclone Tilts Under Vertically Varying Background Flows:Preliminary Results Based upon TCM4 Simulations显示文摘The characteristics of tropical cyclone(TC) tilts under vertically varying background flows were preliminarily examined in this study based on numerical simulations with the Tropical Cyclone Model version 4(TCM4).The tilt magnitudes presented a linearly decreasing tendency in the simulation with the environmental wind speed vertically varying throughout the troposphere and in the simulation with the vertical wind shear concentrated in the lower troposphere,while the vortex tilt showed a linearly increasing tendency in magnitude in the simulation where the vertical shear was concentrated in the upper troposphere.The change in tilt magnitude was found to be related to the evolution of the penetration depth near the eyewall.When the shear was concentrated in the lower troposphere,the vortex tended to tilt downshear right during the early integration and underwent more precession processes.When the shear was concentrated in the upper troposphere,the vortex rapidly tilted downshear left during the early simulation and vortex precession was less frequently observed.The storms simulated in all experiments were finally in downshear-left tilt equilibrium.ZHENG Qikang LI Qingqing 2018Journal of Ocean University of China2018,17,5:1
13Insight into sulfur and iron effect of binary nickel-iron sulfide on oxygen evolution reaction显示文摘Nickel-iron sulfide has shown attractive activity in electrocatalytic oxygen evolution reaction(OER).However,the effects of low valence sulfur(S^(2−))and metal species on OER in binary nickel-iron sulfide have rarely been systematically studied.Works based on post-catalysis characterization have led to the assumption that the real active species are nickel-iron oxyhydroxide,and that nickel-iron sulfide acts only as a precatalyst.Therefore,to study the role of S,Ni,and Fe for the development of nickel-iron sulfide catalyst is of self-evident importance.Herein,a facile solvothermal method is used to synthesize acetylene black coated with nickel-iron sulfide nanosheets.Electrochemical tests show that the presence of low valence S species makes the catalyst have faster OER kinetics,larger active area,and intermediate active species adsorption area.Therefore,the present study reveals the enhancing effect of low valence sulfur species(S^(2−))on OER in binary nickel-iron sulfide.In situ Raman spectroscopy shows that the generation ofγ-NiOOH intermediate is essential and Fe does not directly participate in the oxygen production.Density functional theory(DFT)calculation shows that Ni-OH deprotonation is a rate-determining step for both binary nickel-iron sulfide and nickel sulfide.The addition of Fe into NiSx lightly increases the charge transfer of Ni atom to O atom,which makes deprotonation easier and thereby improves the OER performance.Qikang Wu Songrui Wang Jiahui Guo Xueqing Feng Han Li Shanshan Lv Yan Zhou Zheng Chen 2022Nano Research2022,15,3:1
14An Unexpected Frequency Response of A Piezoelectric Quartz Crystal Sensor to the Density and Viscosity of the Liquid显示文摘An unexpected frequency response for a piezoelectric quartz crystal (PQC) sensor to liquid density and viscosity was reported. For a PQC oscillating in a liquid phase, the frequency shifts (?f ) show a wave-shape response to liquid density (ρ) and viscosity (η) in fine structure, if the longitudinal wave effect was not eliminated. This result is different from the well-known linear relationship between of ?f and (ρη)1/2. 15Studying Aggregate in Lysozyme Solution by Atomic Force Microscope显示文摘The aggregates in lysozyme solution with different NaCl concentration were investigated by Atomic Force Microscope (AFM). The AFM images show that there exist lysozyme monomers, n-mers and clusters in lysozyme solution when the conditions are not suitable for crystal growth. 16Intelligent production optimization method for a low pressure and low productivity shale gas well显示文摘Shale gas wells frequently suffer from liquid loading and insufficient formation pressure in the late stage of production.To address this issue,an intelligent production optimization method for low pressure and low productivity shale gas well is proposed.Based on the artificial intelligence algorithms,this method realizes automatic production and monitoring of gas well.The method can forecast the production performance of a single well by using the long short-term memory neural network and then guide gas well production accordingly,to fulfill liquid loading warning and automatic intermittent production.Combined with adjustable nozzle,the method can keep production and pressure of gas wells stable automatically,extend normal production time of shale gas wells,enhance automatic level of well sites,and reach the goal of refined production management by making production regime for each well.Field tests show that wells with production regime optimized by this method increased 15%in estimated ultimate reserve(EUR).Compared with the development mode of drainage after depletion recovery,this method is more economical and can increase and stabilize production effectively,so it has a bright application prospect.ZHU Qikang LIN Botao YANG Guang WANG Lijia CHEN Man 2022Petroleum Exploration and Development2022,49,4:0
