1Synergistic effects of autophagy/mitophagy inhibitors and magnolol promote apoptosis and antitumor efficacy显示文摘Mitochondria as a signaling platform play crucial roles in deciding cell fate.Many classic anticancer agents are known to trigger cell death through induction of mitochondrial damage.Mitophagy,one selective autophagy,is the key mitochondrial quality control that effectively removes damaged mitochondria.However,the precise roles of mitophagy in tumorigenesis and anticancer agent treatment remain largely unclear.Here,we examined the functional implication of mitophagy in the anticancer properties of magnolol,a natural product isolated from herbal Magnolia officinalis.First,we found that magnolol induces mitochondrial depolarization,causes excessive mitochondrial fragmentation,and increases mitochondrial reactive oxygen species(mtROS).Second,magnolol induces PTEN-induced putative kinase protein 1(PINK1)-Parkin-mediated mitophagy through regulating two positive feedforward amplification loops.Third,magnolol triggers cancer cell death and inhibits neuroblastoma tumor growth via the intrinsic apoptosis pathway.Moreover,magnolol prolongs the survival time of tumor-bearing mice.Finally,inhibition of mitophagy by PINK1/Parkin knockdown or using inhibitors targeting different autophagy/mitophagy stages significantly promotes magnolol-induced cell death and enhances magnolol's anticancer efficacy,both in vitro and in vivo.Altogether,our study demonstrates that magnolol can induce autophagy/mitophagy and apoptosis,whereas blockage of autophagy/mitophagy remarkably enhances the anticancer efficacy of magnolol,suggesting that targeting mitophagy may be a promising strategy to overcome chemoresistance and improve anticancer therapy.Yancheng Tang Liming Wang Tao Yi Jun Xu Jigang Wang Jiang-Jiang Qin Qilei Chen Ka-Man Yip Yihang Pan Peng Hong Yingying Lu Han-Ming Shen Hu-Biao Chen 2021Acta Pharmaceutica Sinica B2021,11,12:2
2A Fast Tongue Detection and Location Algorithm in Natural Environment显示文摘The collection and extraction of tongue images has always been an important part of intelligent tongue diagnosis.At present,the collection of tongue images generally needs to be completed in a sealed,stable light environment,which is not conducive to the promotion of extensive tongue image and intelligent tongue diagnosis.In response to the problem,a newalgorithm named GCYTD(GELU-CA-YOLO Tongue Detection)is proposed to quickly detect and locate the tongue in a natural environment,which can greatly reduce the restriction of the tongue image collection environment.The algorithm is based on the YOLO(You Only Look Once)V4-tiny network model to detect the tongue.Firstly,the GELU(Gaussian Error Liner Units)activation function is integrated into the model to improve the training speed and reduce the number of model parameters;then,the CA(Coordinate Attention)mechanism is integrated into the model to enhance the detection precision and improve the failure tolerance of the model.Compared with the other classical algorithms,Experimental results show thatGCYTD algorithm has a better performance on the tongue images of all types in terms of training speed,tongue detection speed and detection precision,etc.The lighter model can contribute on deploying the tongue detection model on small mobile terminals.Lei Zhu Guojiang Xin Xin Wang Changsong Ding Hao Liang Qilei Chen 2022Computers, Materials & Continua2022,,12:1
3Reliability analysis of retractable drill bit with air reverse circulation used for drilling while casing显示文摘Casing-while-drilling(CWD)with down the hole(DTH)hammer drilling technology has been widely used in unconsolidated formations,due to its advantages in protecting the borehole wall,excellent rock cuttings capacity,and fast penetration rate in hard rock.As an important component of the CWD system,the structure of the retractable drill bit needs not only to ensure to form stronger reverse circulation,but also to be expandable or retractable as needed,otherwise the drill bit cannot be lift and put down smoothly in the casing,and may lead to drilling accidents.This paper developed a new type of reverse circulation DTH hammer drill bit used for CWD drilling technology.The retractable performance of this type of drill bit is studied using ADAMS software.The results show that it is smoothly expandable and retractable as designed under the conditions of the weight of the bit(WOB)of 0.5–2.0 t and the rotation speed of 30–60 r/min.To investigate the reverse circulation effect of the drill bit,Fluent software was used to simulate the flow characteristic inside it.The simulation results indicated that it can form strong reverse circulation,and the entrainment ratio h can reach 9.5%.WANG Minqi YIN Qilei CHEN Baoyi QI Bo BO Kun CAO Pinlu 2023Global Geology2023,26,1:0
4An ARF24-ZmArf2 module influences kernel size in different maize haplotypes显示文摘Members of the ADP-ribosylation factor family,which are GTP-binding proteins, are involved in metabolite transport, cell division, and expansion.Although there has been a significant amount of research on small GTP-binding proteins, their roles and functions in regulating maize kernel size remain elusive. Here, we identified Zm Arf2 as a maize ADPribosylation factor-like family member that is highly conserved during evolution. Maize zmarf2 mutants showed a characteristic smaller kernel size. Conversely, ZmArf2 overexpression increased maize kernel size. Furthermore, heterologous expression of Zm Arf2 dramatically elevated Arabidopsis and yeast growth by promoting cell division. Using expression quantitative trait loci(e QTL) analysis, we determined that Zm Arf2 expression levels in various lines were mainly associated with variation at the gene locus. The promoters of Zm Arf2 genes could be divided into two types, p S and p L, that were significantly associated with both Zm Arf2 expression levels and kernel size. In yeast-one-hybrid screening, maize Auxin Response Factor 24(ARF24) is directly bound to the Zm Arf2 promoter region and negatively regulated Zm Arf2 expression.Notably, the p S and p L promoter types each contained an ARF24 binding element: an auxin response element(AuxRE) in p S and an auxin response region(Aux RR) in p L, respectively. ARF24binding affinity to Aux RR was much higher compared with Aux RE.
