| 1 | Impact of copy number of distinct SV40PolyA segments on expression of a GFP reporter gene显示文摘The presence of Alu repeats downregulates the expression of the green fluorescent protein(GFP) gene.We found that SV40PolyA(PolyA,240 bp),in either orientation,eliminated the inhibition of GFP gene expression induced by Alu repeats when it was placed between the GFP gene and the Alu repeats.In this study,4 different segments(each 60 bp) were amplified from antisense PolyA(PolyAas) by PCR,and inserted upstream of Alu14 in pAlu14 plasmid(14 Alu repeats inserted downstream of the GFP gene in vector pEGFP-C1 in a head-tail tandem manner).Segments 1F1R(the first 60 bp segment at the 5' end of PolyAas) and 4F4R(the fourth 60 bp segment from the 5' end of PolyAas) did not activate GFP gene expression,whereas 2F2R and 3F3R(the middle two segments) did(as detected by Northern blot analysis and fluorescent microscopy).Different copy numbers of 2F2R and 3F3R segments,in a head and tail tandem manner,were inserted downstream of the GFP gene in pAlu14.p2F2R*4-Alu28,p3F3R*4-Alu18 and p3F3R*4-Alu28 were used as length controls to verify that the decrease in the expression of GFP was not due to the increased length of the inserted segment in the expression vectors.We found that 2 and 4 copies of 2F2R or 3F3R activated the GFP gene more strongly than one copy of them.However,more than 8 copies of 2F2R or 3F3R reduced the activation of the GFP gene.We concluded that SV40PolyAas contained at least two gene-activating elements(2F2R and 3F3R) and 2-4 copies of 2F2R or 3F3R were optimal for the expression of the GFP gene. | YIN Kang,WANG XiuFang,MA Huan,XIE Ying,FENG JingJing,YANG QinQing & Lü ZhanJun Hebei Key Lab of Laboratory Animal,Department of Genetics,Hebei Medical University,Shijiazhuang 050017,China | 2010 | Science China(Life Sciences)2010,53,5: | 2 |

| 8 | Superconductivity and strong spin-orbit coupling in a new noncentrosymmetric compound ThIrP显示文摘A new noncentrosymmetric phosphide ThIrP has been synthesized and characterized. X-ray difraction analysis shows that this compound crystallizes in a LaPtSi-type tetragonal lattice(space group I41md, Z = 4), whose lattice parameters are a = b =4.0676(1) ?, c = 14.3354(2) ?, and V = 237.191(8) ?3. Moreover, ThIrP is discovered to be an intermediately coupled,type-Ⅱ superconductor with possibly multiple gaps below Tc= 5.07 K. The upper critical magnetic field, Sommerfield coefficient, and Ginzburg-Laudau parameter are determined based on physical property measurements, which are Bc2= 0.83 T, γ =7.5 mJ mol-1 K-2, and κGL= 7.5, respectively. The electronic band structure calculations point out nearly equal contributions of Ir and Th atoms on the density of states around the Fermi surface. In addition, the spin-orbit coupling induced band splitting reaches as large as 270 meV along the Γ-Z line. | 19 | Study on Intermittent Discharge Characteristics of Typical Solid Insulation Defects in Gas Insulated Switchgear显示文摘In response to the problem of frequent leakage and false alarm of partial discharge insulation defects in GIS,this paper conducts experimental research on intermittent discharge characteristics of common solid insulation defects in GIS.Using true GIS to build amulti-source testing platform for intermittent discharge of solid insulation defects,and using pulse current method,ultra-high frequency method,ultrasonic method,and gas characteristic component detectionmethod to study the variation law of intermittent discharge characteristics of solid insulation defects.The results showthat:the intermittent discharge state of metal fouling defects on the solid insulation surface decreases with the extension of the discharge time,the discharge time interval as a whole shows an increasing trend,and the metal fouling defects on the solid insulation surface in the intermittent discharge state is difficult to develop into a breakdown discharge fault.The overall discharge amount of air gap defects inside solid insulation also shows the law of increasing first and then decreasing,and the discharge amount of air gap defects inside solid insulation stays at a large level for a long time during intermittent discharge,and there is a risk of breakdown,and the discharge will gradually evolve into continuous discharge;ultrasonic method and characteristic component detection method cannot realize the effective perception of intermittent discharge of metal fouling.The 50-period mapping information commonly used in the power field cannot reflect the intermittent discharge variation law of solid insulation,and the 200-period UHF mapping composed of four groups of 50-period UHF mapping data can initially derive the intermittent discharge distribution law of solid insulation defects.The research results of this paper are of great significance to expand the knowledge of intermittent discharge characteristics and the detection of GIS discharge defects in the field. | Xu Yang Jing Zhang Chuanxian Luo Qinqing Huang HuiXu Guozhi Zhang | 2023 | Energy Engineering2023,120,9: | 0 |
